姜黄素对电离辐射诱导胶质瘤免疫原性细胞死亡的影响及其机制研究

摘要:目的:探究姜黄素对电离辐射诱导胶质瘤免疫原性细胞死亡的影响及其机制。方法:CCK-8实验检测姜黄素对人和鼠胶质瘤细胞的毒性作用。流式细胞术检测细胞凋亡、胶质瘤细胞表面CD47表达。流式细胞术和免疫荧光实验检测钙网蛋白细胞膜表面暴露。生物发光实验检测ATP释放。Western Blot实验检测HSP70水平。疫苗接种实验检测小鼠体内是否发生免疫原性细胞死亡。结果:U251和GL261胶质瘤细胞活性随姜黄素浓度的增加而降低。X射线照射前用姜黄素处理24小时,能够明显提高常氧和乏氧(1% O2)胶质瘤细胞的凋亡率。姜黄素能够增强辐射诱导的胶质瘤钙网蛋白细胞膜表面暴露以及HSP70、ATP释放。通过疫苗接种实验证明,姜黄素能够增强辐射诱导小鼠体内胶质瘤免疫原性细胞死亡。结论:姜黄素增强辐射诱导的胶质瘤免疫原性细胞死亡,刺激体内产生强烈的抗肿瘤免疫反应。

Effect of Curcumin on ionizing radiation-induced immunogenic cell death in glioma cells and its mechanism
Abstract:Objective: This study aims to investigate the effect of curcumin on the ionizing radiation-induced immunogenic cell death in glioma cells and its mechanism. Methods: Apoptosis and CD47 expression on the surface of glioma cells were detected by flow cytometry. Surface exposure of calreticulin was measured by flow cytometry and immunofluorescence. ATP release was detected by bioluminescence. The level of HSP70 was detected using western blot. Immunogenic cell death in vivo was detected by vaccination experiment. Results: The viability of U251 and GL261 glioma cells decreased with the increase of curcumin concentration. Glioma cells were incubated with curcumin for 24 hours before X-ray irradiation can significantly increase the apoptosis rate in normoxia and hypoxia (1% O2). Curcumin can enhance radiation-induced cell surface exposure of CRT, and increase radiation-induced HSP70 and ATP release in glioma cells. Conclusion: Ionizing radiation can induce immunogenic cell death in glioma cells. Curcumin can be used as a radiation-induced immunogenic cell death sensitizer, enhance the tumor immunogenicity induced by IR and activate a strong anti-tumor immune response.
