高密度发酵大肠杆菌产酪醇

摘要:为进一步提高本实验室高产酪醇菌株的酪醇产量,通过响应面方法优化摇瓶发酵条件,并通过5L发酵罐进行发酵扩大实验。响应面优化摇瓶结果为:培养基中Ca2+浓度1.63 mmol/L、硫酸铵3.28g/L、温度27℃时,发酵时间60h,酪醇产量为2.80g/L,相较于已发表重组大肠杆菌产酪醇水平提高了80%。5L发酵罐补料分批发酵,发酵时间48h,OD600为36±0.5,相较优化前提高了10左右;酪醇产量为5.06g/L,相较摇瓶产量提高了74.5%,细胞密度和产量明显提升,为工业生产酪醇奠定了基础。

High-density Cultivation of Recombinant Escherchia coli for Tyrosol
Abstract:In order to further increase the tyrosol production of the high-yield tyrosol strains in our laboratory, the shake flask fermentation conditions were optimized by response surface method, and the fermentation expansion experiment was carried out through a 5L fermentor.The results of the response surface optimization shake flask are: the concentration of Ca2+ in the medium is 1.63 mmol/L, the ammonium sulfate is 3.28g/L, the temperature is 27℃, the fermentation time is 60h, and the tyrosol production is 2.80g/L, which is compared with the published recombinant large intestine.The level of tyrosol produced by the bacterium increased by 80%.The 5L fermenter was fed-batch fermentation, the fermentation time was 48h, and the OD600 was 36±0.5, which was an increase of about 10 compared to before optimization; the tyrosol yield was 5.06g/L, which was 74.5% higher than the shake flask yield, and the cell density and cell density.The output has increased significantly, laying the foundation for industrial production of tyrosol.
