甲基斑蝥胺对胃癌细胞的毒作用及其机制研究

摘要:[目的]研究甲基斑蝥胺对正常胃黏膜上皮细胞GES-1和胃癌细胞NCI-N87、MKN-45增殖、周期和凋亡的影响及其作用机制。[方法]采用Cell Counting Kit-8 (CCK-8) 试剂盒检测MCA对正常胃黏膜上皮细胞GES-1和胃癌细胞NCI-N87、MKN-45生存活性的影响;用钙离子依赖性磷脂结合蛋白-异硫氰酸荧光素/碘化丙啶(Annexin V-FITC/PI)双标记方法检测经MCA作用后两种胃癌细胞的凋亡率;用细胞周期检测试剂盒检测经MCA作用后两种胃癌细胞周期变化情况;采用Western-blot检测MCA暴露对胃癌细胞NCI-N87的Survivin蛋白和NF-κB蛋白表达水平的影响。[结果]甲基斑蝥胺对胃癌细胞NCI-N87和MKN-45的增殖抑制作用大于正常胃黏膜上皮细胞GES-1(P<0.05);与对照组相比,甲基斑蝥胺暴露引起人胃癌细胞NCI-N87和MKN-45的凋亡率明显升高(P<0.05);与对照组相比,甲基斑蝥胺暴露能引起人胃癌细胞NCI-N87和MKN-45细胞周期阻滞(P<0.05);与对照组相比,甲基斑蝥胺暴露引起胃癌细胞NCI-N87内Survivin蛋白和NF-κB蛋白表达水平明显降低(P<0.05)。[结论]甲基斑蝥胺可以引起人胃癌细胞NCI-N87和MKN-45增殖抑制、凋亡率增加、细胞周期阻滞、Survivin蛋白和NF-κB蛋白表达水平降低,对胃癌细胞具有明显的细胞毒性作用。

Toxic Effect and the Underlying Mechanism of Methyl-cantharidimide on human gastric cancer cells
Abstract: [Object] Methyl-Cantharidimide (MCA) is a derivative of cantharidin which has potential anticancer activity. This study investigates the cytotoxic effects of MCA on human gastric cancer cells. Human gastric cancer cells NCI-N87 and MKN-45, and normal human gastric epithelium immortalized cells GES-1 were treated with a series of concentrations of MCA, the proliferation rates of these cells was examined using CCK-8 assay. Cell cycle and cell apoptosis were determined using Flow Cytometry. Furthermore, Western blot was used to evaluate biomarkers associated with cell cycle and apoptosis. [Results] Methylcantharidin inhibited cell proliferation in human gastric cancer cells NCI-N87 and MKN-45 in a dose- and time-dependent manner; Compared with the control group, MCA exposure arrested NCI-N87 cells in S phase cell cycle and arrested MKN -45 cells in G-1 phase cell cycle (P <0.05); Compared with the control group, MCA significantly increased the apoptosis rate of human gastric cancer cells NCI-N87 and MKN-45 (P <0.05); compared with the control group, MCA significantly reduced the expression levels of survivin protein and NF-κB protein in gastric cancer cells NCI-N87 (P <0.05).[Conclusion] These results suggest that MCA has cytotoxic effect on NCI-N87 and MKN -45 cells. Its mechanism of action is by inducing cell cycle arrest and promoting cell apoptosis through inhibiting survivin and NF-κB. MCA could be developed into a novel drug for the treatment of human gastric carcinoma.
