栀子果胶的制备与质量分析

摘要:目的:探究从栀子渣中提取果胶(GJP)的方法,并对其进行结构研究和质量分析。方法:采用酸-酶联合提取的方法从栀子渣中提取GJP;利用高效分子排阻色谱法测定其分子量,采用高效离子交换色谱法检测单糖组成,并采用红外、1H和13C NMR波谱分析对其进行结构表征;分别采用酸碱化学滴定法测定其酯化度,间羟基联苯法测定半乳糖醛酸含量,蒽酮硫酸法测定中性糖含量。结果:测定GJP的相对分子量为44.10 KDa,其单糖组成包括半乳糖醛酸、半乳糖、鼠李糖、阿拉伯糖、木糖和甘露糖、葡萄糖、岩藻糖等7种,GJP包括62.78%的半乳糖醛酸聚糖和27.35%的鼠李糖半乳糖醛酸聚糖两种结构。测得GJP的总半乳糖醛酸含量88.10%,中性糖含量7.65%,酯化度72%,属于高酯果胶。结论:从栀子渣中可提取获得栀子果胶副产物,可为其综合加工利用提供参考。

Preparation and Quality Analysis of Gardenia jasminoides Pectin
Abstract:Objective: To explore the extraction method of pectin (GJP) from Gardenia jasminoides Ellis, as well as its structure characterization and quality analysis. Methods: GJP was extracted from Gardenia jasminoides slag by thecombined acid-enzyme extraction.Its molecular weight was determined by HPSEC method, and the composition of monosaccharides was detected by HPEAC-PAD.Its structurewas analyzed by infrared, 1H and 13C NMR spectroscopy.Acid - alkali chemical titration method was used to determine the degree of esterification, the meta-hydroxybiphenyl method was used to determine the content of galacturonic acid, and the anthrone sulfuric acid method was used to determine the content of neutral sugars. Results: The relative molecular weight of GJP was 44.10 KDa, and it was composed of 7 monosaccharideinclude galacturonic acid, galactose, rhamnose, arabinose, xylose, mannose, glucose, and fucose. GJP was HG type pectin. The galacturonic acid content of GJP was measured to be 88.1%, neutral sugar content was 7.65%,. The degree of esterification was 72%, which indicated that it was high DE pectin. Conclusion: GJPcan be extracted from gardenia slag, which can be provided as a valuble by-product.
