砷致HaCaT细胞恶性转化过程中铁死亡的变化情况

摘要:【目的】探究砷人皮肤角质形成细胞(HaCaT)恶性转化中铁死亡的变化,为砷致癌机制的深入研究提供必要的基础资料。【方法】收集经0.0或1.0μM亚砷酸钠(NaAsO2)处理至35代过程中的各关键代数(0、1、7、14、21、28、35)的HaCaT细胞后,用试剂盒检测细胞内的丙二醛(MDA)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶4(GPX4)以及亚铁离子含量,利用Western-blot检测砷致细胞恶性转化过程中xCT和3B8A21的蛋白表达水平。【结果】在砷致细胞恶性转化过程中,GPX4水平和GSH/GSSG水平呈现升高趋势,且与对照组相比差异具有统计学意义(P<0.05);细胞内丙二醛(MDA)和Fe2+含量在砷致细胞恶性转化后期呈现下降趋势。【结论】本实验研究结果表明,在砷致细胞恶性转化过程中铁死亡水平呈降低趋势,砷可能通过抑制铁死亡促进细胞的恶性转化。

Changes of ferroptosis during the malignant transformation of HaCaT cells induced by arsenite
Abstract:[Objective] To investigate the changes of ferroptosis in arsenic-induced malignant transformation of human keratinocytes (HaCaT). [Method] After collecting key generations (0, 1, 7, 14, 21, 28, 35) of HaCaT cells exposed to sodium arsenite at a concentration of 0.0 and 1.0 μM, the levels of glutathione, glutathione peroxidase 4 (GPX4) and Fe2+ were determined. Futhermore, the protein expression levels of xCT and 3B8A21 also were detected by Western-blot. [Results] During the arsenite-induced malignant transformation of HaCaT cells, GPX4 level and the ratio of GSH/GSSG showed an up-regulated trend, which was statistically significant compared with the control group (P<0.05). The levels of malondialdehyde (MDA) and Fe2+ showed down-regulated trend after 14 passages during malignant transformation caused by arsenite (P<0.05). [Conclusion] Ferroptosis was inhibited during the malignant transformation of arsenite-induced HaCaT cells, which might play a role in the malignant transformation.
