功能化DNA四面体用于双肿瘤标志物的荧光响应及活细胞成像

摘要:为了实现癌细胞的精准识别,本文基于碱基互补杂交简单地组装了功能化DNA四面体,其中内置了AS1411序列和共识别miRNA-21和survivin mRNA两种肿瘤标志物的燃料刺激驱动核酸反应元件。通过电泳对其表征后,使用荧光光谱和激光共聚焦成像考察了其对两种肿瘤标志物的识别能力。结果表明,该功能化DNA四面体能够顺利进入细胞内,仅在两种肿瘤标志物的共同触发下能够出现强烈的荧光,在癌细胞和正常细胞中显示明显的荧光信号差异。该功能化DNA四面体具有良好的生物相容性,可在活细胞内可视化miRNA-21和survivin mRNA的共表达情况,为精准有效识别癌细胞提供了有潜力的平台。

Functionalized DNA tetrahedrons for dual tumor markers\' fluorescence response and imaging in living cells
Abstract:For accurate identification of cancer cells, a functionalized DNA tetrahedron based on base complementation hybridization was assembled, embedded with AS1411 sequence and fuel stimulation-driven nucleic acid reaction part for response of tumor markers (miRNA-21 and survivin mRNA). After being characterized by electrophoresis, fluorescence spectrometer and confocal laser microsocpe were used to investigate the ability to recognize two tumor markers. The results showed that the functionalized DNA tetrahedron could be internalized into cells, and strong fluorescence appeared only in the presence of two tumor markers. Significant differences in fluorescence signals were observed between cancer cells and normal cells. Assembled functionalized DNA tetrahedron has good biocompatibility and can visualize the co-expression of miRNA-21 and survivin mRNA in living cells, providing a potential platform for accurate and effective identification of cancer cells.
