无机砷与二甲基一硫代胂酸诱导HaCaT细胞氧化应激的比较

摘要:【目的】比较砷甲基化代谢物二甲基一硫代胂酸(DMMTA)与无机砷诱导人皮肤角质形成细胞(HaCaT)氧化应激的差异,为砷甲基化代谢物的毒性评估以及致癌机制研究提供基础资料。【方法】将人皮肤角质形成细胞传代培养后随机分为7个组。分别用含0.5μM亚砷酸钠,1.0μM亚砷酸钠,5.0μM亚砷酸钠,0.5μM DMMTA,1.0μM DMMTA,5.0μM DMMTA的培养基以及正常培养基(对照组)处理24小时。用相应的试剂盒检测人皮肤角质形成细胞内的丙二醛(MDA), 8-羟基脱氧鸟苷(8-OHdG),过氧化氢,超氧化物以及谷胱甘肽的水平。【结果】相同剂量下,DMMTA处理组的氧化损伤相关指标(MDA,8-OHdG)的水平明显高于无机砷处理组(P<0.05),活性氧(过氧化氢,超氧化物)水平相比无机砷处理组也明显更高(P<0.05),而谷胱甘肽水平则明显低于无机砷处理组(P<0.05)。【结论】相同剂量下,相比于无机砷,DMMTA更易引起HaCaT细胞氧化损伤,这不仅与其更易刺激活性氧自由基的产生有关,也可能与其特异性的消耗了细胞内的抗氧化物质(谷胱甘肽)有关。

Comparison of oxidative stress induced by inorganic arsenic and dimethylmonothioarsinic acid in HaCaT cells
Abstract:[Object] The difference in oxidative stress induced by arsenic methylation metabolite dimethylmonothioarsinic acid (DMMTA) and inorganic arsenic was compared in human keratinocytes (HaCaT). It is to provide basic information for the toxicity assessment of arsenic methylation metabolites and the study of carcinogenic mechanisms. [Methods] Human keratinocytes were randomly divided into 7 groups. The cells were treated with normal medium (control group) and medium containing 0.5μM sodium arsenite, 1.0μM sodium arsenite, 5.0μM sodium arsenite, 0.5μM DMMTA, 1.0μM DMMTA, 5.0μM DMMTA for 24 hours, respectively. The levels of malondialdehyde (MDA), 8-hydroxydeoxyguanosine (8-OHdG), hydrogen peroxide, superoxide and glutathione in human keratinocytes were measured with the suitable kit. [Results] At the same dose, the levels of oxidative damage (MDA, 8-OHdG) in the DMMTA treatment group were significantly higher than those in the inorganic arsenic treatment group (P<0.05) and the levels of reactive oxygen species (hydrogen peroxide, superoxide) were also significantly higher than those in the inorganic arsenic treatment group (P<0.05), while the levels of glutathione were significantly lower than those in the inorganic arsenic treatment group (P<0.05). [Conclusion] At the same dose, compared with the inorganic arsenic, DMMTA is more likely to cause oxidative damage in HaCaT cells .This is not only because it is more readily to stimulate the production of reactive oxygen species, but also because it consumes intracellular antioxidants (glutathione) specially.
