基于pH响应特异富集巯基肽壳聚糖衍生物材料的理性设计、制备及性能研究

摘要:如何实现巯基寡肽的无损吸附/解吸附一直是巯基寡肽分析领域的难点。针对该问题,以壳聚糖(CTS)为原料,通过3-甲酰基苯硼酸与壳聚糖上氨基反应在壳聚糖C2位置上引入苯硼酸基团,并在壳聚糖C6位点引入羧甲基,从而制得具有pH响应组装/解组装性能的双功能O-羧甲基壳聚糖-甲酰基苯硼酸(O-CMC-PBS),其在pH大于6.0时溶解,在pH≤4.5时自组装变浑浊离心形成沉淀,反调pH≥10.0时沉淀又溶解;然后利用5-亚甲基吡咯烷酮(5MP)可以在pH 6.0~8.5时通过共价键结合含有巯基的寡肽,在pH≥9.5时又可以释放所结合的巯基寡肽的特点,通过在O-CMC-PBS的氨基上引入5MP基团,从而制得了具有pH响应自组装/解自组装和巯基吸附/解吸附功能的双功能壳聚糖衍生物O-CMC-PBS-5MP。该材料在pH 6.0~8.0时捕获巯基寡肽,2小时吸附率可高达64%,最高吸附率可达95%;在pH≤3.0时,吸附巯基寡肽的材料则因发生自组装凝聚,离心即可实现巯基寡肽的富集;然后将沉淀溶解于少量蒸馏水中通过加入氨水调节pH≥10.0,沉淀复溶巯基寡肽释放到溶液中, 然后再调节pH≤3.0,富集材料沉淀,上清中即为富集的高纯度巯基寡肽,总回收率达到50%。本研究开发的巯基肽富集/浓缩材料O-CMC-PBS-5MP第一次实现了对巯基寡肽的无损富集和浓缩,为巯基寡肽的质谱分析奠定了基础。

Design,preparation and application of chuitosan derivatives materials for specific enriching thiol oligopeptides base on pH-responsive
Abstract:How to realize the non-destructive adsorption / desorption of thiol oligopeptides has always been a difficult problem in the field of thiol oligopeptide analysis. To solve this problem, chitosan (CTS) was used as the raw material, the phenylboric acid group was introduced at the position of chitosan C2, through the reaction of 3-formylphenylboric acid with the amino group on chitosan, and carboxymethyl was introduced at the position of chitosan C6 by chloroacetic acid under alkaline condition, so as to O-carboxymethyl chitosan-formylphenylboric acid (O-CMC-PBS) with pH responsive ability was prepared. It was dissolved at pH≥6.0, self-assembly becomes turbid and centrifuges to form precipitates at pH≤4.5, and precipitates was dissolved at pH≥10.0. Then, the 5-methylenepyrrolidone (5MP) had the characteristics of covalently bind thiol oligopeptides at pH 6.0-8.5 and release bound thiol oligopeptides at pH ≥9.5. Therefore, the chitosan derivative O-CMC-PBS-5MP with pH responsive self-assembly / disassembly and thiol oligopeptides adsorption/desorption functions was prepared by introducing 5MP group into the amino group of O-CMC-PBS, which can be covalently bonded to thiol oligopeptide at pH 6.0-8.0, and released at pH≥10.0; At pH 6.0-8.0, O-CMC-PBS-5MP dissolves in water and could specifically react with mercapto on oligopeptide to realize the function of binding thiol oligopeptides, the adsorption rate is up to 64% in 2 hours, and the maximum adsorption rate can reach 95%; At pH≤3.0, the material that adsorbs thiol oligopeptide is condensed due to self-assembly, and the thiol oligopeptide can be enriched by centrifugation; Then, the precipitate is dissolved in a small amount of distilled water to adjust the pH ≥10.0 by adding ammonia, the precipitate reconstituted thiol The oligopeptide is released into the solution, Adjust pH≤3.0 with formic acid again to realize the self-assembly precipitation of O-CMC-PBS-5MP and the enrichment of high-purity thiol oligopeptides in the supernatant, with a total recovery rate of 50%.The thiol oligopeptides enrichment / concentration material developed in this study, O-CMC-PBS-5MP, achieved the first non-destructive enrichment and concentration of thiol oligopeptides, which laid the foundation for mass spectrometry analysis of thiol oligopeptides.
