摘要:MALDI-TOF-MS常用于检测蛋白质、多肽、聚合物等大分子化合物,该方法具有灵敏度高、检测速度快、样品制备方便、高耐盐性、低样品需求量等优点。为了提高该方法的检测范围,新型纳米基质应运而生。例如石墨烯、碳点、多孔硅、金属纳米颗粒等。虽然这些纳米材料具有良好的基质性能,可以避免传统基质自身带来的干扰,但它们存在制备复杂、批量合成困难、不能同时用于富集和检测等缺点。本文采用自上而下的合成方法,合成了硼酸功能化的二维硼纳米片(2DBS),该方法方便,对设备要求低,制备简单,易于批量制备,且产品纯度高。实验表明,合成的2DBS在350 nm波长附近有明显的紫外线吸收,这为2DBS作为基质提供了条件,在正离子模式下,对糖类和氨基酸这类小分子化合物用于MALDI-TOF-MS分析,具有很高的检测灵敏度。由于硼酸根官能团的存在,还能特异性的富集带有邻二醇结构的糖类化合物,具有优异的富集效果,能同时达到富集和检测的目的。

Coupled Enrichment and MALDI-TOF-MS Analysis of Vicinal Diols Using As-Prepared Boron Nanosheets as Matrix
Abstract:MALDI-TOF-MS is commonly used to detect large molecular compounds such as proteins, peptides, polymers, etc. This method has the advantages of high sensitivity, fast detection speed, convenient sample preparation, high salt tolerance, and low sample requirements. In order to increase the detection range of this method, a new type of nano-matrix came into being. Examples include graphene, carbon dots, porous silicon, and metal nanoparticles. Although these nanomaterials have good matrix properties and can avoid the interference caused by the traditional matrix itself, they have the disadvantages of complex preparation, difficult batch synthesis, and cannot be used for enrichment and detection at the same time. This paper uses a top-down synthesis method to synthesize boric acid-functionalized two-dimensional boron nanosheets (2DBS). This method is convenient, requires low equipment, simple preparation, easy batch preparation, and high product purity. The experiments show that the synthesized 2DBS has obvious ultraviolet absorption near the 350 nm wavelength, which provides conditions for 2DBS as a matrix. In the positive ion mode, the small molecule compounds such as carbohydrates and amino acids are used for MALDI-TOF-MS analysis, which has a high detection sensitivity. Due to the presence of borate functional groups, it is also possible to specifically enrich carbohydrates with vicinal diols structure, which has excellent enrichment effects and can simultaneously achieve the purpose of enrichment and detection.
