黄锡荣,男,1963年8月生,1980-1984年在扬州大学化学系读本科化学专业,1986-1989年在山东大学化学系攻读无机化学专业硕士学位,1989年7月留校任教从事分析化学教学与科研工作;2001年获山东大学分析化学专业博士学位。自1989年工作以来,历任山东大学化学系助教、讲师、副教授。现为山东大学教授、博士生导师、胶体与界面化学教育部重点实验室研究员、微生物技术国家重点实验室客座研究员。近期研究方向:胶束酶学,环境与生物分析等。工作以来,主持或参与承担了12项省部级及以上纵向科研课题,在国内外核心刊物上共发表研究论文50多篇(其中约2/3在SCI刊物上),获校级及校级以上科研成果奖8项(其中省部级奖2项(均为第一作者)),省部级技术鉴定成果3项。近年来,依据学科发展趋势,结合我校在微生物技术及胶体与界面化学等领域的优势。在胶束酶学这一研究热点领域积极开展跨专业、跨学科合作研究,在疏水性芳香化合物的微生物降解及机制研究方面取得重要进展。

AOT)/isooctane/toluene/water reverse micelles were studied in this paper to understand the microheterogeneous effect of the medium on the catalytic properties of LiP hosted in the reverse micelle. Results showed that LiP from Phanerochaete chrysosporium could express its activity in the reverse micelles, but its activity depended, to a great extent, on the composition of the reverse micelles. Optimum activity occurred at a molar ratio of water to AOT (ω0) of 11, a pH value of 3.6, and a volume ratio of isooctane to toluene of 7-9. Under optimum conditions, the half-life of LiP was circa 12 h. The dependence of LiP activity on the volume fraction of water in the medium (θ), at a constant ω0 value of 11, indicated that VA was mainly solubilized in the pseudophase of the reverse micelle. Based on the pseudobiphasic model and the corresponding kinetic method, a linear line can be obtained in a plot of apparent Michaelis constant of VA vs θ, and the partition coefficient of VA between the pseudophase and the organic solvent phase was determined to be 35.8, which was higher than that (22.3) between bulk water and the corresponding mixed organic solvent. H2O2 inhibited LiP at concentrations higher than 80μM; this concentration value seems to be different from that in aqueous solution (about 3mM). The differences mentioned above should be ascribed to the microheterogeneity and the interface of the AOT reverse micelle.

The interaction of bovine serum albumin (BSA) with cationic surfactant cetylpyridinium bromide (CPB) in aqueous solution (pH 7.00) was studied quantitatively with ultraviolet (UV)-visible, far-UV, and near-UV circular dichroism, fluorescence, small angle x-ray scattering, and nuclear magnetic resonance measurement. It was found that CPB at low and high concentrations could induce the unfolding and refolding of BSA, respectively. We suggest that in the unfolding process, there existed BSA-CPB complex with the "necklace and bead" structure in which the unfolded BSA wrapped around CPB micelles, and that the hydrophobic interaction between the complexes led to the formation of large aggregates. The aromatic headgroup of CPB interacted with the tryptophan residues of BSA, resulting in the aromatic ring stacking between BSA and CPB. During the refolding process, the BSA molecule was penetrated into the rod micelle of CPB and the hydrophobic moiety of the BSA molecule was exposed outside while its hydrophilic part was hidden inside, thereby disrupting the aromatic ring stacking.

The activity of lignin peroxidase (LiP) in reversed micelles of polyoxyethylene lauryl ether (Brij30) changed with the molar ratio of water to the surfactant and the denaturant concentration of guanidinium chloride. At low water contents the activity of LiP could be enhanced by the denaturant at moderate concentration. This phenomenon, together with the spectral characteristics of the intrinsic fluorescence of LiP, suggested that the conformation of the active center of LiP was flexible.

根据研究发现,在有醇作助表面活性剂的CTAB反胶束中木素过氧化物酶(LiP)不能表现活力,而在水介质中CTAB对LiP的催化活性影响又不是很大。为了揭示其中醇的影响,本工作就不同碳链长度的醇对LiP酶催化性能的影响进行了研究。由于CTAB反胶束体系中醇浓度较高,且碳原子数大于4的直链醇在水中的溶解度又很小,为此采用了LiP可在其中显示催化活性的CTAB正胶束、AOT反胶束和Brij30反胶束作介质,通过研究这些介质中不同链长的醇对LiP催化活力的影响,来探讨CTAB反胶束中木素过氧化物酶(LiP)不能表现活力的原因。结果表明,不管表面活性剂聚集体的结构、电性质及反胶束大小如何,只要醇的浓度超过500mmol•L-1(丁醇≥1200mmol•L-1),LiP在上述原本可显示活力的介质中均无催化活性。据此推测CTAB反胶束中木素过氧化物酶(LiP)不能表现活力的原因主要是由助表面活性剂醇造成的。

微乳凝胶中小分子传质研究对于拓宽胶束酶学研究内涵、加速酶在生物合成与转化领域中的应用、研制高性能生物传感器等具有重要理论意义和潜在应用价值。以微乳液中二价金属离子与紫脲酸胺之间的配位反应为指示反应,采用分光光度技术,研究了金属离子在由阳离子表面活性剂十六烷基三甲基溴化胺构建的微乳凝胶中的传质问题。结果表明,在由含紫脲酸胺微乳液及含金属离子微乳凝胶构成的体系中,金属离子在微乳凝胶中的传质是金属离子与紫脲酸胺配位反应的限速步骤。为进一步证实上述结论,还对影响金属离子在微乳凝胶中传质的各种因素(如微乳液中水与表面活性剂的摩尔比值、分散相中甘油与水的配比等)进行了研究,结果也证实了上述结论。

Veratryl alcohol (VA) at higher concentration stimulated the lignin peroxidase (LiP)-catalyzed oxidation of phenolic compounds remarkably. This novel phenomenon was due to its competition with the phenols for the active site of the enzyme and to the high reactivity of the formed cation radical of VA (VAk) which resulted in an additional oxidation of the phenols. The influence of the nonionic surfactant Tween 80 on the VA-enhanced LiP-catalyzed oxidation of phenols depended on its concentration. At lower concentration it had a small synergetic effect but at higher concentration it decreased the initial rate. Studies of the capillary electrophoretic behavior of LiP in the presence of Tween 80 showed that this effect was caused by the surfactant aggregation on LiP which, at higher surfactant concentrations, might impede the access of VA to its binding site on LiP and, consequently, the VA+. formation.

酶是生物合成与转化的关键因素,酶催化性能既取决于酶蛋白分子自身结构,又取决于介质微环境。非水酶学和胶束酶学是近二三十年来兴起的酶学研究新领域,主要研究有机溶剂和反胶束中酶的催化性能并探索其潜在应用。表面活性剂既是区分这两个分支领域的标志,又是联系两者的桥梁。本文在对这两个领域的研究概况作一简要介绍的同时,着重介绍了表面活性剂修饰酶、微乳液凝胶固定化酶等研究进展。

The kinetics of ligninase-catalyzed oxidation of veratryl alcohol (VA) by H2O2 in the aqueous medium containing cationic surfactant cetyltrimethylammonium bromide (CTAB) has been investigated using spectrophotometric technique. Steady-state kinetic studies at different concentrations of CTAB indicate that the reaction follows a ping pong mechanism and the mechanism always holds but the kinetic parameters vary with CTAB concentrations. CTAB is a weak inhibitor for ligninase; it lowers the maximum initial velocity. CTAB also causes the Michaelis constant of H2O2 to decrease dramatically and that of VA to increase markedly. Based on the changes in kinetic parameters of the enzyme-catalyzed reaction at different CTAB concentrations (lower than, near to and larger than its critical micelle concentration) and the effects of the CTAB monomer and the micelles on the spectra of VA and its corresponding aldehyde, a conclusion could be made that modification of the enzymatic protein by the surfactant monomer should be responsible for the above-mentioned results.

The inhibition of ligninase by hydrogen peroxide in the presence of cationic surfactant CTAB was studied by kinetic spectrophotometric technique. Results showed that addition of CTAB enhanced the inhibition by H2O2, but it did not alter the inhibition pattern and the inhibition constant changed little with the concentration of CTAB. Modification of the enzymic protein by the surfactant monomer may be responsible for the above mentioned results.

Based on an unusual reversible sol-gel transition phenomenon, a novel method for the fabrication of a lipase electrode was developed. The response characteristics of the biosensor was studied by potentiometric technique using olive oil as substrate. After optimization, the lipase electrode demonstrated high activity and good stability.
