王彦广教授,男,1964年11月8日出生于陕西扶风,浙江大学化学系教授,博士生导师。毕业于兰州大学化学系,先后获化学学士(1981-1985)、硕士(1985-1988)和博士学位(1990-1993)。历任天津大学化学系助教(1988)、讲师(1989)和副教授(1993),浙江大学化学系博士后、副教授(1994-1997),香港中文大学化学系博士后研究员(1997-1998)。1998年至今任浙江大学化学系教授、博士生导师,现兼任浙江大学化学系系主任、中国化学会理事、浙江省化学会副理事长、中国化学会有机化学学科委员会委员、中国化学会化学生物学专业委员会委员。主要从事有机合成方法学、药物与生物活性有机化合物的合成、小分子荧光探针等研究。曾主持国家自然科学基金项目、国家重点攻关项目、973项目子课题等多项国家级科研项目。已发表SCI收录论文80余篇,编著教材3部,获教育部高校青年教师奖(2003)和中国化学会青年化学奖(1999)。

An unexpected 1-methylimidazole-catalyzed reaction of dimethyl acetylenedicarboxylate (DMAD) with in situ generated arylketenes leading to the synthesis of dimethyl 2-arylidenesuccinates under mild conditions is described. A plausible mechanism has been proposed.

Methyl 3,4-dihydropyrimidin-2(1H)-one-5-carboxylate derivatives 7 were effectively synthesized on the soluble polymer of polyethylene glycol (PEG) 4000 by heating or solvent-free microwave irradiation through the Biginelli three-component cyclocondensation. Compared with the classical solution-phase Biginelli reactions, the yields could be considerably improved and the reaction time could be shortened dramatically under microwave promotion in a liquid-phase protocol. Moreover, the polymer-supported synthesis provided the target compounds in high purity.

A three-step liquid-phase protocol for the synthesis of furanoquinolines and pyranoquinolines is described. The one-pot three-component aza-Diels-Alder reaction of PEG-supported benzaldehyde, anilines and 2,3-dihydrofuran or dihydropyran under TFA gave PEG-supported tetrahydroquinolines, which was assuredly oxidized by DDQ and then cleaved from the support to afford the corresponding furanoquinolines and pyranoquinolines with reasonable yields and purity.

A general method for the liquid-phase syntheses of 1,2,4-oxadiazolines through a 1,3-dipolar cycloaddition is described. The poly(ethylene glycol) (PEG)-supported imines reacted with nitrile oxides generated in situ from aldoximes, and then the adducts were cleaved from the PEG, to give 1,2,4-oxadiazolines in high yields and excellent purities.

A general and efficient one-step procedure has been developed for the synthesis of biaryl ethers via direct coupling of electron-deficient arylhalides or aryl triflates to aryl t-butyldimethylsilyl (TBS) ethers using Cs2CO3. The protocol was also applied to electron-rich arylhalides by addition of a catalytic amount of copper(I) iodide. The reaction is rapid and provides good to excellent yields.

Efficient and diastereoselective syntheses of trans b-lactams and anti b-aminocarbonyl compounds from carboximide 1 and imines 2 under the classical Reformatsky reaction conditions are described. An enolate-imine mechanism has been proposed for this reaction.

An efficient and rapid parallel liquid-phase synthesis of pyrazolines has been developed. The one-pot three-components reaction of polyethylene glycol (PEG)-supported acrylate 1, aldehyde 2 and aryl hydrazine 3 in the presence of chloramine-T in methanol gave the corresponding PEG-supported pyrazolines 5. Cleavage from the support under mild conditions afforded pyrazolines 6 in good yields (69-91%) and high purities (91-100%).

The liquid-phase synthesis of oxadiazolines via 1,3-dipolar cycloaddition reaction on poly(ethylene glycol) support (PEG) is described. 1,3-Dipolar cycloadditions of nitrile oxide generated in situ on soluble polymer with a variety of imines provided a library of 1,2,4-oxadiazolines in good yields and purity. The reactions were run in the homogeneous phase and the synthetic sequences were performed in parallel one-pot fashion. Quantitative conversion in the 1,3-dipolar cycloaddition reaction was verified by 1H NMR analysis. The polymer bound products were isolated in excellent yields by simple precipitation of the soluble support.

PEG-supported 2-methylphenacyl ester as a new photocleavable linker is reported. The photocleavage based on an efficient intramolecular hydrogen abstraction was carried out with 280-366nm UV irradiation in benzene or methanol to give the corresponding carboxylic acid in high yields and purities. The linker was suitable for aliphatic and aromatic carboxylic acids as well as protected amino acids.

7-Arylseleno-7-deoxydaunomycinone derivatives 3a-e and 7-thiophenyl-7-deoxydaunomycinones (7 and 8) were synthesized and the antitumor activities of them were evaluated against human stomach cancer SGC-7901 and human leukaemia HL60. The cytotoxic assay show that seleno daunomycinone derivatives are much better inhibitory activity than thiodaunomycinone and the structure-activity relationship was discussed. 7-Deoxydaunomycinone 4 was obtained when selenophenols were used in excess and the possible mechanism was proposed.
