琥珀酸对高脂饮食小鼠代谢和肠道菌群的调节作用

摘要:目的:通过不同浓度的琥珀酸钠干预,研究琥珀酸对高脂饮食小鼠代谢和肠道菌群的影响作用。方法:将32只6周龄C57BL/6J小鼠高脂饮食8周诱导小鼠肥胖模型后,随机分为4组,即高脂对照组(HFD)、低剂量琥珀酸钠干预组(L-SA)、中等剂量琥珀酸钠干预组(M-SA)、高剂量琥珀酸钠干预组(H-SA)。分别通过定期记录体重,代谢笼实验、检测血清血脂四项和炎症因子水平,组织学分析、16S rDNA测序等方法来研究琥珀酸对高脂饮食小鼠代谢和肠道菌群的影响。结果:琥珀酸可以减缓高脂饮食小鼠体重的增加,改善血脂和炎症水平、减少附睾白色脂肪细胞的扩张以及棕色脂肪白色化,并且调节了肠道菌群的结构和组成。结论:琥珀酸通过提高高脂饮食小鼠的呼吸代谢作用增加能量消耗,提高Akkermansia、Peptostreptotocococcaceae、Allobaculum的丰度,降低Lachnospiraceae、Ileibacterium、Desulfovibrionaceae丰度来改善高脂饮食小鼠的代谢紊乱和体重增加。

Regulatory effects of succinic acid on metabolism and intestinal flora in mice fed a high-fat diet.
Abstract:Objective: Intervene high-fat diet mice with different concentrations of sodium succinate to study the effects on metabolism and intestinal flora.Methods: Thirty-two 6-week-old C57BL/6J mice were randomly divided into four groups: a high-fat control group (HFD), a low-dose sodiumsuccinic intervention group (L-SA), a medium-dose sodiumsuccinic intervention group (M-SA), and a high-dose sodiumsuccinic intervention group (H-SA).The effects of succinic acid on the metabolism and intestinal flora of high-fat diet mice were studied by regular body weight recording, metabolic cage test, detection of serum lipid four items and inflammatory factor levels, histological analysis, 16S rDNA sequencing, and other methods.Results: Succinic acid could slow the weight gain, improve blood lipid and inflammation levels, reduce the expansion of epididymal adipose white tissue and the whitening of brown adipose tissue, and regulate the structure and composition of intestinal flora in HFD-fed mice.Conclusions: Succinic acid by raising a high-fat diet in mice respiratory metabolism increased energy consumption, improve Akkermansia, Peptostreptotocococcaceae, Allobaculum abundance, Reducing the abundance of Lachnospiraceae, Ileibacterium, and Desulfovibrionaceae can improve metabolic disorders and body weight gain in high-fat diet mice.
