硒化秦巴硒菇对A549细胞的抗肿瘤作用研究

摘要:目的初步探讨Se-ABM对A549细胞DNA损伤和细胞周期阻滞的分子机制。方法体外培养HT29、HepG2、HeLa和A549细胞,用Se-ABM和拓扑替康(Topotecan)处理A549细胞,分别用CCK-8实验,流式细胞分析和免疫印迹实验检测Se-ABM对A549细胞的抗肿瘤作用。结果 Se-ABM能够抑制HT29、HepG2、HeLa和A549细胞的增殖,其中对A549细胞的增殖抑制最为明显。细胞流式实验表明Se-ABM能够阻滞A549细胞在G2/M期并呈浓度依赖性。Western blot 实验结果显示,Se-ABM 能够使 A549 细胞中DNA损伤相关因子ATM,ATR,p53,γ-H2AX的表达量明显升高,同时诱导细胞周期阻滞在G2/M期。此外,Se-ABM能够调控凋亡相关蛋白的表达,使BAX,BRCA1,Bcl-2,Case3和Case 9的表达发生明显变化。结论 Se-ABM在体外能够明显诱导DNA损伤,从而导致细胞凋亡。对比拓扑替康,Se-ABM具有更高的抑制A549细胞增殖的能力。?????

Antitumor efficacy of Se-Agaricus blazei Murill (Se-ABM) on A549 cells
Abstract:Objective: Preliminary explore the molecular mechanism of Se-ABM on cell DNA damage and cell cycle arrest of A549 cells. Methods: HT29, HepG2, HeLa and A549 cells were cultured in vitro, and exposure of A549 cells to Se-ABM and Topotecan. The antitumor efficacy of A549 cells was then detected by CCK-8 assay, cell flow assay and western blots assay, respectively. Results: Se-ABM inhibited the proliferation of HT29, HepG2, HeLa and A549 cells and the inhibition of proliferation of A549 cells is most obvious. Cell flow cytometry experiments showed that Se-ABM nduced the G2/M phase arrest of the cell cycle in A549 cells and exhibited a dose-dependent effect in A549 cells. Westen blot experiment results showed that Se-ABM highly increased the expression of DNA damage-related factors ATM, ATR, p53 and γ-H2AX in A549 cells and induced cell cycle arrest in G2/Mphase. Moreover, Se-ABM regulated the expression of apoptosis-related proteins and the expression of BAX, BRCA1, Bcl-2, Case3 and Case 9 showed significantly changs. Conclusion: Se-ABM can induce DNA damage and induction of cell apoptosis in vitro. When compared to topotecan, Se-ABM has a higher potency to inhibit the growth of A549 cells. ???
