PYCR1在卵巢癌细胞中的表达及作用机制研究

摘要:目的:探讨PYCR1在卵巢癌细胞中的表达,探讨其对卵巢癌细胞的增殖、迁移、侵袭以及EMT的影响。方法:使用GEPIA分析PYCR1在卵巢癌中的表达水平;Q-PCR、Western blotting以及免疫荧光技术检测PYCR1在正常卵巢细胞(IOSE 80)以及卵巢癌细胞中(SK-OV-3、OVCAR-3)的表达;利用RNA 干扰技术敲降卵巢癌细胞系中 PYCR1的表达,CCK-8和细胞平板克隆实验检测卵巢癌细胞的增殖能力,Transwell实验检测卵巢癌细胞的迁移、侵袭能力,Western blotting检测EMT以及Wnt/β-catenin通路相关基因蛋白表达水平。结果:PYCR1在卵巢癌组织中以及卵巢癌细胞中高表达。下调PYCR1的表达可明显降低卵巢癌细胞系的增殖、迁移、侵袭能力(P <0.05);EMT相关蛋白E-cadherin表达量升高,N-cadherin表达量降低。Wnt/β-catenin通路相关蛋白β-catenin和C-Myc表达量降低。结论:PYCR1可能通过Wnt/β-catenin通路促进卵巢癌细胞的增殖、迁移以及侵袭。

Expression and mechanism of PYCR1 in ovarian cancer cells
Abstract:Objective:To investigate the expression of PYCR1 in ovarian cancer cells, and its effects on proliferation, migration, invasion and EMTof ovarian cancer cells. Methods: GEPIA was used to analyze the expression level of PYCR1 in ovarian cancer. The expression of PYCR1 in normal ovarian cells (IOSE 80) and ovarian cancer cells (SK-OV-3 and OVCAR-3) was detected by Q-PCR, Western blotting and immunofluorescence techniques. RNA interference technique was used to knock down the expreExpression and mechanism of PYCR1 in ovarian cancer cellsssion of PYCR1 in ovarExpression and mechanism of PYCR1 in ovarian cancer cellsian cancer cell lines, CCK-8 and cell plate cloning assay were used to detect the proliferation of ovarian cancer cells, and Transwell assay was used to detect the migration and invasion of ovarian cancer cells. The expression levels of EMT and Wnt/β-catenin pathway related genes were detected by Western blotting. Results: PYCR1 was highly expressed in ovarian cancer tissues and ovarian cancer cells. Down-regulation of PYCR1 expression significantly reduced proliferation, migration and invasion of ovarian cancer cell lines (P < 0.05); The expression of EMT-related protein E-cadherin increased, while N-cadherin decreased. The expression levels of Wnt/β-catenin pathway related proteins β-catenin and C-Myc decreased. Conclusion: PYCR1 may promote the proliferation, migration and invasion of ovarian cancer cells through the Wnt/β-catenin pathway.
