严洁,女,1941年生,现为湖南中医药大学针灸推拿学院针灸学教授、博士生导师,湖南省首批名中医,湖南省第二届优秀中青年专家,中国针灸学会常务理事、经络分会副会长,湖南省针灸学会会长,享受国务院特殊津贴。
从事教学、科研、临床40余年,指导培养海内外博士生26名,硕士生21名。长期以来,带领一支团结齐心,无私奉献的科研团队,以“经脉脏腑相关的临床与实验研究”作为主攻方向。开展了30余项课题研究。其中主持了国家自然科学基金课题3项,国家教育部博士学科点专项科研基金课题1项,国家中医管理局课题1项。曾担任国家“七•五”攻关,“八•五”攀登,“九•五”攀登预选项目“经络的研究”课题中“经脉脏腑相关”专题组长,子课题负责人,及“九•五”攀登预选项目“经络研究”专家委员会委员。除外尚参与了国家“973”课题2项,国家自然科学基金课题与省部级课题多项,先后获省部级科技成果奖13个(其中二等奖6项,三等奖8项,9项排名第一),省级教学成果奖1项(排名第二),出版个人专著《图解中国针灸技法》1部,副主编普通高等中医院校规划教材《针灸学》及配套著作共4部,协编教材1部(《实验针灸学》),整理出版针灸医籍4部,参编著作6部,发表论文60余篇(其中SCI源刊与收录共7篇)。1996年至2005年分别赴美国、意大利、澳大利亚参加世界针联主办的国际针灸学术交流会,1997年赴马来西亚讲学。在“足阳明经与胃相关”的研究中,发现了针刺四白穴可使胃窦容积明显增大,胃运动功能明显增强,胃粘膜损伤指数明显降低,其效应不亚于针刺足三里,为临床治疗胃病提供了有效穴,亦为经络学说“上下对应”的标本理论提供了实验依据。并进一步从针刺→脑肠肽→细胞膜受体→G蛋白偶联→胞内第二信使→生物学效应等多水平阐明经穴与脏腑联系的通路及物质基础。深化了针刺对脏腑调控过程及机制的研究,丰富了传统针灸对经脉--脏腑相关理论的认识。

Acupuncture has been widely applied to treat gastrointestinal diseases in the clinic. However, its precise mechanism remains unknown. In the present study, effects of electroacupuncture (EA) at Zusanli acupoints (ST36) on gastric myoelectric activity and substance P (SP) in the dorsal vagal complex (DVC) were observed. EA at ST36 significantly increased the frequency of slow waves and the frequency of the spike potentials superimposed on the slow waves, which persisted for 90 min after cessation of EA and was completely abolished after bilateral vagotomy. Meanwhile, the mean optical density of SP immunoreactive fibers in the DVC increased following EA. The results suggested that EA at ST36 promoted the gastric myoelectric activity, which was regulated by the vagus, and SP in the DVC may be involved in the excitatory effects. Furthermore, EA had a relative long-term post-effect. We presumed that DVC may be a supraspinal center in the effect of EA on gastric function. q 2003 Elsevier Ireland Ltd. All rights reserved.

Clinical practice has shown that acupuncture at facial acupoints has curative effects onsome visceral diseases (especially gastrointestinal diseases). However, the physiological basis has not been clarified yet. In the present study, expression of c-fos in the nucleus of the solitary tract (NTS) of rats following gastric distension and electroacupuncture (EA) at Yangbai (GB14) and Sibai (ST2) as well as Jiache (ST6) acupoints was observed by using immunohistochemistry technique. After EA at the three facial acupoints, c-fos immunoreactive (c-fos-IR) neurons were mainly distributed in the medial (mNTS) and intermediate subnucleus of the NTS, and a few were scatteredly distributed in the dorsalmedial and commissural subnucleus of the NTS. Furthermore, there is difference in the number of c-fos-IR neurons in the mNTS following EA at the three facial acupoints. The number in the EA at ST2 and GB14 group is the highest and the lowest, respectively. Gastric distension induces obviously the expression of c-fos, which is mainly confined in the mNTS. The results suggest that the noxious visceral and somatic afferent information from the stomach and face may converge in the mNTS, which may be involved in the effect of EA at facial acupoints on the gastrointestinal pain.

AIM: To investigate electroacupunture(EA) at the acupoints of Stomach Meridian of Foot-Yangming(SMFY), Gallbladder Meridian of Foot-Yangming(SMFY) on gastric mucosal intestinal trefoil factor (ITF) gene expression detection in stress-induced rats with gastric mucosal lesion, and to explore the regulatory mechanism and signify cance of EA-related gastric mucosal protective effect. METHODS: Forty rats were randomly divided into 4 groups: Blank group, Model group, Model group+EA at acupoints of SMFY group(“SMFY group”), and Model group+EA at acupoints of GMFY group(GMFY group). All rats (except blank group) were made model by water immersion and restraint stress (WRS). Then the gastric mucosa tissue in each rat was taken off after assessment of gastric mucosal lesion index(GUI), and the expression of ITF mRNA of the tissues was detected by reverse transcription-polymerase chain reaction(RT-PCR) method. RESULTS: Compared with Model group(54.3 ± 1.34), the GUI value in SMFY group (31 ± 2.21) decreased significantly(P < 0.01), so did that in GMFY group (39.8 ± 1.62, P < 0.05), meanwhile GUI value in SMFY group was significantly lower than in GMFY group(P < 0.01). Compared with Model group (0.65 ± 0.01), EA had a tendency to improve the expression of gastric mucosal ITFmRNA gene: such tendency existed in GMFY group (0.66 ± 0.01) but with no signficant difference(P > 0.05), in SMFY group(0.76 ± 0.01) with an extremely obvious difference (P < 0.01), furthermore the expression in SMFY group was significantly higher than in GMFY group (P < 0.01). CONCLUSION: The gastric mucosal protective effect by EA at the acupoints of SMFY and GMFY was related to the expression variance of ITF, indicating certain meridian specifi city exists. It could be one proof for the TCM theory “Relative particularity between SMFY and stomach”.

Objective:To study the effect of medicinal herb-cake-separated moxibustion on serum lipoprotein levels in hyperlipemia rabbits. Methods: A total of 55 New-Zealand rabbits were randomly divided into control group (n=13), model group (n=14) and medicinal herb-cake-separated moxibustion (indirect moxibustion) group (n=14). Hyperlipemia model was established by feeding the animals with specialized forage (15% vitellus powder, 5% cholesterol and common forage )for 6 weeks. Moxibustion was applied to “Juque”(CV14), “Tianshu”(ST40), etc, 4 moxa-cones for every acupoint, once daily and continuously for 40 days. Serum triglyceride (TG) and total cholesterol (TCH) contents were assayed with colorimentric method. Results:Compared with control group, TCh and TG levels of model group increased significantly (p<0.01). TCh and TG contents of direct moxibustion and indirect moxibustion groups were significantly lower than those of model group (p<0.01).Comparison between two moxibustion groups showed that serum TCh level of indirect moxibustion group was strikingly lower than that of direct moxibustion group (p<0.01). It indicated that both direct and indirect moxibustion could effectively lower hyperlipemia and the therapeutic effect of indirect moxibustion was significantly superior to that of direct moxibustion in lowering serum TCh level. Conclusion: Both direct and indirect moxibustion canregulate lipid metabolism and the therapeutic effect of medicinal herb-cake-separated moxibustion is superior to that of direction moxibustion in hyperlipemia rabbits.

expression in gastric mucosal cells. Such tendency existed in the model group (0.7104 ± 0.0457) but with no signifi cant difference (P = 0.495 > 0.05) and in model serum group (0.8516 ± 0.0409) with an extremely obvious difference (P = 0.001 < 0.01). Furthermore, the EGFR gene expression in stomach serum group was significantly higher than that in gallbladder serum group (P = 0.00<0.01). CONCLUSION: The present study shows that serum derived from rats treated with electroacupuncture atstomach meridian acupoints can distinctly increase the EGFR gene expression of gastric mucosal cells. Therefore, there is certain meridian specificity in the serum, which could provide a proof for the TCM theory “particular relation between meridian and internal organ”.

【期刊论文】电针对应激性胃黏膜损伤大鼠胃黏膜肠三叶因子基因表达的影响

目的:探讨电针预处理对胃黏膜损伤大鼠胃黏膜组织肠三叶因子(intestinal trefoil factor, ITE)基因表达的影响及意义。方法:将40只大鼠随机分为空白组、模型组、胃经组、胆经组。空白组不做任何处理,而模型组仅捆缚7d后造模,胃经组、胆经组须先捆缚电针7d后再造模。造模采用水浸束缚法,水浸束缚10 h,经相应处理后检测胃黏膜损伤指数(UI),同时取各组大鼠胃黏膜组织,采用RT-PCR方法测胃黏膜组织肠三叶因子基因(ITFmRNA)表达。结果:①胃经组、胆经组与模型组的UI比较差异有显著或非常显著性意义(p<0.05或p<0.01),说明造模成功;②电针可提高胃黏膜组织ITFmRNA表达水平,胆经组较模型组ITFmRNA表达增高,胃经组较胆经组ITFmRNA表达非常显著升高(p<0.01)。结论:电针对胃黏膜组织特异性调整作用,与肠三叶因子基因表达差异有关。

【期刊论文】电针胃经穴后胃溃疡大鼠胃黏膜细胞蛋白激酶C活性的变化

目的:观察电针胃经(穴)对应激性胃溃疡大鼠胃黏膜细胞蛋白激酶C活性的影响。方法:实验于2005-07/12在湖南中医药大学针灸基础实验室完成。选择健康SD大鼠60只,随机数字表法分为5组,胃溃血清组(12只)、胃经血清组(12只,取穴;四白、梁门、足三里)和胆经血清组(12只,取穴:阳白、日月、阳陵泉)、胃经血清+PD153035组(12只)和胆经血清+PD153035组(12只)。采用水浸束缚法制作胃溃疡动物模型。电针刺激用G6805型电针仪,疏密波,电针频率疏波4Hz,密波20Hz,强度以肌肉或针柄微颤为度,电针时间30min。利用链酶蛋白酶消化法分离胃黏膜细胞,分别用表皮生长因子受体抑制剂PD153035和血清孵育胃黏膜细胞,应用同位素掺人法检测蛋白激酶C活性。结果:纳入动物60只,均进入结果分析。溃疡血清大鼠胃黏膜细蛋白激酶C有少量表达[(29.16±30.94)nkat/g];胃经血清组和胆经血清组大鼠胃黏膜细胞蛋白激酶C呈现较强上增性表达,其中胃经血清组大鼠上增性表达特别明显,两组相比差异有显著性意义[分别为(114.92±65.61),(36.47±27.54)nkat/g,P<0.01];胃经血清+PD153035组和胆经血清+PD153035组大鼠胃黏膜细胞蛋白激酶C可见微弱表达[分别为(17.39±23.39),(23.44±14.42)nkat/g],胃经血清组胃经血清+PD153035组比较差异有显著性意义(p<0.01)。结论:针刺能刺激大鼠胃黏膜细胞信号转导通路中蛋白激酶C的活化,并且存在经脉-脏腑的特异性联系。

目的比较电针足阳明、足少阳经穴对实验性胃溃疡大鼠胃黏膜损伤修复作用的异同,探讨足阳明经与胃相关的特异性及电针足阳明经穴对胃黏膜损伤细胞修复作用的可能机制。方法将40只大鼠随机分为空白组、模型组、胃经组、胆经组。造模采用水浸束缚法。实验结束后取血清及胃黏膜组织,采用放射免疫分析法测定表皮生长因子(EGF)、转化生长因子α(TGF-α)含量,采用逆转录-聚合酶链反应法(RT-PCR法)分析胃黏膜EGFRmRNA表达。结果电针足阳明经穴均能使胃黏膜损伤指数显著降低,能显著升高胃黏膜EGF、TGF-α含量;电针上调EGFRmRNA在胃黏膜的表达水平,其中以足阳明经穴组作用最强,与足少阳经组比较差异显著。结论电针足阳明经穴对大鼠胃黏膜损伤的细胞修复作用是通过促进EGF、TGF-α等相关因子的合成、分泌与释放实现的;增强EGFRmRNA在胃黏膜局部的表达,可能是电针足阳明经穴对大鼠胃黏膜损伤修复作用的另一机制;足阳明经与胃具有相对的特异性。
