两歧双歧杆菌缓解洛哌丁胺诱导便秘的机制存在种内差异

摘要:本文利用3株不同来源的两歧双歧杆菌(M130R01M51、FHuNMY2M1、FHNFQ21M6)分析两歧双歧杆菌缓解便秘机制中存在的差异。将25只雄性BALB/c小鼠随机分为对照组、模型组与3个菌处理组。采用盐酸洛哌丁胺(10mg/(kg bw))诱导便秘模型,同时对小鼠灌胃两歧双歧杆菌悬液(10^9CFU/mL),共计8天。结果显示3株两歧双歧杆菌都能够通过抑制水通道蛋白的表达从而有效降低粪便含水量,同时对小肠推进具有促进作用。但是这三株菌对肠蠕动的作用机制却存在显著差异,其中,FHNFQ21M6能够促进兴奋型胃肠活性肽的分泌,增加机体代谢过程中的脑源性神经营养因子,并促进粪便中短链脂肪酸的产生,从而对肠道蠕动存在积极作用。而其他两株菌则主要是通过减少抑制型神经递质的分泌从而有利于肠道蠕动。因此,两歧双歧杆菌能够以不同的机制来缓解便秘,存在显著的种内差异。

The mechanisms of Bifidobacterium bifidum relieving loperamide induced constipation are intraspecially different
Abstract:In this study, three Bifidobacterium bifidum strains from different sources (M130R01M51, FHUNMY2M1, and FHNFQ21M6) were used to analyze the differences in the mechanism of B. bifidum relieving constipation. Twenty-five male BALB/c mice were randomly divided into control group, model group and three strain intervention groups. The constipation model was induced by loperamide hydrochloride (10mg/(kg bw)), and at the same time, B. bifidum (10^9CFU/mL) were administered to the mice for 8 days. The results showed that all the three B. bifidum strains could effectively reduce fecal water content by inhibiting the expression of aquaporin 8, and at the same time had a promoting effect on intestinal propulsion. However, the three strains had significant differences in the mechanism of intestinal peristalsis. Among them, FHNFQ21M6 could promote the secretion of excitatory gastrointestinal active peptides and brain-derived neurotrophic factor. Besides, it could stimulate the production of short-chain fatty acids in the feces, so it had a positive effect on intestinal peristalsis. The other two strains are mainly beneficial to intestinal peristalsis by reducing the secretion of inhibitory neurotransmitters. Therefore, B. bifidum can relieve constipation by different mechanisms, and there are significant intraspecies differences.
