棕榈酸对人前列腺癌DU145细胞增殖和迁移的影响

摘要:本研究主要探索棕榈酸(Palmitic acid, PA)在前列腺癌(Prostatic cancer, PCA)发生发展中的作用及机制。PA是一种长链饱和脂肪酸,是脂肪酸合成酶(Fatty acid synthase, FASN)的主要产物。高脂饮食已经被广泛认为与PCA的发生发展有关,但外源性添加饱和脂肪酸对PCA发生发展的影响少有报道。首先我们通过检索TCGA数据库发现FASN在前列腺癌组织中呈现高表达。然后采用MTT法、克隆形成实验以及细胞划痕实验分别研究了PA对DU145细胞增殖、克隆形成能力以及细胞迁移能力的影响。最后,通过Western blot实验检测N-钙黏蛋白和E-钙黏蛋白等上皮间质转化(Epithelial-mesenchymal transition, EMT)相关标志物及FASN的表达情况。结果表明,在PA处理下,DU145细胞增殖、迁移能力增强,并一定程度影响EMT相关标志物的表达。这些结果提示PA能够促进PCA的发生发展,并一定程度诱导EMT过程。

Effects of palmitic acid on DU145 cell proliferation and migration
Abstract:This study mainly explored the role and mechanism of palmitic acid (PA) in the development of prostate cancer (PCA). Palmitic acid, a long-chain saturated fatty acid, is the main product of fatty acid synthase (FASN). The high-fat diet has been widely believed to be associated with the development of PCA, but the effect of exogenous addition of saturated fatty acids on the occurrence and development of PCA has not been reported. By searching TCGA database, we found that FASN expression was high in tissue of PCA. Then, the effects of PA on the ability of DU145 cell proliferation, clone formation and cell migration were detected by MTT method, clone formation experiment and wound-healing experiments. Finally, we detected the expression of epithelial-mesenchymal transition (EMT) markers such as N-Cadherin and E-Cadherin by western blot experiment. Results showed that PA diet can enhanced the proliferation and migration ability of DU145 cells, and to some extent affected the expression of EMT related markers. These results suggest that PA can promote the development of PCA and induce EMT process.
