现任南京农业大学食品科技学院院长,中国农学会农产品贮藏加工分会常务理事、中国微生物学会理事、江苏省微生物学会理事、江苏省生物技术学会理事、农业部重点开放实验室副主任、国务院“政府特殊津贴”获得者、江苏省政协委员、“九三”学社江苏省委委员、“九三”学社南农分社主委、江苏省“333”工程第二层次培养对象。1995.3 毕业于日本近畿大学,获博士学位,在攻读博士学位期间,利用辐射合成高分子,进行固定化细胞机理研究和固定化细胞对纤维素有效利用的基础研究,以及纤维素酶水解的反应动力学和反应机理的研究,阐明和发展了微生物在高分子表面的吸附机理和纤维素酶水解的反应机理,开发了固定化木霉和固定化酵母的新方法;1995.3~1996.6在日本RADIA工业株式会社做博士后主要从事医疗用品辐射灭菌保证水平的基础研究;2001~2002.3作为高级访问学者在英国诺丁汉大学从事酶基因克隆和表达研究。目前主要从事酶工程、食品微生物学等领域的教学及科研工作,已招收4届博士研究生,8届硕士研究生,现指导硕士研究生十几名,指导博士研究生近十名。讲授硕士研究生课:酶工程技术、研究班讨论;讲授博士课程:高级食品生物化学、食品生物技术进展等。主编21世纪教材《现代食品生物技术》和正在主编国家“十五规划教材”《食品质量管理学》教材2部。目前主持国家“十五”大科技项目、教育部、农业部和江苏省自然科学基金重点项目等研究项目十余项。1998年“辐射合成高分子材料上微生物细胞的吸附研究”获教育部科技进步二等奖;1999年“辐射与生物技术相结合提高纤维素水解效率的基础研究”获农业部科技进步二等奖。发表论文90多篇,其中SCI论文20篇,EI论文5篇,SCI引用40多篇次。

In the present investigation, response surface methodology based on a five-level, six-factor central composite rotatable design of experiments was employed to optimize the levels of six factors significantly affecting the yield of extracellular polysaccharide produced by submerged cultivation using Pholiota squarrosa (Pers. ex Fr.) Quel. AS 5.245. The selected factors, i.e., yeast extract, fructose, magnesium sulfate, maltose, zinc chloride and initial pH value were based on the results of our previous Plackett-Burman design. By solving the quadratic regression equation using appropriate statistic methods, the optimum concentration values for obtaining 876.32g extracellular polysaccharide per milliliter of cultivation medium were determined: yeast extract 6.0gl−1, fructose 11.5gl−1, magnesium sulfate 0.5gl−1, maltose 9.6gl−1, zinc chloride 38.6mgl−1 and initial pH value 5.3. The validation data under various experimental conditions have also confirmed the correlation between the experimental and theoretical values.

The recognition of microbial cells by the surface of polymers with oxyethylene units was investigated using the organism Trichoderma reesei, in which the surface was modified by the radiation polymerization method. The cells were recognized by their adhesion to the surface of the polymer giving a bioaffinity similar to cellfusion. The adhesion was affected by the molecular structure of the surface relating to the length of the oxyethylene unit, and enzyme productivity from the adhered cell walls was affected by the property of the polymer.

Gibberella fujikuroi cells were immobilized on the surface of fibrous carriesrs with copolymers obtained by a radiation polymerizsation techique. The gibberellc acid(GA) production in the immmobilized cells was investigated by varying the immobilization conditions and using various monmoers. GA productivity in cells immobilized with the carrier-covered copolymer of hydrophylic hydrozyethyl acrylate andhydrophobic trimethylpropane triacrylate was higher than that in the free cells. the stability of the immobilized cells was evaluated by repetition of the batch fermentaiton. The immoblized cells produced GA with high stability GAproductivity by immobilized cellsvaried with the chemical nature of the carriesrs covered by the polymers.

Fresh-cut celery was dipped with ozonated water and evaluated for changes of microbiological population and physiological quality during storage at 4 C. The polyphenoloxidase (PPO) activity and respiration rate of fresh-cut celery was much inhibited by treatment of ozonated water and sensory quality of fresh-cut celery treated with ozonated water was better (α=0.05) than that of non-treated. However, there is no significant difference (α=0.05) between Vitamin C and total sugar of fresh-cut celery treated with ozonated water and non-treated. The best effect of preservation of fresh-cut celery appeared to be the treatment of water ozonated to a 0.18 ppm of concentration, with which the microbial population was lowered to 1.69 LogCFU/g.

A new immobilization method of filamentous cells such as Trichoderma reesei with thin paper carrier surfaces modified by electron beam irradiation has been developed, in which the immobilization was carried out by adhesion of the cells to hydrophobic polymer surfaces. The cells adhered firmly and grew on the surface of the carrier producing the enzyme cellulase. The productivity of the enzyme in immobilized growing cells was higher than that in free ceils. The diffusional barrier for substrate, nutrients and products (enzymes) in the immobilized carrier was improved. The effect of the condition of carrier preparation and immobilization on enzyme productivity was studied to get the optimum condition of immobilization.

The quantitative effects of fermentation time, temperature and broth content on mycelial growth in terms of dry cell weight (DCW) and extracellular polysaccharide content (EPC) produced by the fungus Boletus spp. ACCC 50328 in submerged fermentation were investigated separately using response surface methodology (RSM). The three factors chosen for the present investigation were based on the results of a previous Plackett-Burman (PB) design. The experimental data obtained were fitted to a second-order polynomial equation using multiple regression analysis and also analyzed by appropriate statistical methods. By solving the regression equation and also by analyzing the response surface contour plots, the optimal batch fermentation conditions were determined: under conditions of fermentation time, 10.7 days; temperature, 24.23℃; and broth content, 50.0ml,EPCreached 1053.987μml−1 of fermentation liquor.Whenfermentation time, emperature and broth content were 10.9 days, 26.77℃ and 64.9ml, respectively, DCW reached 4.216mgml−1. In order to obtain simultaneously the maximum yield of EPC and DCW, the above conditions would be 10.5 days, 26.23℃ and 58.70ml; and under these conditions, the maximum predicted yield of EPC and DCW were 992.258gml−1 and 4.163mgml−1, respectively. These predicted values were also verified by validation experiments.

Polymer hydrogels were obtained by radiation-induced copolymerization at-78℃ of aqueous solutions of acrylic and methacrylic esters. The matrices were characterized by equilibrium water content measurements, by optical microscopy observations and by scanning electron microscopy analysis. Yeast cells were immobilized on these hydrogels and the ethanol productivity by batch fermentation was determined. Matrix hydrophilicity and porosity were found to deeply intluence the adhesion of yeast cells and, hence, the ethanol productivity. The latter as well as other physico-chemical properties were also affected by the presence of a crosslinking agent added in small amounts to the polymerizing mixture.

Trichoderma reesei cells were immobilized on paper covered with hydrophobic monomer, trimethylpropane triacrylate by radiation polymerization. The effect of immobilization condition on enzyme productivity was studied by measuring filter paper and cellobiose activity. The cells were adhered and grew on the surface of the carrier with the polymer giving high enzyme productivity in the immobilized cells in comparison with the free cells. Optimum concentration and volume of the coating monomer for the preparation of the immobilized cells were obtained.

Radiation pretreatment of rice straw in the presence of alkafi (sodium hydroxide) solutions was studied using an electron beam accelerator. As irradiation dose was increased, the effect of a combination of sodium hydroxide and irradiation also increased. The glucose yield was increased by lengthening the treatment time with sodium hydroxide solution. The combination of irradiation and pulverization was effective for reducing the size of rice straw. The radiation pretreatment of the rice straw was also enhanced by heating after irradiation.

Fifamentous cells of Trichoderma reesei were immobilized using afibrous carrier covered with ionic-hydrophobic polymers, prepared by radiation copolymerization. The effect of the ionic properties of the polymers was investigated by changing the monomer composition in a trmethylpropane triaqlate (A-TMPT)- acrylic acid (AA) or methacryIic acid diethylaminoethyl ester (DEAEMA) system. More positive charge or less negative charge in the poIymers led to an increase in the growth of the cells immobilized on their surface. Enzyme productivity in the immobilized cells with AA-A- TMPTpolymer was higher than with a DEAEMA-A-TMPTpolymer.
