Locoweeds cause significant livestock poisoning and economic loss all over the world. The purpose of this study was to investigate the immune effects of locoweed toxin, swainsonine (SW) and human serum albumin (HSA) conjugate (SW–HSA), on goats. Twenty-four Sannon goats were randomly separated into immune control group (eight goats), immune poisoning group I (six goats), immune poisoning group II (six goats) and poisoning control group (four goats). Immune control group, immune poisoning groups I and II were first vaccinated with SW–HSA conjugate. The poisoning control group, immune poisoning groups I and II were then fed with 10.0 g/kg BW/day dry powder of Oxytropis kansuensis Bunge everyday morning. The immune control group was supplied with an alfalfa-based diet. Blood samples of these experimental animals were collected at different time interval. Immunoassay was performed using indirect ELISA and E-rosette technique. The results show that, after second booster immunization: (1) anti-SW antibody level in some goats increased to 28, which proves that SW-HSA conjugate can induce experimental animals to produce high-level anti-SW antibody in their bodies; (2) the high-level antibody in their bodies could maintain 30 days, and decreased gradually after poisoning experiment (in our experiment, there was a return of the antibody level on day 21 after poisoning experiment); (3) the decreasing of the E-rosette rate of the immune poisoning group was delayed 14 days, which suggests that SW-HSA could low down the loss of the immunity of the goats; (4) swainsonine concentration in the blood was significantly lower (p<0.01) in the immune poisoning groups than that in the poisoning control group, and there was no significant difference (p>0.01) between the two immune poisoning groups within the poisoning experiment.

The challenge of sampling blood from small animals has hampered the realization of quantitative small-animal PET. Difficulties associated with the conventional blood-sampling procedure need to be overcome to facilitate the full use of this technique in mice. Methods: We developed an automated blood-sampling device on an integrated microfluidic platform to withdraw small blood samples from mice. We demonstrate the feasibility of performing quantitative small-animal PET studies using 18F-FDG and input functions derived from the blood samples taken by the new device. 18F-FDG kinetics in the mouse brain and myocardial tissues were analyzed. Results: The studies showed that small (~220 nL) blood samples can be taken accurately in volume and precisely in time from the mouse without direct user intervention. The total blood loss in the animal was, 0.5%of the body weight, and radiation exposure to the investigators was minimized. Good model fittings to the brain and the myocardial tissue time–activity curves were obtained when the input functions were derived from the 18 serial blood samples. The R2 values of the curve fittings are.0.90 using a 18F-FDG3-compartmentmodeland.0.99 for Patlakanalysis. The 18F-FDG rate constants K*1, k*2, k*3, and k*4, obtained for the 4 mouse brains, were comparable. The cerebral glucose metabolic rates obtained from4 normoglycemic mice were 21.564.3mmol/min/100g (mean 6SD) under the influence of 1.5%isoflurane. By generating the whole-body parametric images of K FDG (mL/min/g), the uptake constant of 18F-FDG, we obtained similar pixel values as those obtained from the conventional regional analysis using tissue time–activity curves. Conclusion: With an automated microfluidic blood-sampling device, our studies showed that quantitative small-animal PET can be performed in mice routinely, reliably, and safely in a small-animal PET facility.

In this paper, we introduce a new approach for the in situ electrochemical fabrication of an individually addressable array of conducting polymer nanowires (CPNWs) positioned within an integrated microfluidic device and also demonstrate that such an integrated device can be used as a chemical sensor immediately after its construction.

An inorganic/organic composite hybrid nano-system has been successfully synthesized in which nanocrystalline quantum dots (QDs) were effectively immobilized within a photo-cross-linked poly(ethylene glycol) (PEG) hydrogel. The immobilization of 3.5-6.0 nm CdTe and 2.0-3.5 nm CdSe QDs within the PEG hydrogel network has been shown to be effective through utilization of physical trapping. These QD-immobilized gel systems demonstrated luminescence characteristics unique to semiconductor QD nanocrystals. Controlled particle extraction from the PEG hydrogel matrix may be possible via a photocleavage process. The solubility property of QDs was controlled through surface functionalization. It is envisioned that the unique photophysical properties of this new material can be utilized as a convenient signal transducer for chemo-/bio-sensing. A promising application of the described QD/PEG-NC hybrid system may be in the fields of fluoroimmunoassay and as a monitoring system for drug delivery and wound healing.

Since luminescent CdSe quantum dots (QDs) have shown great potential in biological labeling, the surface chemistry behavior of QDs at interfaces is of great research interest. In the present study, green-colored CdSe QDs were modified with hydrophobic chains of varying lengths [from C6 to C18]. These modified QDs can be utilized to form stable monolayers at the air/water interface. Surface pressure-area isotherms of modified QDs have been measured and limiting molecular areas have also been extrapolated in order to analyze the size of the QDs. UV absorption spectra of modified QDs at various surface pressures were also determined. Surface chemistry, as well as the topographic properties, of modified QDs in Langmuir and L-B films was discussed.

In this paper we present our surface chemistry studies of enzymatically polymerized, poly-coniferyl alcohol lignin model compound (dehydrogenate polymer a.k.a. ZL-DHP) at the air-water interface. Using the CHCl3yMeOH (5:1 v/v) spreading solvent, we found an average molecular area of ZL-DHP of approximately 1200 A2. The monolayer expresses a high compressibility with a collapsed area of 500 A? 2 and collapsed surface pressure of 28 mN m-1. In the range of applied surface pressures, ZL-DHP polymer have no phase changes, as shown by the very high linearity (Rs0.994) of absorbance vs. surface pressure cure. There was no symmetry transitions observed as shown by absence of shifts of absorption peak maximums.

Self-assembly property of a new liquid crystalline perylene diimide derivative, N,N?-bis[3-[2-[2-[2-(2-cyanoethoxy) ethoxy]ethoxy]ethoxy]-propyl]perylene-3,4: 9, 10-tetracarboxyldiimide (PPE4CN), was studied at the air/water interface by epifluorescence microscopy and on mica by environmental scanning electron microscopy (ESEM) and atomic force microscopy (AFM). The self-assembly process started at surface pressure 5mN/m. Molecular orientation changes of the PPE4CN molecules accompanied the self-assembly process at the air/water interface. It was found that the molecular orientation changed from face-on to edge-on orientation in the self-assembly process. These orientation changes were strongly supported by the analysis of the surface pressure-area isotherms, and by the opographic measurements at micro and nanoscale levels, including ESEM and AFM microscopies.

Two amphiphilic PAMAM dendrimers are synthesized by attaching 12-hydroxydodecanoic acid (HA) chains to a poly (amido amine) (PAMAM) dendrimer core (including generation I and generation II). The limiting molecular area obtained from the surface pressure-area isotherm at the air/water interface suggests the edge-on configuration for both dendrimers in Langmuir films. The edge-on arrangement is also supported by the atomic force microscopic (AFM) studies of the Langmuir–Blodgett films.
