富硒绿茶对四氯化碳诱导的小鼠肝纤维化的影响

摘要:目的:探讨富硒绿茶对四氯化碳(CCl4)诱导的雄性C57BL/6J小鼠肝纤维化的影响。方法:雄性C57BL/6小鼠48只,随机分为四组:CON组、CCl4组、CCl4+普通绿茶组及CCl4+富硒绿茶组,每组12只。CCl4组和茶干预组每周两次注射1ml/kg的CCl4,CON组则每周两次注射相同量的纯橄榄油,干预组小鼠自由饮用茶溶液。干预4周之后处死,收集样品。结果:CCl4组小鼠出现明显的肝纤维化,肝功能严重损害。肝脏组织病理学显示茶干预改善肝纤维化,降低肝脏I型胶原(COL1A1)、III型胶原(COL3A1)和平滑肌激动蛋白α(α-SMA)水平。茶干预显著提高了肝脏超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)、过氧化氢酶(CAT)的活性,降低了肿瘤坏死因子-α(TNF-α)、转化生长因子(TGF-β1)、白介素-1β(IL-1β)、白介素-6(IL-6)和丙二醛(MDA)的含量。和普通绿茶比较,富硒绿茶效果更为明显。结论:两种茶均可改善CCl4诱导的小鼠肝纤维化、氧化应激和炎症反应,富硒绿茶效果优于普通绿茶。

Effects of Selenium-enriched green tea on liver fibrosis induced by carbon tetrachloride in mice
Abstract:Objective: To investigate the effect of selenium-enriched green tea on liver fibrosis induced by carbon tetrachloride (CCl4) in male C57BL/6J mice. Methods: A total of 48 male C57BL/6 mice were randomly divided into four groups as follows: CON group, CCl4 group, CCl4+common tea group and CCl4+selenium-rich tea group, with 12 animals each group. Liver fibrosis was induced by intraperitoneal injection of 0.1 ml/100 g CCl4 (10% CCl4 in olive oil) twice per week. The mice in intervention groups drank tea and selenium-enriched tea solution for 4 weeks. At the end of the 28 days, the mice were deprived of food for 12 h and sacrificed. Results: CCl4 resulted in hepatic fibrosis and damaged hepatic function. Histological analysis of liver showed that tea interventions alleviated hepatic fibrosis and decreased hepatic collagen type I (COL1A1), collagen type III (COL3A1) and α-smooth muscle actin (α-SMA). Both tea interventions significantly increased serum levels of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT), and decreased tumor necrosis factor-α (TNF-α), transforming growth factor-beta1 (TGF-β1), interleukin-1β (IL-1β) and interleukin-6 (IL-6), malondialdehyde (MDA) content. Compated with in CCl4+common tea group, The improvemment was more obvious in CCl4+selenium-rich tea group. Conclusion: Both types of tea improve CCl4-induced liver fibrosis oxidative stress and inflammatory response in mice, with pronounced efffect in selenium-rich tea. ?????
