维生素D对颗粒所致急性肺损伤的作用及机制研究

摘要:目的:研究维生素D在颗粒所致急性肺损伤中的作用。方法:C57BL/6J小鼠采用非暴露式气管灌注二氧化硅颗粒构建急性肺损伤模型,通过HE、肺泡灌洗液细胞总数和分数计数、总蛋白量检测,炎症相关蛋白p-P65、细胞凋亡及DNA损伤免疫组化等实验技术检测维生素D对颗粒所致小鼠肺组织损伤的作用。结果:维生素D对颗粒所致小鼠急性肺损伤具有缓解作用。(1)与对照组相比,颗粒组小鼠肺组织炎症细胞浸润明显,且肺重增加(P<0.05);肺泡灌洗液中细胞总数和分数、总蛋白量明显增加(P<0.05),给予维生素D干预后,肺组织炎症损伤明显减轻(P<0.05);(2)与对照组相比,颗粒组小鼠DNA氧化损伤、细胞凋亡水平显著增加;给予维生素D干预后,DNA氧化损伤及细胞凋亡水平降低(P<0.05)。结论:颗粒暴露可使肺组织炎症细胞浸润、肺泡损伤引起总蛋白增加,细胞凋亡及DNA损伤水平升高,给予维生素D干预可使以上指标明显降低。表明维生素D对颗粒所致急性肺损伤具有缓解作用

Study on the effect and mechanism of vitamin D of acute lung injury caused by particles
Abstract:Objective: To investigate the role of vitamin D in particles-induced acute lung injury and explore the potential mechanism. Methods: A model of acute lung injury was constructed by non-exposed tracheal instillation of particles in C57/BL6 mice. The effects of vitamin D on particles-induced lung tissue injury were examined by HE, alveolar lavage fluid total cells and differential cells, total protein amount assay, IHC etc. Results: Vitamin D had protective effects on acute lung injury: (1) Compared with the Ctrl group, the infiltration of inflammatory cell was significant and the lung weight increased (P<0.05)in the particles group ; the total cells, differential cells and the total protein amount in the alveolar lavage fluid were significantly increased (P<0.05), the inflammatory damage in the lung tissue was significantly reduced after the administration of vitamin D (P<0.05); (2) Compared with the Ctrl group, the oxidative DNA damage and apoptosis in the particles group were significantly increased, the expression levels of 8-oxo-dG and CC3 were reduced after vitamin D intervention(P<0.05). Conclusion: Particles exposure can cause inflammatory cell infiltration and alveolar damage, the levels of apoptosis and DNA damage were increased, intervention with vitamin D can significantly reduce the above indicators, indicating that vitamin D has a alleviating effect on particles-induced acute lung injury.
