miRNA398与miRNA408对UV-B辐射胁迫下拟南芥叶片光合功能及抗氧化系统的调控

摘要:植物microRNA参与各种非生物胁迫响应。本文研究了miRNA398和miRNA408对UV-B辐射下拟南芥叶片光合功能及抗氧化系统的调控。检测了光合作用相关的叶绿素荧光参数等指标、抗氧化系统相关的SOD、GSH酶活性及MDA含量。结果表明:在UV-B辐射下,过表达miRNA398转基因拟南芥与过表达miRNA408转基因拟南芥相比野生型植株的叶绿素荧光动力学参数Fv/Fm、qP数值降低、qN数值升高,影响植物的光合作用过程,降低光合效率;抗氧化系统酶类SOD、GSH活性降低,氧化物质MDA含量上升,活性氧积累,影响植物抵抗辐射逆境胁迫,损害植株生长发育过程。表明过表达miR398和过表达miR408的拟南芥在UV-B辐射胁迫下生长状况更差,对UV-B辐射胁迫的抗性降低,光合作用和抗氧化系统均受到影响,因此miR398与miR408对拟南芥抗逆性有重要的调控作用。

The regulations of miRNA398 and miRNA408 on photosynthetic function and antioxidant system of Arabidopsis thaliana under UV-B radiation stress?
Abstract:microRNA was involved in various abiotic stress responses in plants. In this paper, we studied the regulation of miRNA398 and miRNA408 on the photosynthetic function and antioxidant system of Arabidopsis thaliana leaves under UV-B radiation. The chlorophyll fluorescence parameters related to photosynthesis and antioxidant system related indicators including SOD, GSH enzyme activity and MDA content were detected. The results showed that the chlorophyll fluorescence kinetic parameters Fv / Fm, qP, and qN increased in wild-type plants compared to miRNA398 transgenic Arabidopsis and miRNA408 transgenic Arabidopsis under UV-B radiation. The photosynthesis process of plants reduces the photosynthetic efficiency; the activities of antioxidant enzymes SOD and GSH decrease, the content of oxidizing substances MDA rises, and the accumulation of active oxygen affects the resistance of plants to radiation stress and damages the growth and development of plants. It showed that overexpressed miR398 and miR408 Arabidopsis grew worse under UV-B radiation stress, resistance to UV-B radiation stress decreased, photosynthesis and antioxidant system were affected, so miR398 and miR408 Arabidopsis stress resistance has an important regulatory role.
