光学图谱技术研究进展

摘要:光学图谱技术是一种DNA单分子水平分析手段,能够对兆碱基对级别的DNA片段实现可视化,提供DNA上的远程序列信息。通过对DNA进行标记,光学图谱技术可以检测染色体结构变异。随着DNA拉伸方法的不断优化,光学图谱技术已经具备强大的定位功能,在DNA测序领域的应用十分广泛。本文主要包含DNA标记方法、DNA拉伸方法以及光学图谱技术的应用三个方面内容,梳理了近年来光学图谱相关技术的研究概况,以期为光学图谱技术的进一步优化和应用提供参考意见。

Advance in Optical Mapping
Abstract:Optical mapping(OM) is a DNA analysis tool, at a single DNA molecule level. It can visualize ultra-long DNA molecules, up to mega-basepairs, and reveal long range sequence information on DNA. After targeted labling, OM isallowed to interrogate genomicstructural variation.With the optimization of DNA stretching methods, OM possess a powerfullocalization capabilityand is widely used as as auxiliary technique in the field of DNA sequencing.The article mainly includes the three aspects ofDNA labeling, DNAelongation and application of OM, aiming to provide some guidance for further optimization and application of OM.
