THE EFFECT OF CHEMICAL TREATMENT OF SILICON GEL ON TISSUE COMPATIBILITY
Objective To study if the epithelial proliferation on the surface of the silicon gel is affected by chemical agents. Methods Silicon gel was treated with two chemical agents. One is treated with mixed acid solution containing 23.2% H2SO4 and 0.8% K2Cr2O7, the other is treated with 30% H2O2 solution. The treated time is 5, 10, 15minutes in mixed acid solution and 10, 15, 20minutes in H2O2 solution. After treatment the cultural corneal epithelial cells were seeded onto the silicon gel. Immunohistochemical study was carried out for integrin subunits alpha6, beta4 and actin. Results The growth of the epithelial cells was much better in acid treated silicon gel for 10 and 15 minutes than the control. After cultured for 12 hours, we found a small amount of cell proliferate on the surface of the silicon gel treated with 30% H2O2 for 15 minutes. After 3 days culture, the cell proliferate and fuse together. The cells were not growing well on the surface treated with H2O2 for 10 and 20 minutes. Immunostainning revealed that the expression of actin and integrin subunits alpha6, beta4 were found on the silicon gel treated with acid for 10 minutes and H2O2 for 15 minutes. Conclusion The silicon gel treated with H2O2 for 15 minutes and 10, 15 minutes with mixed acid solution were better treatment to improve the cell proliferation.
