研究领域:能源生物技术,酶工程,水体生态学。生物柴油制备关键技术、工程化及产业化,生物柴油高值化技术及工程化应用;脂肪酶基因工程、蛋白质工程,发酵工程、酶固定化新技术及在生物能源、保健食品、手性药物拆分及新型有机物合成等领域的应用开发;生物制氢分子机制、代谢调控、制氢关键技术及工程化。水体自然态及胁迫态下,生态系统结构与功能动态,生产力生态学、生态能量学、精细食物网构建、水体生物膜的形成、结构及在物质循环中的作用。

Aspergillus niger lipase (ANL) is an important biocatalyst in the food processing industry.However, there is no report of its detailed three-dimensional structure because of difficultiesin crystallization. In this article, based on experimental data and bioinformational analysisresults, the structural features of ANL were simulated. Firstly, two recombinant ANLsexpressed in Pichia pastoris were purified to homogeneity and their corresponding secondarystructure compositions were determined by circular dichroism spectra. Secondly, the primarystructure, the secondary structure and the three-dimensional structure of ANL weremodeled by comparison with homologous lipases with known three-dimensional structuresusing the BioEdit software, lipase engineering database (http://www.led.uni-stuttgart.de/),PSIPRED server and SwissModel server. The predicted molecular structure of ANL presentedtypical features of the a/b hydrolase fold including positioning of the putative catalytictriad residues and the GXSXG signature motif. Comparison of the predicted threedimensionalstructure of ANL with the X-ray three-dimensional structure of A. niger feruloylesterase showed that the functional difference of interfacial activation between lipase andesterase was concerned with the difference in position of the lid. Our three-dimensionalmodel of ANL helps to modify lipase structure by protein engineering, which will furtherexpand the scope of application of ANL. During June 2003 to June 2004, an investigation on life cycle, production and trophic basis of twospecies of mayfly in a second-order river of Hanjiang River Basin, Hubei, China was conducted. Theresults showed Epeorus sinensis UMLER and Caenis nigropunctata WU both developed two generationsa year. The mean annual production and P/B ratio of E. sinensis were 9.154 g m-2 a-1 dry weight and16.0, and those of C. nigropunctata were 1.554 g m-2 a-1 and 9.6, respectively. For E. sinensis, the proportionscontributing to secondary production of the main food types were: amorphous detritus 33.46%,fungi 10.8%, vascular plant detritus 1.8%, diatoms 53.9%; for C. nigropunctata, the proportions were70.8%, 6.90%, 3.5% and 18.8%, respectively. Compared with those species reported in North Americaand Europe, although land use mode and local climate were greatly different in China, life history andtrophic basis of the mayflies seemed roughly similar, yet secondary production appeared to be muchhigher.

Although the lipase of Geotrichum candidum has been extensively reported, little attention has been focused on molecular genetic and biochemical characterizations of Galactomyces geotrichum lipases. A lipase gene from G. geotrichum Y05 was cloned from both genomic DNA and cDNA sources. Nucleotide sequencing revealed that the ggl gene has an ORF of 1692 bp without any introns, encoding a protein of 563 amino acid residues, including a potential signal sequence of 19 amino acid residues. The amino acid sequence of this lipase showed 86% identity to lipase of Trichosporon fermentans WU-C12. The mature lipase gene was subcloned into pPIC9K vector, and overexpressed in methylotrophic Pichia pastoris GS115. Active lipase was accumulated to the level of 100.0U/ml (0.4mg/ml) in the shake-flask culture, 10.4-fold higher than the activity of the original strain (9.6U/ml). This yield dramatically exceeds that previously reported with 23-50U/ml, 0.06mg/ml and 0.2mg/ml. The purified lipase exhibited several properties of significant industrial importance, such as pH and temperature stability, wide organic solvent tolerance and broad hydrolysis on vegetable oils. Such a combination of properties makes it a promising candidate for its application in non-aqueous biocatalysis, such as biodiesel production, selective hydrolysis or esterification for enrichment of PUFAs and oil-contaminated biodegradation, which have been drawn considerable attention currently.
