LuxR在睾丸酮丛毛单胞菌对甾体激素降解中的作用

摘要:LuxR蛋白在睾丸酮丛毛单胞菌对甾体激素降解中的作用是细菌的一种基因调控蛋白,LuxR蛋白在睾丸酮丛毛单胞菌(Comamonas testosteroni,C.T)ATCC11996中表达。本文利用基因同源重组技术,构建LuxR基因敲除菌株MC.T。利用HPLC对野生型C.T菌与突变株MC.T的甾体激素降解对比分析,MC.T降解睾丸酮的水平上明显提高;对雌二醇、胆固醇、雌酮的降解差异较小。结果表明, LuxR蛋白对C.T菌中降解甾体类化合物代谢起到调控作用,对于睾丸酮的代谢具有负调控作用。

Effect of LuxR on degradation of steroidal hormones by Comamonas testosteroni
Abstract:LuxR in testosterone tufting effect on degradation of steroid hormones in the bacterium is one of the types of bacteria gene regulatory proteins, LuxR protein in testosterone fetlock single cell bacteria (Comamonas testosteroni, C.T) expression in ATCC11996.In this paper, MC.T of LuxR gene knockout strain was constructed by homologous recombination technique.The steroid hormone degradation analysis of wild-type C. T. and mutant MC.T by HPLC showed that the testosterone degradation level of MC.T was significantly increased.There was little difference in the degradation of estradiol, cholesterol and estrone.The results showed that LuxR protein had a regulatory effect on the metabolism of degraded steroidal compounds in C.T bacteria, and had a negative regulatory effect on the metabolism of testosterone.
