利用RNA-Seq技术探究三法三穴对坐骨神经损伤大鼠的基因表达影响

摘要:目的:探究推拿对坐骨神经损伤大鼠神经损伤点基因表达的影响,从转录组水平阐明推拿促进周围神经损伤的恢复机制。方法:27只SD大鼠随机分为假手术组、模型组和三法三穴组。三法三穴组大鼠在造模7天后,每日以点法、拨法和揉法定性定量刺激大鼠术侧殷门穴、承山穴和阳陵泉穴1次,干预10次休息1天,共干预20次。RNA测序技术检测神经损伤点基因变化,并做GO分析和KEGG分析。结果:假手术组与模型组比较,共计431个基因发生显著性差异变化;模型组与三法三穴组比较,共计221个基因发生显著性差异变化;假手术组与三法三穴组比较,共计322个基因发生显著性差异变化。结论:推拿促进周围神经损伤大鼠的恢复是通过改变神经损伤点基因序列实现的。

RNA-Seq technique to explore the effect of three methods and three points on gene expression of sciatic nerve injury rats
Abstract:Objective: To explore the effect of massage on the gene expression of nerve injury point in rats with sciatic nerve injury, and to elucidate the mechanism of massage promoting the recovery of peripheral nerve injury at the transcriptome level. Methods: 27 SD rats were randomly divided into sham operated group, model group and three points group. In the three methods and three points group, after 7 days of modeling, the rats were stimulated with point method, dial method and rubbing method once a day, 10 times for rest and one day for a total of 20 times. RNA sequencing was used to detect the gene changes of nerve injury points, and go analysis and KEGG analysis were performed. Results: compared with the model group, 431 genes in the sham operation group changed significantly; compared with the three methods and three points group, 221 genes in the model group changed significantly; compared with the three methods and three points group, 322 genes in the sham operation group changed significantly. Conclusion: massage can promote the recovery of rats with peripheral nerve injury by changing the gene sequence of nerve injury point.
