摘要:快速可靠和具有成本效益的细菌区分是人类健康保健,食品安全,法医和环境科学等许多领域的重要问题。本实验通过种子生长法合成了Au@Ag纳米颗粒(Au@Ag NPs),通过浸涂法使其自组装在滤纸片上,制备了基于滤纸的柔性SERS基底,使用拉曼信标分子4-巯基苯甲酸(4-MBA)对基底的SERS性能进行评估。分别将五种食源性致病菌(鼠伤寒沙门氏菌、金黄色葡萄球菌、大肠埃希氏菌、福氏志贺菌、单核细胞增生李斯特氏菌)悬浮液滴加在基底上进行SERS信号测量。五种细菌SERS光谱主要峰的位置接近,但振动峰的相对强度显著不同,且不同菌种具有独特的拉曼峰。将主成分分析(PCA)方法与细菌SERS光谱结合,用于食源性致病菌的快速鉴别。结果显示,五种细菌的数据点聚集成五个独立的簇,没有重叠,菌种之间具有明显差异。表明使用该方法可以有效地对细菌进行分类和鉴定。同时对金黄色葡萄球菌和鼠伤寒沙门氏菌进行定量检测,最低检测限均为105 cfu/mL。

Rapid detection of foodborne pathogens by flexible SERS substrate based on filter paper
Abstract:I Fast, reliable and cost-effective bacterial differentiation is an important issue in many areas such as human health care, food safety, forensics and environmental science. In this experiment, Au@Ag nanoparticles (Au@Ag NPs) were synthesized by seed growth method, and were self-assembled on a filter paper sheet by dip coating to prepare a flexible SERS substrate based on filter paper. Raman beacon molecule 4-mercapto Benzoic acid (4-MBA) was used to evaluate the SERS performance of the substrate. Five foodborne pathogens (Salmonella typhimurium, Staphylococcus aureus, Escherichia coli, Shigella flexneri, Listeria monocytogenes) were added to the substrate respectively for SERS signal measurement. The positions of the main peaks of the SERS spectra of the five bacteria are similar, but the relative intensities of the vibration peaks are significantly different, and different strains have unique Raman peaks. The principal component analysis (PCA) method was combined with bacterial SERS spectra for rapid identification of foodborne pathogens. The results showed that the data points of the five bacteria were clustered into five independent clusters. At the same time, quantitative detection of Staphylococcus aureus and Salmonella typhimurium, the minimum detection limit was 105 cfu / mL.
