边专,男,1961年10月6日出生于湖南省株洲市,中共党员,博士导师。1983年毕业于湖北医学院口腔系,1987年获硕士学位,1998年获博士学位。1992年赴日本大阪大学进修,1993年回国。1997年破格晋升为主任医师、教授。现任武汉大学口腔医学院院长,同时担任中华口腔医学会常务理事,中国医师协会口腔医师分会副主任委员,湖北省医师协会副会长,IADR中国分会秘书长,IADR会员发展委员会委员,中华口腔医学会牙体牙髓病专业委员会副主任委员,中华口腔医学会口腔医学教育专业委员会副主任委员和十余家杂志的副主编、编委等多项社会兼职。主要从事龋病病因及预防方面的研究。在国家自然科学基金的连续资助下,他与课题组专家们一起成功地构建了变链菌葡糖基转移缺陷株,并在国际上首次构建了变链菌葡糖基转移酶高表达株,已成功将该株免疫奶牛,所获含高效价抗体的防龋牛奶能显著地减少致龋菌在人口腔中的定居,为免疫防龋开辟一条全新的思路。目前正在进行α-1,3-葡聚糖酶分子防龋的基础研究,口腔遗传病的家系收集及相关基因的分子遗传学研究,口腔微生物及菌斑形成机制的研究。近年来在国内外核心期刊发表论文60余篇,SCI全文收录8篇。2004年有2篇论文全文被SCI收录,皆为通讯作者。参编论著《口腔生物学》等12部。目前承担国家级、省、部级基金4项,协助完成的“口腔医学教学建设的探索实践”获省教学成果一等奖,“免疫防龋的系列研究” 2001年湖北省科技进步一等奖,参与编著的《口腔生物学》获卫生部科技进步二等奖,由他主持的“变形链球菌葡糖基转移酶高表达株被动免疫防龋疫苗的研制”获2000年省自然科学奖二等奖。自1997年起享受国家级政府特殊津贴,1998年被评为湖北省卫生系统先进工作者,2001年被评为湖北省优秀教育工作者,2002荣获湖北省优秀留学回国人员称号,2002年被评为卫生部有突出贡献的中青年专家。

目的 探索一种快速、简便地从人类唾液中同时检测变形链球菌和远缘链球菌的方法。方法 分别以变形链球菌gtfI和远缘链球菌gtfB基因设计两组成套引物,首先用套式PCR(二次PCR)检测变形链球菌和远缘链球菌标准株和临床株,然后用套式PCR 直接从唾液中检测这两种细菌。结果 变链菌(血清型c,e,f) 的标准株及临床株第1 次PCR 扩增产物为517 bp, 第2 次扩增产物为468bp;远缘链球菌(血清型d,g) 及道勒链球菌(血清型h)的标准株及临床株第1次PCR 扩增产物为712bp, 第2次扩增产物为663bp;其他异种菌均不能扩增出产物, 因此该PCR 检测具有高度的特异性。细菌纯培养物及唾液PCR 检测的敏感性分别是: 第1 次PCR 为105CFU, 第2次PCR 为103CFU。结论 套式PCR能快速在人类唾液中同时检测变形链球菌和远缘链球菌。该检测方法有望运用于临床检测,对揭示两种细菌与龋病发生关系的研究具有一定价值。

The aim of this study was to clarify the genotypic stability of mutans streptococci (MS) longitudinally during orthodontic treatment. Plaque samples were obtained from the supragingival smooth surface of the upper right teeth at four stages: prior to and after 1, 3 and 6 months of orthodontic treatment. Levels of total viable count, total streptococci and MS in dental plaque of 17 patients were recorded. Streptococci isolated from dental plaque samples were identified as MS on the basis of their morphological and biochemical properties. DNA was prepared from 713 strains of MS and the strains were then identified with polymerase chain reaction (PCR) again. Arbitrarily primed PCR (AP-PCR) fingerprinting was applied in determining the genotypes of MS. The results indicated that levels of total viable count, total streptococci and MS increased significantly after the fixed appliances were bonded. A maximum of 3 different genotypes were found in an individual. All the genotypes were found again after the application of the fixed appliances in 17 patients. A new AP-PCR typing pattern was found after the application of fixed appliances for 1 month in patient 1. That strain was not detected either prior to or after 3, or 6 months of treatment. The result indicated that the MS clones were very stable during orthodontic treatment.

【期刊论文】Transmission of Mutans Streptococci in Adults within a Chinese Population

The purpose of this study was to investigate whether there was transmission between adults in Chinese families using chromosomal DNA fingerprinting. Plaque samples were obtained from buccal and occlusal surfaces of 11 married couples. The colonization levels of mutans streptococci were estimated as colony-forming units per milliliter, and caries experience was scored by decayed, missing and filled teeth. Information about medical history, diet regimes and age at marriage was obtained. The isolates were serotyped by biochemical test and genotyped using the restriction endonuclease HaeIII. The procedure was repeated after 3 months. The results showed that 1 couple had the same genotype of mutans streptococcus at the first examination, but this could not be repeated for the husband who had lost his mutans streptococci at the second examination. On the contrary, another couple that did not have the same mutans streptococcal genotype at the beginning had the same genotype after 3 months. No matching of genotypes was observed within 8 couples. In 1 male, no mutans streptococci were detected, therefore that couple was not considered. These data indicate that spouses had a chance to be infected by strains of mutans streptococci from another person. The results suggest that there may be transmission between adults in Chinese families, but it may be difficult for mutans streptococci to colonize another mouth permanently.

To study and evaluate root canal preparation using greater taper(GT) hand files in curved root canals. Material and Methods: Twenty-three teech with 39 curved root canals in outpatients were instrumented with GT hand files, using the balance force technique, and obturated with standard gutta-percha points and a sealer(AHphus) using the lateral condensation method. Radiographs taken before, during and after the operation were used to evaluate the efficiency of preparation and obturation. Results: In all cases the master apical GT hand file followed the course of the canal so that a continuously tapered root canal shape was achieved. No apical blockage or obvious aberrations were found. Only one file fractured during instrumentation. The mean preparation time for each curved canal was 9.75

Fusion DNA Vaccine against Caries J Dent Res 83(3) 2004,-0001,():

Glucosyltransferases (GTFs) and A cell-surface protein (PAc) are two important virulence factors of the cariogenic organism Streptococcus mutans. They may mediate sucrose-independent or sucrose-dependent attachment of Streptococcus mutans to tooth surfaces, respectively. Thus, inhibiting both virulence factors is predicted to provide better protection against caries than inhibiting a single factor. To develop a highly efficient vaccine against caries, we constructed a fusion DNA vaccine, pGLUA-P, by cloning the GLU region of GTF into a DNA vaccine, pCIA-P, which encodes two highly conservative regions of PAc. In this report, we provide evidence that fewer caries lesions were observed in rats following subcutaneous injection of pGLUA-P, compared with pCIA-P, near the submandibular gland. Our findings suggest that a multigenic DNA vaccine may be more caries-preventive than a single-gene DNA vaccine.

Objective: Streptococcus mutans produces 3 distinct ghtcosyhransferases (GTase): GTase-1, GTase-SL and GTase-S. These enzymes synthesize from sucrose water-soluble and insohtble glucan that act cooperatively in mediating S. mutans adherence to tooth surface and dental plaque formation. This study investigated the relationship between the sucrose-dependent adherence and GTase-l expression level. Methods: In this study a Streptococcns-Escherichia eoli shuitter vector carrying GTase-I gene, pZBI, was constracted. The gene pZBI was then transformed into S. mutans GTase-I-deficient muttant B29, Two types of GTase-I expressing strain were obtained: GTase-I expression strain similar to the parent and a GTase-I overe.overexpression strain. Results: Sucrose-dependent adherence of these mutants was closely related to the amount of initial bacterial inoculum in the experiment, that is a small inoculum resulted in firm adherence and vice versa. The plasntid was reiso/ated from the GTase-I overexpressing strain. Nucleotide sequencing revealed that no mutation was found upstream of the GTase-I gene when compared with pZB1. Conclusion: The balance in the quantity of GTase-I and GTase-SI may play an important role in the adherence of S. matans in the presence of sucrose. (CJDR 1998; I:26-42)

DNA Vaccine against Caries J Dent Res 81 (11) 2002,-0001,():

A cell-surface protein antigen (PAc) of Streptococcus mutans is considered a virulence factor because it may mediate initial attachment of Streptococcus mutans to tooth surfaces. Thus, inhibiting PAc is predicted to provide protection against caries. To develop vaccines against dental caries, we constructed a DNA vaccine, pCIA-P, which encodes two high-conservative regions of PAc. Expression of the recombinant protein was obtained in eukaryotic cells in vitro and in vivo. In this report, we provide evidence that fewer caries lesions, and high levels of PAc-specific salivary IgA antibody and serum IgG antibody, were observed in gnotobiotic rats following targeted salivary gland (TSG) administration of pCIA-P. This study shows that the recombinant DNA vaccine pCIA-P could induce protective anticaries immune responses and that TSG immunization is a promising strategy for the inhibition of dental caries.

防龋疫苗的研究已有近40年的历史,多种防龋疫苗经证实确有防龋效果,然而防龋疫苗进入临床应用前必须解决使用安全性、防龋高效性、成本经济性等问题。近年来,随着免疫学、分子生物学和基因工程技术的飞速发展,从分子水平对变形链球菌(Mutans Streptococci,以下简称变链菌) 主要毒力因子表面蛋白抗原( surfaceprotein antigen, PAc,又称为抗原B、I/ II、IF、P1) 和葡糖基转移酶( glucosyl-transferaes, GTFs) 的认识逐步深入,极大地推动了防龋疫苗的研究。国内外学者就防疫苗的免疫原、免疫佐剂和免疫途径进行了大量的研究,取得了许多新的进展
