副格氏乳杆菌FTJWQ2L9通过提高短链脂肪酸水平缓解小鼠高尿酸血症

摘要:为探究副格氏乳杆菌对高尿酸血症的影响,该研究选用30只5周龄的雄性昆明小鼠,实验期间每天灌胃109CFU副格氏乳杆菌,最后2周采用氧嗪酸钾和次黄嘌呤处理引起小鼠高尿酸血症。实验结束后,测定小鼠血清及尿液尿酸浓度、黄嘌呤氧化酶活性和肾损伤程度,并分析短链脂肪酸水平和肠道菌群变化。结果表明,副格氏乳杆菌FTJWQ2L9能够有效降低小鼠血清和尿液尿酸浓度,抑制肝脏和血清中黄嘌呤氧化酶(XOD)的活性,并能够使小鼠的肾损伤得以恢复。此外,副格氏乳杆菌FTJWQ2L9还能够提高小鼠粪便6种短链脂肪酸(SCFAs)的浓度,上调Ruminococcaceae UCG-005、Prevotellaceae NK3B31 group、Ruminococcaceae UCG-014等的丰度。菌群、短链脂肪酸水平与尿酸代谢指标的相关性分析结果显示,血清尿酸及黄嘌呤氧化酶活性与短链脂肪酸水平及产短链脂肪酸相关菌群具有较强相关性。因此,副格氏乳杆菌FTJWQ2L9对短链脂肪酸水平及其相关菌群的调节可能是其发挥降尿酸作用的重要途径。该研究为探讨益生菌缓解高尿酸血症的机制提供了新的思路,为后续研究提供了依据。

Lactobacillus paragasseri FTJWQ2L9 relieves hyperuricemia in mice by increasing short-chain fatty acids production
Abstract:In order to explore the effect of Lactobacillus paragasseri on hyperuricemia, 30 male Kunming mice aged 5 weeks were selected for this study. During the experiment, 109 CFU of Lactobacillus paragasseri was administered daily, and potassium oxonate and hypoxanthine were daily administrated for the last 2 weeks to make mice hyperuricaemia. After the animal experiment, uric acid concentration in serum and urine, xanthine oxidase activity and the degree of kidney damage were measured, and the levels of short-chain fatty acids and changes in intestinal flora were analyzed. The results showed that Lactobacillus paragasseri FTJWQ2L9 can effectively reduce the concentration of uric acid in serum and urine of mice, inhibit the activity of xanthine oxidase (XOD) in the liver and serum, and restore the kidney damage in mice. In addition, Lactobacillus paragasseri FTJWQ2L9 could also increase the concentration of 6 short-chain fatty acids in mouse feces, and up-regulate the abundance of Ruminococcaceae UCG-005, Prevotellaceae NK3B31 group and Ruminococcaceae UCG-014. The results of correlation analysis between intestinal flora, short-chain fatty acid (SCFA) levels and uric acid metabolism index showed that uric acid and xanthine oxidase activities in serum had a strong correlation with short-chain fatty acid concentrations and SCFA-producing related taxa. Therefore, the regulation of Lactobacillus paragasseri FTJWQ2L9 on the level of SCFAs and its related taxa may be an important way for it to lower uric acid. This study provides a new idea for exploring the mechanism of probiotics to relieve hyperuricemia and provides a basis for follow-up research.
