PDMS微阵列中真皮成纤维细胞向毛乳头细胞转化的研究

摘要:本文旨在探究聚二甲基硅氧烷(PDMS)微孔三维环境是否能够诱导小鼠真皮成纤维细胞向毛乳头样细胞转化。利用3D打印技术设计出不同孔径大小的PDMS微孔阵列,接种小鼠真皮成纤维细胞形成细胞聚集体,分别通过RT-PCR的碱性磷酸酶染色的方法检测细胞聚集体中毛乳头特异性标志物的基因和蛋白的表达。实验结果表明,孔径大小的改变可以控制所形成的细胞聚集体的大小,并且毛乳头特异性标志物在基因水平和蛋白水平上都有增强。此研究证明了PDMS微孔环境诱导小鼠真皮成纤维细胞重编程为毛乳头样细胞的可能性。

Transformation of dermal fibroblasts into dermal papilla cells in PDMS micropores
Abstract:The purpose of this study was to investigate whether the three-dimensional environment of PDMS micropore array could induce the transformation of mouse dermal fibroblasts into dermal papilla-like cells.PDMS micropore arrays with different pore sizes were designed by 3D printing technology and add mouse dermal fibroblasts to form cell aggregates. Gene and protein expressions of specific markers of dermal papilla in the cell aggregates were detected by alkaline phosphatase staining by RT-PCR, respectively.The results showed that the change of pore size could control the size of the aggregates formed, and it was enhanced at both the gene and protein levels.It was suggested that aggregates formed by mouse dermal fibroblasts in PDMS micropore arrays could be reprogrammed into dermal papilla-like cells.
