阿魏酸类似物对应激小鼠的氧化还原状态和免疫功能的影响

摘要:本文研究了阿魏酸及其类似物姜黄素、谷维素对应激小鼠体内氧化还原状态和免疫功能的影响。方法:将90 只雄性ICR小鼠随机分为6 组,正常组、模型组、阿魏酸组、姜黄素组、谷维素组和阳性对照(辅酶Q10)组。每3 天,正常组肌肉注射生理盐水,其他组注射40 mg/kg体重的强的松进行造模。正常组和模型组饲喂正常饲料,其他四组饲料中分别添加了40 mg/kg的阿魏酸、姜黄素、谷维素和辅酶Q10。饲养8 周后处死取样,测定血浆、肝脏、心脏、肌肉和小肠氧化还原状态和免疫功能相关的指标。结果:(1)造模第九天开始,模型组、阿魏酸组和姜黄素组小鼠日饮水量显著高于正常组(P<0.05)。(2)阿魏酸、姜黄素、谷维素和辅酶Q10小鼠下丘脑促肾上腺素释放激素(CRH)和垂体促肾上腺皮质激素(ATCH)的含量均显著低于模型组(P<0.05);谷维素和辅酶Q10组小鼠血浆皮质酮(CROT)的含量显著低于模型组(P<0.05)。(3)谷维素组和辅酶Q10组心脏、肝脏和肌肉组织中总抗氧化能力(T-AOC)显著高于模型组(P<0.05);谷维素组和辅酶Q10组血浆、肝脏和肌肉组织中总超氧化物歧化酶(SOD)活力显著高于模型组(P<0.05);谷维素组和辅酶Q10组血浆和组织里的丙二醛(MDA)含量均显著低于模型组(P<0.05)。(4)姜黄素组、谷维素组和辅酶Q10组血浆和空肠组织中促炎因子IFN-γ、TNF-α和IL-6水平都显著低于模型组,谷维素组和辅酶Q10组血浆和空肠组织中抑炎因子IL-10显著高于模型组(P<0.05);辅酶Q10组血浆IgA的含量、谷维素组血浆IgG的含量、姜黄素组、谷维素组和辅酶Q10组IgM含量均显著高于模型组(P<0.05)。以上结果表明,在本实验条件下,阿魏酸类似物都有抗应激、抗氧化能力以及提高免疫力的作用,且作用效果:阿魏酸<姜黄素<谷维素,其中谷维素作用接近于作为阳性对照的辅酶Q10,阿魏酸效果较差。

Effects of Ferulic Acid Analogs on Redox State and Immune Function in Stress Mice
Abstract: Experiments were conducted to evaluate the effects of ferulic acid and its analogs, curcumin and oryzanol, on the redox state and immune function in stress mice. Methods: Ninety male ICR mice were randomly divided into 6 groups: normal group, model group, ferulic acid group, curcumin group, oryzanol group and positive control (coenzyme Q10) group. Every 3 days, normal group was injected with normal saline, and other groups were injected with 40 mg/kg body weight of prednisone. The normal group and the model group were fed with normal feed. The other four groups were fed with 40 mg/kg of ferulic acid, curcumin, oryzanol, and coenzyme Q10. After 8 weeks of feeding, samples were sacrificed to determine the redox status and immune function related indicators of plasma, liver, heart, muscle and intestine. Results: (1) From the ninth day of modeling, the daily water intake of mice in the model group, ferulic acid group and curcumin group was significantly higher than that in the normal group. (P <0.05) (2) The contents of hypothalamic adrenergic hormone (CRH) and pituitary adrenocorticotropic hormone (ATCH) in ferulic acid, curcumin, oryzanol and coenzyme Q10 group were significantly lower than those in the model group (P <0.05); The content of plasma corticosterone (CROT) in oryzanol and coenzyme Q10 group was significantly lower than that in the model group (P <0.05). (3) Total antioxidant capacity (T-AOC) in heart, liver and muscle tissues of oryzanol group and coenzyme Q 10 group was significantly higher than that of model group (P <0.05); Total superoxide dismutase (SOD) activity in plasma, liver and muscle tissues of oryzanol and coenzyme Q 10 group was significantly higher than those in the model group (P <0.05); the levels of malondialdehyde (MDA) in the plasma and tissues of the oryzanol group and the coenzyme Q 10 group were significantly lower than those in the model group (P <0.05). 0.05).
