张俐娜,女,武汉大学化学与分子科学学院教授(博士生导师)。1963年毕业于武汉大学化学系。1985年获日本政府学术振兴协会奖学金(JSPS)赴大阪大学合作研究。1993年晋升为武汉大学教授,现兼任全国政协委员,民盟中央委员,中国化学会高分子专业委员会高分子表征学科组副主任,湖北省高校女高知科技创新协会副会长,担任《高分子学报》编委。长期从事高分子物理及天然高分子的基础和应用研究,主要包括真菌多糖二级结构、溶液性质及其与生物活性的构效关系;生物可降解材料、聚氨酯 / 天然高分子半互穿网络聚合物(semi-IPNs)、纤维素新溶剂和再生纤维素丝和膜及其共混材料、高分子纳米复合材料和蛋白塑料的结构与性能等。已在国内、外刊物发表220余篇论文,其中SCI收录180多篇。应用研究成果已获准发明专利17项、公告专利22多项,此外主编和合作2部专著“高分子物理近代研究方法”(2003年)和“高分子科学技术发展简史”(1994年)。她获得湖北省科技成果二等奖(1985年)、国家教委科技进步(甲类)三等奖(1995年)、国家教委科技进步(甲类)二等奖(1997年)和国家教育部自然科学二等奖(2004年)共四项奖。近5年主持科研项目12项,包括国家“863”项目、国家重点自然科学基金项目、香港卓越学科领域(AoE)项目的境外合作子课题、湖北省重点科技项目、国际合作及境外合作项目等,总经费近500万元。目前主讲三门课,包括“高分子化学与物理”本科生基础课,“高分子物理近代研究方法”和“高分子链形态学”硕士研究生课程。已累计指导36名博士生和34名硕士生。她在国际学术界较活跃,多次被国外及境外邀请出国讲学、作邀请报告及担任分会场主席,例如2000年在华盛顿第220届美国化学会学术会议担任高分子材料科学与工程学术会的分会主席;1994年和2002年两次在日本京都国际纤维素会议作特邀报告并任分会主席。曾赴美国、瑞典、日本、香港等地学术交流14次。已与日本和香港建立长期科研合作和学术交流关系,并获多种国外、境外基金派出20多名研究生和本科生出国、出境合作研究和交流。近年已接受外国留学博士生和访问学者。先后荣获全国优秀教师(1993年)、全国先进女职工(1994年)、全国先进工作者(2000年)及全国优秀巾帼发明者(2002年)称号。

In this article, a series of O-2-[2-(2-methoxyethoxy)ethoxy]acetyl celluloses with different degree of substitution (DS) values was synthesized by a homogeneous reaction of cellulose with 2-[2-(2-methoxyethoxy)ethoxy]acetyl chloride in a 10% (w/w) dimethylacetamide/lithium chloride solution, combined with pyridine as the acid acceptor. The total DS values of the derivatives in anhydroglucose units was determined by 1H and 13C NMR spectra, and ranged from 0.4 to 3.0, depending on the amount of acid chloride in the reaction. The effects of the total DS values and the O-2-[2-(2-methoxyethoxy) ethoxy] acetyl substituent distribution on the solubility of the derivatives were investigated. The lowest limit of the DS value for water-soluble O-2-[2-(2-methoxyethoxy) ethoxy] acetyl cellulose was approximately 0.5, which is lower than thatof methylcellulose. The amphiphilic derivatives with higher DS values than 1.7 exhibited a good solubility in both water and organic solvents, such as dimethyl sulfoxide, tetrahydrofuran, and chloroform. Sol-gel transition in aqueous solution was observed for the amphiphilic derivatives with a higher DS value than 1.7; the precipitation temperature (Tp) decreased as the DS value increased, showing that the derivatives are highly temperature sensitive. The thermal properties of the fully substituted derivative were measured using polarized microscopy, DSC, and X-ray diffraction; and are discussed in terms of phase transition of the sample derivatives.

β-D-Glucan PC3 (weight-average molecular mass=9.15•104, polydispersity=1.6), a linear glucan from Poria cocos sclerotium, was fractionated by a preparative size-exclusion chromatography (SEC) column packed with regenerated cellulose gels using dimethyl sulfoxide (DMSO) as mobile phase. The molecular masses and aggregation of the fractions were investigated by using analytical SEC, viscometry and membrane osmometry under different conditions. The results of SEC analysis proved that glucan PC3 forms partial aggregates in 0.25M LiCl/DMSO (namely, 0.25M LiCl in DMSO) at 40℃, showing a new shoulder peak of a higher-molecular-mass component in the chromatogram, and dissolves as a single-stranded chain at 80℃, giving a sharper peak. The mean apparent aggregation number (Nap) of the fractions in 0.25M LiCl/DMSO at 40℃ was 1.9, and much smaller than that of glucan PC3 in 20% cadoxen. It is indicated that the analytical SEC is more sensitive and direct in detection of the aggregates than other methods, and aggregation can be avoided by carrying out SEC measurement at 80℃. The Mark-Houwink equation for glucan PC3 in DMSO at 25℃ was established as [η]=0.107M0.50W in the Mw range of 7-11•104, suggesting that it behaves as a flexible coil.

Two kinds of regenerated cellulose microporous gel particles with mean pore sizes of 135nm and 370nm were satisfactorily prepared by mixing cellulose cuoxam with polyethylene glycol (PEG) as pore former. A preparative size-exclusion chromatography (SEC) column (550mm×320mm) packed with the gel particles mentioned above was used for the fractionation of a dextran in water. The exclusion limit and fractionation range of the stationary phase were molecular masses 7•105 and 3•103~7•105, respectively. The dextran (weight-average molecular mass=7.14•104, polydispersity=2) was fractionated into ten fractions with molecular masses range from 4•104 to 1•105, which were analyzed by an aqueous phase 21 analytical SEC. A daily throughput of 6.5g of the dextran was given with a flow-rate of 2.88ml min. The preparative SEC is simple, fast and cheap, and suitable for large-scale fractionation of the polysaccharides and other biopolymers.

Molecular morphologies and conformation transition of lentinan, a β-(1→3)-D-glucan from Lentinus edodes, were studied in aqueous NaOH solution by atomic force microscopy (AFM), viscometry, multiangle laser light scattering, and optical rotation measurements. The results revealed that lentinan exists as triple-helical chains and as single random-coil chains at NaOH concentration lower than 0.05M and higher than 0.08M, respectively. Moreover, the dramatic changes in weight-average molecular weight Mw, radius of gyration <s2>1/2, intrinsic viscosity [η], as well as specific optical rotation at 589nm [α]589 occurred in a narrow range of NaOH concentration between 0.05 and 0.08M NaOH, indicating that the helix-coil conformation transition of lentinan was carried out more easily than that of native schizophyllan and scleroglucan, and was irreversible. For the first time, we confirmed that the denatured lentinan molecule, which was dissolved in 0.15M NaOH to be disrupted into single coil chains, could be renatured as triple helical chain by dialyzing against abundant water in the regenerated cellulose tube at ambient temperature (15℃). In view of the AFM image, lentinan in aqueous solution exhibited the linear, circular, and branched species of triple helix compared with native linear schizophyllan or scleroglucan.

Seven water-insoluble (1→3)-β-D-glucan fractions TM8-1 to TM8-7 with weight-average molecular mass Mw ranged from 2.22 to 77.4×104 obtained from the sclerotia of Pleurotus tuberregium were carboxymethylated to produce the water-soluble fractions CTM8-1 to CTM8-7 with Mw ranged from 3.87 to 87.8×104. The degree of substitution (DS) of CTM8 fractions was analyzed by ir and elemental analysis (EA) to be 0.3-0.68. The Mw and the intrinsic viscosity [η] of the CTM8 fractions were measured by size-exclusion chromatography combined with multiangle laser light scattering (SEC-MALLS), MALLS, and viscometry in phosphate buffer solution (PBS) at 37℃. The dependencies of [η] and radius of gyration <s2>z1/2 on|Mw for the CTM8 samples were found to be [η]=(8.82±0.03)×10-3Mw 0.78±0.04 (cm3g-1) and <s2>z1/2=(3.09±0.05)×10-3Mw 0.75±0.06 (nm) in the Mw range from 3.87×104 to 53.2×104. Based on current theories for wormlike chain model, the conformational parameters of the CTM8 were obtained to be 790 (nm-1) for ML, 9.6 (nm) for q, which were higher than those of the native TM8 fractions, suggesting a more extended flexible chain of CTM8 in PBS. On the whole, the CTM8 fractions showed higher antitumor activity than their corresponding TM8 fractions. In view of data from molecular parameters and bioactivity, the antitumor activity of the CTM8 fractions may be correlated to its water solubility and relatively extended chain.

Dynamic light scattering measurements have been made on 15 fractions of aeromonas (A) gum, an extracellular heteropolysaccharide produced by the strain Aeromonas nichidenii, with dimethylsulfoxide containing 0.2M lithium chloride as the solvent at 25℃. Data for the ranslational diffusion coefficient D covering a molecular weight range from 4.5×105 to 2.1×106 and ratios of the z-average radius of gyration <s2>z 1/2 to the hydrodynamic radius RH (calculated with previous <s2>z data) suggest that the polymer behaves like a semiflexible chain in this solvent similar to the stiffness of cellulose derivatives. Thus the D data are analyzed on the basis of the Yamakawa-Fujii theory for the translational friction coefficient of a wormlike cylinder by coarsegraining the heteropolysaccharide molecule. Excluded-volume effects are taken into account in the quasi-two-parameter scheme, as was done previously for <s2>z and [η] (the intrinsic viscosity) of A gum in the same solvent. The molecular weight dependence of RH is found to be explained by the perturbed wormlike chain with a persistence length of 10nm, a linear mass density of 1350nm 1, an excluded-volume strength parameter of 1.3nm, and a chain diameter of 2.8nm. These parameters are in substantial agreement with those estimated previously from <s2>z and [η] data, demonstrating that the solution properties (D, <s2>z, and [η]) of the heteropolysaccharide are almost quantitatively described by the current theories for wormlike chains in the molecular weight range studied.

A water-insoluble polysaccharide (TM8) was isolated from sclerotium of Pleurotus tuberregiumby extraction with 0.5M NaOHaqueous solutions at 120℃. Its chemical structure was confirmed by infrared, high performance liquid chromatography, gas chromatography, and 13℃NMR in imethylsulfoxide (DMSO) to be composed of β-(1→3)-D-glucan backbone chain linked with a branched glucose, one out of every three glycosyl units being substituted at C6 position. The glucan TM8 in DMSO was fractionated by nonsolvent addition method into ten fractions, and the solution properties were studied by size exclusion chromatography combined with multiangle laser light scattering (SEC-MALLS) and viscometry in DMSO at 30℃ The dependencies of intrinsic viscosity[η] and radius of gyration <2>z 1/2 on weight-average molecular mass Mw for this glucan were found to be [η]=(9.24±0.2)×10-2Mw0.51±0.02(cm3g-1) and <s2>z 1/2=(3.67±0.3)×10-2Mw 0.56±0.02(nm) in the range of Mw from 1.07×104 to 77.4×104. Based on current theories for a wormlike chain, the conformational vparameters of the glucan TM8 were found to be 408(nm-1) for ML, 3.1(nm) for q, and 16.8 for C∞, suggesting that the polysaccharide exists as a dense random-coil chain in DMSO, due to branched structure.

Triple helical lentinan, β-(1-3)-D-glucan from Lentinus edodes, was denatured in dimethlysulfoxide (DMSO) into single random coils. The DMSO solutions of randomly coiled lentinan were diluted with pure water to different WH (the weight fraction of water in the mixed solvent), and their specific optical rotation[R]D, reduced viscosity (lnηr)/c, and hydrodynamic radius Rh were investigated as a function of wH and storage time t. With an increase of WH from 0.1 to 0.2, [α]D increased sharply, suggesting that transition of conformation of the macromolecules has occurred. When wH was lower than 0.1, (lnηr)/c of lentinan in water-diluted DMSO exhibited the almost same value as that in pure DMSO and changed hardly with increasing t. Interestingly, (lnηr)/c decreased to reach a minimum with a further increase of wH from 0.1 to 0.25 and then increased with a continuous increase of wH from 0.25 to 0.5. Both (lnηr)/c and Rh of the denatured lentinan in water-diluted DMSO with wH of 0.25 both exhibited a minimum, indicating that collapsed coil chains have occurred. All of the experimental findings revealed that the behaviors of lentinan in water-diluted DMSO solution with wH <0.1 were consistent with that in good solvent, DMSO. When wH=0.25, the quality of the mixed solvents became worse, and the dominant intramacromolecular hydrogenbond interaction enhanced, leading to minimum of viscosity and size of the chains as a result of the collapsed coils. When wH >0.25, the quality of the mixture weakens further, and the intermolecular hydrogen-bond interaction enhanced and was dominant, leading to aggregation of the collapsed chains.
