miRNAs are a large family of small regulatory elements that direct messenger RNA degradation or disrupt mRNA translation. High-throughput Solexa sequencing approach was adopted to expand the repertoire of porcine miRNAs and to explore their roles on spermatogenesis. 237,590 total reads (792 unique reads) representing 65 known mature miRNAs in the miRBase release 14.0 were detected. 116 miRNAs including 12 known miRNAs in the miRBase release 15.0 and 104 novel miRNAs were identified from the un-annotated small RNAs. 466,757 targets for 104 novel miRNAs were predicted by MIREAP. The highly expressed miRNAs in porcine testes (let-7c, let-7f, miR-148a, miR-140* and miR-199b) had a very high rate of SNP mutation, and the most popular SNP was T to G mutation at position 6. Three known miRNAs including miR-514, miR-507, miR-508-5p were significantly differentially expressed between some groups (p<0.05), while two novel miRNAs- DB-m0102 and DB-m0071 were not significantly differentially expressed between all detected groups.

Oxidative stress is thought to be a major cause of light induced retinal neurodegeneration. The protective role of 17β-estradiol (βE2) in neurodegenerative disorders is well known, however, the underlying mechanism remains unclear. Here, we applied light induced retinal damage model to explore the mechanism by which βE2 exerts its neuroprotection. Adult male and female- ovariectomized (OVX) rats were exposed to 8000 lx white light for 12h to induce retinal light damage. Electroretinogram (ERG) assay and hematoxylin and eosin (H&E) staining show that exposure to light for 12h resulted in functional damage to the rat retina, histological changes and retinal neurons loss, while intravitreal injection (IVI) with βE2 significantly rescued impaired retina function in both female and male rats. By detecting malonylodialdehyde (MDA) production (a biomarker for oxidative stress) indicated an increasing retinal oxidative stress following exposure to light, and βE2 reduced the light induced oxidative stress. qRT-PCR indicated that antioxidant enzymes SOD and Gpx mRNA level were diminished in female-OVX rats but up-regulated in male rats after exposing to light, suggesting a gender differences in regulating the genes of these antioxidant enzymes in response to light. However, administration of βE2 recovered or enhanced the SOD and Gpx expression upon light stimulation. In spite of the CAT expression was not sensitive to light, βE2 also increased the gene expressions both in female-OVX and male rats. Further study indicated the antioxidant proteins Trx and Nrf2 were also involved in βE2 mediated anti-oxidation, while cytoprotective HO-1 exerts a key role in endogenous defense mechanisms against light but not via βE2. Taken together, we provide evidence that βE2 protected the retinal from light damage via anti-oxidative effect, the underline mechanism involved in regulation of the genes of antioxidant enzymes (SOD, CAT, Gpx) and proteins (Trx and Nrf2). Our study will provide the theory evidence for estrogen replacement therapy in postmenopausal women for reducing the risk of Age-related macular degeneration.

Major histocompatibility complex (MHC) class II B molecules play an important role in the adaptive immune response in fish. Previous study has reported that two highly polymorphic class II B genes, Cyse-DAB and Cyse-DBB exist in half-smooth tongue sole (Cynoglossus semilaevis). In this study, the polymorphism within exon 2 of the class II B genes following bacterial challenge was evaluated. Two hundred individuals were injected intraperitoneally with Vibrio anguillarum. Muscle tissue from the first 20 dead and 20 of the survivors was collected for genotyping. Sixty alleles from the 40 individuals were isolated, of which 32 belonged to Cyse-DAB and 28 belonged to Cyse-DBB. The rate of dN (non-synonymous substitution) was higher than that of dS (synonymous substitution) in the PBRs (peptide binding residues) of both class II B genes. Conversely, the rate of dS was higher than dN in the non-PBRs and the complete exon 2 sequence. Thus, the results suggest that positive selection has occurred in the PBRs and purifying selection in the non-PBRs and exon 2. Thirteen class II B alleles were used to study the association between alleles and resistance to infection. Though not significant, alleles Cyse-DAB*0601, Cyse-DAB*0706, and Cyse-DBB*0101, Cyse-DBB*1301 were only found in surviving individuals and may represent alleles that have resistance against V. anguillarum infection. Alleles Cyse-DAB*0701 and Cyse-DAB*1301 were significantly more prevalent in dead individuals than in surviving ones and may represent alleles that are associated with increased susceptibility to V. anguillarum infection.

Major histocompatibility complex class II antigens are important in vertebrate immune system. In the present study, the full length cDNA sequence of class II A gene was got by RACE-PCR from half-smooth tongue sole (Cynoglossus semilaevis), and its open reading frame (ORF) polymorphism was studied. The whole cDNA sequence was 992 bp in length, including the ORF of 717 bp. Twenty-five alleles were identified and clustered into two distinct groups for the specific nucleotides/ amino acids in specific positions, so they belonged to two class II A genes and dominated as Cyse-DAA and Cyse-DBA with 11 and 14 alleles respectively. Four Cyse-DAA alleles were observed in one individual, and three to five Cyse-DBA alleles in each of three detected individuals, which indicated that at least two loci existed in each gene. Moreover, in order to study the association of the alleles of A genes and resistance to infection, 200 individuals were intraperitoneally injected with Vibrio anguillarum and the first 20 dead individuals and 20 surviving ones were selected for genotypes of A genes. Fifty-six alleles were identified among the 40 individuals, 29 of which belonged to Cyse-DAA and 27 belonged to Cyse-DBA. Eighteen alleles were selected for the association study between alleles and resistance to infection. Though not significant, allele Cyse-DAA*0201, Cyse-DAA*1101, Cyse-DBA*0401, Cyse-DBA*1102, Cyse-DBA*1801 and Cyse-DBA*2201 appeared only in surviving individuals, while allele Cyse-DAA*0901, Cyse-DBA*1101 and Cyse-DBA*1401 occurred more frequently in dead individuals. This study confirmed the existence and polymorphism of two class II A genes and also the association between alleles of class II A genes and disease susceptibility/ resistance in half-smooth tongue sole.

Selection of suitable reference gene is important for accurate measurement of gene expression data obtained using the quantitative real-time PCR (qPCR) method. There has been no systematic investigation of reference genes for porcine chorioallantois which plays important roles in mediating concepts/fetus growth and survival. In this study, six candidate reference genes expressed in pig chorioallantois from Meishan and Yorkshire pigs on gestational days 26 (GD26) and 50 (GD50) were evaluated by qPCR. These genes included TATA box binding protein (TBP), glyceraldehydes-3-phosphate dehydrogenase (GAPDH), ribosomal protein L32 (RPL32), actin-beta (ACTB), ribosomal protein S20 (RPS20) and RNApolymerase polypeptide G (RPG). The expression stability of these genes was investigated by using the GeNorm software program and statistical analyses. The rank order of the expression stability for these genes was (from the most stable to the least stable): TBP, GAPDH, RPL32, ΒACTIN, RPS20 and RPG. Collectively, the results suggest that the TBP and GAPDH could be considered as the suitable reference genes for investigation of gene expression in the pig chorioallantois from the two breeds at different gestational stages.

This study tested the hypothesis that abscisic acid (ABA) and ethylene may mediate the effect of postanthesis soil-drying on grain filling of rice (Oryza sativa L.) through regulating antioxidative systems. Two rice cultivars were pot-grown, and three soil-drying treatments, well-watered (WW), moderately soil-drying (MD) and severely soil-drying (SD), were imposed from 9 days post anthesis until maturity. Levels of ABA and ethylene, activities of catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD), and contents of superoxide anion (O2-) and malondialdehyde (MDA) and their relationships with grain filling rate were investigated. The results showed that, when compared with the WW treatment, the MD treatment increased, whereas the SD treatment decreased, grain filling rate and activities of CAT, POD, and SOD in both superior and inferior spikelets. Both MD and SD treatments increased contents of ABA, superoxide anion and MDA in grains, with more increases under the SD than under the MD. The MD treatment decreased, whereas the SD treatment increased, ethylene evolution rate and 1-aminocycopropane-1-carboxylic acid (ACC) content. The ratio of ABA to ACC (ABA/ACC) was the highest under the MD and the lowest under the SD among the three soil-drying treatments. ABA content, activities of CAT, POD, SOD, and ABA/ACC very significantly and positively, while the levels of ethylene and ACC and contents of superoxide anion and MDA significantly and negatively, correlated with grain filling rate. Application of cobalt ion (inhibitor of ethylene synthesis) or ABA at early grain filling stage significantly decreased contents of superoxide anion and MDA, and increased activities of CAT, POD, SOD and grain filling rate. Spraying with ethephon (ethylene-releasing agent) or fluridone (inhibitor of ABA synthesis) had the opposite effect. The results suggest that an increase in ABA and decrease in ethylene in the grains under postanthesis moderate soil-drying could enhance the activities of antioxidative enzymes and reduce free radicals in grains, and therefore, accelerate the grain filling rate of rice.

G4DNA, which widely exists in the structure of the telomeres in normal cells, plays a pivotal part in the process of prolonging the telomere DNA by catalyzing the enzyme telomerase. Bloom (BLM) helicase, an important member of the RecQ DNA helicase family, plays an important role in DNA metabolism, including DNA replication, repair, transcription, and recombination. The unwinding of G4DNA requires DNA helicase participation, which is crucial for maintaining chromosomal stability. This study was conducted to determine the DNA-binding and unwinding properties of the BLM helicase catalytic core to G4DNA using fluorescence polarization and the electrophoretic mobility shift assay (EMSA). The results revealed that the BLM helicase catalytic core could bind and unwind G4DNA. The molecular affinity of G4DNA binding by the helicase was dependent on the single-stranded DNA (ssDNA) terminals in the G4DNA; the helicase binds to the G4DNA where one helicase monomer covers approximately 10 nucleotides at the 3' or 5' ssDNA tail. The unwinding of G4DNA was dependent on the presence of a 3' ssDNA tail and ATP; the G4DNA with only a 3' ssDNA tail was the most favorable substrate to be unwound by the BLM helicase catalytic core, and required 3' ssDNA tails of at least 10 nt in length for efficient unwinding. The blunt-ended G4DNA was loosely bound and partly unwound by the helicase catalytic core. The experimental results presented are beneficial to further the understanding the functionality of BLM helicase in cells.

IgA nephropathy (IgAN) is one of the most common primary glomerulonephritis. Previously-identified GWAS loci explain only a fraction of disease risk. To identify novel susceptibility loci in Han Chinese, we conduct a four-stage GWAS comprising 8,313 cases and 19,680 controls. Here, we show novel associations at ST6GAL1 on 3q27.3 (rs7634389, OR=1.13, P=7.27x10-10), ACCSon 11p11.2 (rs2074038, OR=1.14, P=3.93x10-9) and ODF1-KLF10on 8q22.3 (rs2033562,OR=1.13, P=1.41x10-9), validate a recently reported association at ITGAX-ITGAM on 16p11.2 (rs7190997, OR=1.22, P= 2.26x10-19),and identify three independent signals within the DEFA locus(rs2738058, P=1.15x10-19; rs12716641, P=9.53x10-9; rs9314614, P=4.25x10-9, multivariate association).The risk variants on 3q27.3 and 11p11.2 show strong association with mRNA expression levels in blood cells while allele frequencies of the risk variants within ST6GAL1, ACCS and DEFA correlate with geographical variation in IgAN prevalence. Our findings expand our understanding on IgAN genetic susceptibility and provide novel biological insights into molecular mechanisms underlying IgAN.

景观遗传学是目前正在兴起的一门涉及生态学、遗传学、环境学和进化理论等多个学科交叉的热点研究方向。由于涉及到从大尺度的景观生态学和种群生态学到小尺度的分子遗传学多层面研究,相关工作在试验材料的选择方面对研究进展影响较大。试验所选材料一般应具备地理分布广、种群分化高、逆境适应强和取材方便等特点。综述前期研究结果发现,沙棘属植物具有以上重要特点,是景观遗传研究中的理想材料。本文对沙棘属植物的形态学特征、生态学特征、遗传学特征及分子生物学特征进行总结,结合当前景观遗传学中的研究进展前沿,阐述沙棘属植物在景观遗传学中的科研价值和实践意义,为今后景观遗传学的深入研究和生态恢复与治理实践提供基础研究材料。

Objective: TBX5 gene is a member of T-box transcriptional factors that mainly expressed in cardiovascular system and play a significant role in the development of embryonic cardiovascular. The aim of this study is to investigate the association of TBX5 gene polymorphisms with the risk of congenital heart disease (CHD) in the Tibetan population in China. Methods: One hundred and twenty Chinese Tibetan patients with CHD as well as one hundred and twelve matched healthy controls were recruited for this study. And the tagging SNPs were genotyped using Illumina GoldenGate chips. Potential association was evaluated with the chi-square (Χ2) test. Results: The selected tagging SNPs were well genotyped and we found significant differences in allele frequencies and genotype distributions of rs55646156 as well as rs4533090 between CHD patients and controls. The AA genotype of rs55656156 may be am increased risk factor of CHD, while the TT genotype of rs4533090 might be the protective factor against CHD development in Chinese Tibetan population. Conclusions: We have firstly established the association between the tagging SNPs of TBX5 and CHD in the Tibetan population of China in the present study.
