中性粒细胞耗竭影响肠道微生物多样性:一项宏基因组学层面的研究

摘要:目的:探究中性粒细胞对肠道健康的影响。方法:通过构建小鼠中性粒细胞耗竭模型,采样小鼠肠内容物进行宏基因组测序分析。结果:小鼠中性粒细胞耗竭模型构建成功,在门水平上,中性粒细胞耗竭使梭杆菌门和迷踪菌门升高;在纲水平上,中性粒细胞耗竭使梭杆菌纲、迷踪菌纲、噬几丁质菌纲、甲烷微菌纲、甲烷球菌纲、亚硝酸球藻纲、异常球菌纲升高;在属水平上,中性粒细胞耗竭使梭杆菌属、迷踪菌属、乳球菌属、漫游菌属、黄杆菌属、甲烷八叠球菌属升高;在种水平上,中性粒细胞耗竭使艰难梭菌、化脓性链球菌、大肠杆菌和金黄色葡萄球菌升高。结论:中性粒细胞可以抑制梭杆菌门-梭杆菌纲-梭杆菌目-梭杆菌科-梭菌属-艰难梭菌、迷踪菌门-迷踪菌纲-迷踪菌目-迷踪菌科-迷踪菌属-迷踪菌、化脓性链球菌、大肠杆菌和金黄色葡萄球菌这些致病菌从而维持肠道稳态。

The Impact of Neutrophil Depletion on Gut Microbial Diversity: A Metagenomic Perspective
Abstract: Purpose: To explore the impact of neutrophils on gut health. Methods: The mouse neutrophil depletion model was established, and the intestinal contents of mice were sampled for metagenomic sequencing. Results: The mouse neutrophil depletion model was established successfully. At the phylum level, neutrophil depletion increased Fusobacteria and Elusimicrobia. At the class level, neutrophil depletion increased Fusobacteriia、Endomicrobia、Chitinophagia、Methanomicrobia、Methanococci、Nitrososphaeria and Deinococci. At the genus level, neutrophil depletion increased Fusobacterium、Endomicrobium、Lactococcus、Vagococcus、Tenacibaculum and Methanosarcina. At the species level, neutrophil depletion incerased Fusobacterium varium、Streptococcus pyogenes、Escherichia coli and Staphylococcus aureus. Conclusion: Neutrophils can inhibit Fusobacteria-Fusobacteriia-Fusobacteriales-Fusobacteriaceae-Fusobacterium-Fusobacterium varium、Elusimicrobia-Endomicrobia-Endomicrobiales-Endomicrobiaceae-Endomicrobium-Endomicrobium proavitum、Streptococcus pyogenes、Escherichia coli and Staphylococcus aureus to maintain intestinal homeostasis.
