复方中药对腰椎韧带细胞作用的研究

摘要:目的:探究新拟复方中药(Compound Traditional Chinese Medicine, CTCM)对兔腰椎棘间韧带成纤维细胞(Interspinous Ligament Fibroblasts, ILFs)分泌胞外基质蛋白及其降解酶和内源性生长因子的影响。方法:体外分离培养新西兰大白兔ILFs,将第3代ILFs随机分为3组:培养基对照组、CTCM组(200μg/mL)、舒筋健腰丸对照组(S组,30μg/mL)。体外培养不同时间段后检测细胞活性,用RT-PCR检测不同组细胞内Ⅰ、Ⅲ型胶原、弹性蛋白、mTOR以及各生长因子和MMP-2、MMP-9基因表达量的变化,采用免疫荧光染色半定量检测不同组细胞Ⅰ、Ⅲ型胶原、弹性蛋白表达情况以及观察胶原组装形态,并采用Western blot定量检测不同组细胞Ⅰ、Ⅲ型胶原、弹性蛋白表达量的变化以及mTOR磷酸化的情况。结果:PCR结果显示CTCM组和S组在第3d能够显著上调ILFs内Ⅰ、Ⅲ型胶原、弹性蛋白以及与PI3K/AKT信号通路有关的mTOR基因的表达,并且能够上调EGF、bFGF、MGF基因的表达,免疫荧光染色结果显示CTCM组和S组的Ⅰ、Ⅲ型胶原、弹性蛋白在第3d显著增加,但胶原组装形态没有观察到明显的变化,Western blot结果显示CTCM组和S组在第3d和第4d能够显著上调ILFs内Ⅰ、Ⅲ型胶原、弹性蛋白的表达以及促进mTOR的磷酸化,此外,CTCM组和S组在5d时上调基质金属降解酶的表达。结论:CTCM能促进ILFs胞外基质蛋白及内源性生长因子的表达,而在后期能显著上调MMP-2和MMP-9的表达,促进ILFs的迁移,这对韧带的修复是有利的。?????

The study on the effect of Compound Chinese Medicine on lumbar ligament cells
Abstract:Objective: To investigate the effects of Compound Traditional Chinese Medicine (CTCM) on the expression of extracellular matrix proteins, degrading enzymes and endogenous growth factors in Interspinous LiThe study on the effect of Compound Chinese Medicine on lumbar ligament cellsgament Fibroblasts (ILFs) of rabbits. Methods: The ILFs of New Zealand white rabbits were isolated and cultured in vitro, and the third-generation ILFs were randomly divided into three groups: the normal control group, the CTCM group (200 μg / mL), and the Shujinjianwan group (S group, 30 μg / mL). Cell viability was measured after in vitro culture for different periods of time. The gene level of collagenⅠ, collagen Ⅲ, elastin, mTOR, growth factors and MMP-2, MMP-9 in different groups of cells was detected by RT-PCR. Immunofluorescence staining was used to semi-quantitatively detect the expression of collagenⅠ, collagen Ⅲ and elastin in different groups of cells and observe the morphology of collagen assembly. Besides, Western blot was used to quantitatively detect the expression of collagen, elastin and phosphorylated mTOR. Results: PCR results showed that the CTCM group and the S group could significantly up-regulate the gene level of type I, III collagen, elastin, and mTOR related to the PI3K / AKT signaling pathway after 3 days, and similar to EGF, bFGF, and MGF. Immunofluorescence staining results showed that collagen I, collagen III and elastin in the CTCM group and the S group increased significantly on the 3rd day, but no obvious changes were observed in the collagen assembly morphology. Western blot results showed that the expression of type I, III collagen, elastin, and phosphorylated mTOR of CTCM group and the S group were significantly up-regulated compared to normal group on the 3rd day and 4th day. In addition, the CTCM group and the S group down-regulated the expression of matrix metalloproteinase on the 1st day, and up-regulated on the 3rd and 5th days. Conclusion: CTCM promote the expression of ILFs extracellular matrix proteins and endogenous growth factors, and down-regulate matrix metalloproteinases in the early stage of repair, in addition, its significant up-regulation in the late stage of matrix shaping, which is conducive to the repair of ligaments.
