In the past several years, a lot of symmetric mono-carbonyl analogues of curcumin (MACs) have been designed and synthesized in our laboratory. In the recent study, Eleven asymmetric MACs were designed and synthesized. Their bioactivities were evaluated by inhibit LPS-stimulated macrophages release proinflammation cytokines TNF-α and IL-6. The compound 1d exhibit the most inhibit activity among the test compounds. These data show that asymmetric MACs might serve as potential agents for the treatment of inflammatory diseases.

The cell cycle inhibitor flavopiridol was previously shown in animal models to improve SCI recovery. However, the systematic dosage of flavopiridol is with side effects and the mechanism is not clear. This study aims to develop a strategy for local delivery of flavopiridol and investigate its mechanisms of function as well. Poly(lactic-co-glycolic acid) (PLGA) nanoparticles (NPs) was used for the sustained delivery of flavopiridol. The spinal cords were right-hemisectioned and NPs were administered into the injury site. Transparent spinal cord technology was applied for the three-dimensional observation of the anterograde tracing. The results showed that flavopiridol NPs had a susteined release of up to 3 days in vitro. Flavopiridol NPs significantly decreased inflammatory factor synthesis of astrocytes, including tnf-alpha, il-1beta, and il-6, while the il-10 expression was elevated. In-vivo study demonstrated that flavopiridol NPs decreased inflammatory expression and glial scarring, and facilitated neuronal survival and neural transmitting. Cavitation volume was decreased by proximately 90%. Administration of flavopiridol NPs also improved the motor recovery of injured animals. These findings illustrated that local delivery of flavopiridol in PLGA NPs improved SCI recovery through inhibiting astrocyte growth and inflammatory factor synthesis.

炎症是机体遭受微生物入侵、组织损伤时必不可少的宿主防御反应。健康状态下,炎症是自限性的,可以自然消退,这说明可能存在内源性的机制来控制炎症的消退。导致炎症发生的促炎机制已逐渐被阐明,而炎症的消退一直被认为是被动的过程,最近的研究显示,炎症消退是一个由内源性抗炎、促炎症消退介质控制的主动的程序化过程,脂氧素就是其中一种非常重要的内源性抗炎促消退介质,被誉为炎症的"刹车信号"。本文就脂氧素在炎症消退中的作用及其机制简要进行综述。

背景及目的:牙髓炎症是影响牙髓修复的关键,轻微的炎症或者及时去除刺激,有利于牙髓的愈合;持续的炎症,使牙髓修复能力减弱甚至引起牙髓坏死。YAP是一种与细胞的矿化过程有关的转录共激活因子,不仅参与调节炎症过程还与多种细胞的矿化过程有关。但是,YAP是否参与牙髓炎症及炎症损伤后的修复过程尚不清楚。因此,本课题探究了YAP是否参与牙髓炎症损失及修复过程。方法:建立大鼠牙髓炎模型;收取人的健康牙髓和牙髓炎组织,免疫组织化学染色检测牙髓组织中YAP的表达。10 ng/mL TNF-α体外诱导牙髓细胞的炎症反应。qPCR检测mRNA的表达,Western blot检测蛋白的表达,茜素红S染色检测牙髓细胞的矿化能力。结果:牙髓炎组织中YAP的表达明显上调,TNF-α也上调牙髓细胞中YAP 的表达。诱导牙髓细胞矿化前,TNF-α预处理0.5小时促进了矿化,TNF-α预处理3小时和TNF-α持续刺激抑制了矿化。与对照组相比,矿化诱导液组中牙髓细胞的YAP蛋白表达上调。与矿化诱导液组相比,TNF-α预处理0.5小时抑制了YAP的表达;然而,TNF-α预处理3小时和TNF-α持续刺激上调了牙髓细胞YAP蛋白的表达。结论:YAP参与了牙髓炎症过程。短时间的炎症刺激促进了牙髓细胞的矿化,长时间炎症刺激抑制矿化。YAP参与了正常条件下牙髓细胞的矿化过程和受到炎症刺激的牙髓细胞的矿化过程。

目的:研究中性粒细胞在小鼠急性炎症中的性别差异。方法:小鼠腹腔注射酵母多糖,12 h后流式细胞术检测雌雄小鼠腹膜洗液、肠系膜淋巴结、骨髓中中性粒细胞的比例。结果:雌性小鼠腹膜洗液中中性粒细胞的比例明显比雄性小鼠中的高,分别为72.80% ± 8.649%、41.07% ± 8.649%;雌性小鼠肠系膜淋巴结中性粒细胞比例高于雄性小鼠,分别为0.526% ± 0.295%、0.047% ± 0.295%;然而雄性小鼠骨髓中的中性粒细胞比例比雌性小鼠高分别为20.73% ± 3.121%、10.25% ± 3.121%。结论:中性粒细胞在小鼠急性炎症模型中具有性别差异性,炎症高峰期时雌性炎症明显高于雄性。

鼻息肉是一种常见病,其发病细菌、病毒、炎症、组织重塑与鼻息肉率及复发率均较高。其发病机制至今尚没有明确,普遍认为是多种因素共同作用的结果,例如感染性因素、细菌超抗原、细胞因子释放等。本文就目前该疾病病因学的研究现状进行综述。着重从细菌、病毒、炎症、组织重塑4个方面对鼻息肉的发病机制的研究现状进行小结。

Toll样受体(Toll like receptors,TLRs)是与果蝇Toll蛋白具有同源性的表达于细胞膜上的一类受体家族。TLRs可以识别相应配体后引起信号转导,激活相关基因的表达。近来研究发现在动脉粥样硬化中存在多种炎症因子,这些炎症因子的表达很大程度上依赖于TLRs信号通路的激活。总结TLRs信号通路在动脉粥样硬化炎症反应中的作用,将有利于更清楚地了解动脉粥样硬化的分子机制及其与炎症的关系。

肥胖及其相关代谢异常疾病目前成为严重的公共卫生问题。慢性炎症反应与肥胖、胰岛素抵抗等疾病密切相关,这种炎症反应可由脂肪组织分泌的因子诱发,此外肠道菌群相关的内毒素水平异常增高也引起慢性炎症反应,动物试验中有较多相关报道,却缺乏足够的相关人体试验。本文介绍了LPS在慢性炎症反应中作用的研究进展,为肥胖的治疗方向提供思路。
