氚水对斑马鱼的心脏发育毒性作用及机制研究

摘要:目的:探讨氚水对斑马鱼的心血管毒性及机制。方法:2~3 hpf斑马鱼胚胎用3.7*10^4 Bq/ml氚水进行染毒,观察斑马鱼心率变化;对氚水暴露60 h和120 h后的胚胎进行转录组学检测;用RT-PCR实验检测TnC/TPM4通路相关基因(tnnc1a、tpm4a、tpm4b)在氚水暴露后的表达水平;CRISPR/Cas9技术建立tnnc1a、tpm4a基因敲除斑马鱼模型,并观察氚水染毒后心脏发育情况。结果:斑马鱼在36、60 hpf心率明显加快,tnnc1a、tpm4a、tpm4b基因表达水平在36 hpf、48 hpf、60 hpf显著下调,在120 hpf显著上调。tnnc1a或tpm4a基因敲除后,氚暴露36 hpf、60 hpf、120 hpf时心率显著下降;tnnc1a基因敲除后斑马鱼胚胎畸形率升高,tpm4a基因敲除后,斑马鱼胚胎形态未发生明显改变。结论:氚水暴露通过TnC/TPM4通路诱导斑马鱼胚胎/幼鱼心率升高,其中tnnc1a与tpm4a对心率的调控主要在胚胎发育后期(120 hpf),tnnc1a对心脏及全身成长发育具有重要调控作用。

Study on the Toxicity and Mechanism of Tritiated Water on the Cardiac Development of Zebrafish
Abstract:Objective: To investigate the cardiovascular toxicity of tritiated water to zebrafish and its mechanism. Methods: 2~3 hpf zebrafish embryos were exposed to 3.7*10^4 Bq/ml tritiated water to observe the changes of heart rate of zebrafish RT-PCR was used to detect the expression levels of tnnc1a, tpm4a, tpm4b related genes after exposure to tritium water; CRISPR/Cas9 technology was used to establish tnnc1a, tpm4a gene knockout zebrafish model, and to observe the cardiac development after exposure to tritium water. Results: the heart rate of zebrafish increased significantly at 36 hpf and 60 hpf, and the gene expression levels of tnnc1a, tpm4a and tpm4b decreased significantly at 36 hpf, 48 hpf and 60 hpf, and increased significantly at 120 hpf. After tnnc1a or tpm4a gene knockout, the heart rate decreased significantly after tritium exposure at 36 hpf, 60 hpf and 120 hpf; the malformation rate of zebrafish embryos increased after tnnc1a gene knockout, but the morphology of zebrafish embryos did not change significantly after tpm4a gene knockout. Conclusion: exposure to tritiated water can induce the increase of heart rate in zebrafish embryos/juveniles through TnC/TPM4 pathway. The regulation of tnnc1a and tpm4a on heart rate is mainly in the late embryonic development (120 hpf), and tnnc1a plays an important role in the growth and development of heart and whole body.
