Cur@Hb纳米颗粒增强乏氧肝癌细胞放射敏感性

摘要:恶性肿瘤已成为危害国民健康的重要原因,而放射治疗早已成为肿瘤治疗的重要组成部分。但是,恶性实体瘤中乏氧普遍存在,乏氧肿瘤细胞对放射治疗的抗性增加,严重影响了肿瘤放射治疗的效果。姜黄素血红蛋白纳米颗粒(Cur@Hb)可通过抑制SMMC 7721肝癌细胞细胞增殖和迁移能力,促进细胞凋亡。Cur@Hb联合4 Gy X射线照射使SMMC7721细胞发生G2/M 期阻滞。Cur@Hb纳米颗粒明显抑制体外培养的SMMC7721细胞血管管腔样结构生成,EMT相关蛋白VE-cadherin、Twist1和MMP2表达量明显降低。Cur@Hb纳米颗粒明显增强常氧或乏氧SMMC7721细胞的放射敏感性,放射增敏比分别约为1.385、1.309; Cur@Hb纳米颗粒明显增加常氧或乏氧SMMC7721细胞X射线诱导的彗星尾部DNA百分含量。这些结果表明,Cur@Hb可增强乏氧肝癌细胞放射敏感性。

Cur@Hb nanoparticles enhance radiosensitivity of hypoxic hepatocellular carcinoma cells
Abstract:Malignant tumors have become an important cause of harm to national health, and radiation therapy has long been an important part of tumor treatment. However, hypoxia is common in malignant solid tumors, and the resistance of hypoxic tumor cells to radiation therapy is increased, which seriously affects the efficacy of tumor radiation therapy. Incubation Cur@Hb in normoxia or hypoxia (1% O2) for 24 h can significantly inhibit the proliferation and migration of SMMC 7721 cells, and promote apoptosis. Cur@Hb combined with 4 Gy X-ray irradiation caused G2/M phase arrest of SMMC7721 cells. In addition, Cur@Hb nanoparticles can significantly inhibit the generation of vascular tube-like structures, and the expression of EMT-related protein VE-cadherin, twist1 and MMP2 was significantly decreases.Cur@Hb nanoparticles significantly enhanced the radiosensitivity of normoxic and hypoxic SMMC7721, with radiosensitization ratios of approximately 1.385 and 1.309, respectively; Cur@Hb nanoparticles significantly increased the x-ray-induced percent of DNA tail of normoxic or hypoxic SMMC7721 cells quantity. Cur@Hb nanoparticles can significantly enhance the effect of radiotherapy.
