FAP探针的设计合成及性质研究

摘要:生物蛋白酶在人体中发挥着很重要的作用,并且参与人体的新陈代谢和重要物质的合成。现阶段,已经证实酶和诸多疾病的产生有着密切关系,并对人体内生理的异常代谢和表达有着重要作用。FAP存在于成纤维细胞的基质中并异常表达,在上皮肿瘤细胞中存在异常的高表达,与诸多肿瘤细胞的发生和发展有着紧密联系,如何高效、灵敏地检测FAP成为了当前相当重要的研究工作之一。因此本文设计并合成了用于检测FAP的探针,借助FAP可以切断蛋白质末端的酰胺键机理,我们选择了Cbz-Gly-Pro-OH蛋白质作为响应多肽,并选择了自己设计的荧光素类荧光染料作为荧光团,在染料一端通过对氨基苄醇链接蛋白质,另一端缩合可以使染料红移的环二己胺醛基化合物,设计并合成了可用于FAP的体外检测和细胞成像的近红外染料荧光探针(NIR-FAP),最终测得探针对FAP的检测下限为1.0 U/L。

Design, Synthesis and Properties of FAP Probe
Abstract:In this paper………..Biological protease plays an important role in human body, and participates in metabolism and synthesis of important substances. At present, it has been proved that enzymes are closely related to the production of many diseases and play an important role in the abnormal metabolism and expression of human physiology. FAP (Fibroblast Activation Protein) is a serine protease, which exists in the matrix of fibroblasts and expresses abnormally. The high expression in many tumor cells, meanwhile development of these tumor cells. The detectiion of FAP has become a necessary research work. Therefore, this paper can design, synthesize and detect FAP, and the mechanism of cleavage of amide bond at the end of protein by FAP, we chose Cbz-Gly-Pro-OH as the response peptide, the fluorescein dye as the fluorescent group. At one end of the dye, the protein is linked by p-aminobenzyl alcohol, and the other end is condensed to make the dye red shift, near infrared dye fluorescence probe (NIR-FAP) for FAP detection in vitro and cell imaging was designed and synthesized, and the detectiong limit of probe is 1.0 U/L.
