郑文杰,暨南大学生命技术学院教授,主要从事无机化学、配位化学、生物无机化学、微量元素的化学与生物化学的教学科研工作。在无机化学专业招收培养硕士研究生,在生化与分子生物学专业招收培养博士研究生。在硒杂环化合物、硒的生物有机化等领域主持并完成了多项省部级科研项目,包括国家自然科学基金和省、教育部多项基金;获厅局级奖项两项(广东省高教厅科技进步奖,广东省教育厅自然科学奖);获国务院侨办所属高校优秀教师称号。在国内外各类学术期刊上发表重要研究论文90余篇(多篇被SCI和EI收录)。在纳米硒及其制备与保存和抗肿瘤药物先导化合物方面申请专利5项。
【研究兴趣与主要方向】
主要从事硒及其化合物的生物功能、化学基础及其应用研究。(1)在硒(Se)、碲(Te)的生物转化及其转化物的抗氧化、抗肿瘤活性研究。(2) 含硒小分子化合物设计合成及其生理、药理活性。(3)活性小分子对纳米硫、硒、碲的形貌调控、定向生长及其活性协同效应。
【学习及工作简历】
1998年获得博士学位,2000.7博士后出站,作为访问学者曾经在香港大学支志明教授实验室从事科研合作;1985- 暨南大学,从事教学、科研工作,现为硒化学与化学生物学实验室负责人。

Changes in photosynthetic pigment and protein contents, growth and the spectral characteristics in Chlorella vulgaris in response to selenium stress were investigated. Carotenoids (β-carotene and xanthophylls) and chlorophyll (Chl a and Chl b) contents in cells exposed to Se 50mg/L increased primarily and decreased afterwards, while photosynthetic pigments in cells exposed to Se 800mg/L decreased significantly. Chlorophyll (Chl) absorption peak at 693nm and prominent Chl emission peak at 700 nm, weakened significantly after Se stress. The excitation spectra showed a decrease in excitation energy transfer efficiency in Se-stressed cells. Total soluble protein decreased after Se stress. The changes in total Se, Mg2+, Ca2+, K+and Na+ concentrations in culture medium and cells were also determined by ICP-AES.

Accumulation of Se in mixotrophic culture of Spirulina platensis was investigated in this study. Results indicated that glucose was better than acetate as an organic carbon source for mixotrophic culture of S. platensis. Supplementation of glucose (2gL-1) significantly enhanced the biomass concentration (2.57gL-1) and the production of phycocyanin (0.279gL-1)-and-allophycocyanin-(0.126gL-1) in S. platensis, which were much higher than those of photoautotrophic culture (1.08gL-1,0.119g-L-1and0.042g-L-1,respectively). Stepwise addition of Se during the growth phase avoided the inhibitory effect of high Se concentration on the growth of S. platensis. The Se enrichment favored the production of phycocyanin and allophycocyanin in the algal cells. The highest Se yield (1033 lgL-1) was obtained at an accumulative Se concentration of 250mgL-1, with organic Se percentage, biomass concentration, phycocyanin and allophycocyanin yields of 92.3%, 2.55gL-1, 0.295gL-1and 0.153gL-1, respectively. These results indicated that the application of mixotrophic culture S. platensis with stepwise addition of Se to the medium could offer an effective and economical way for the production of high Se enriched algal products.

The feasibility of mixotrophic culture of high selenium-enriched Spirulina platensis was investigated in this study. The results indicated that supplementation of acetate to the mixotrophic culture of S. platensis led to a significant enhancement in biomass concentration, chlorophyll α, lutein, β-carotene, phycocyanin and allophycocyanin production when compared to the photoautotrophic culture. Stepwise addition of selenium (Se) source during the exponential growth phase of S. platensis can avoid the inhibitory effect of high Se concentration on cell growth. At an accumulative Se concentration of 450mgL−1 undermixotrophic culture conditions, the highest Se yield was obtained; while significant enhancement in Se accumulation capability, algal biomass concentration and photosynthetic pigment yield was also observed. These results indicated that the application of mixotrophic culture with stepwise addition of Se to culture of S. platensis offers a more effective and economical way for the production of high Se-enriched biological compounds.

用激光散射(LLS)及透射电镜(TEM)技术研究了Te (IV)-I--RhB水溶液中液相纳米粒子在自然状态下的聚集行为及粒径分布,并用共振瑞利散射光谱、荧光发射光谱、紫外-可见光谱对该液相纳米体系的光谱性质进行了表征.结果表明:在0.8 mol/ L HCL介质中,当 Te (IV)浓度为 4、8、12、16、20μg/ L时,Te (IV)-I--RhB 体系平均粒径分别为 192、228、262、278、300 nm;纳米粒子的形成及其聚合行为导致 RhB在558 nm处的吸收峰(A558)的减色效应及 598 nm处的发射峰(F598)的荧光猝灭和623 nm处的共振散射峰(I623)增强,并与 Te(IV)浓度呈良好的线性关系;体系在623 nm处的散射强度 I1/ 3与其粒径成正比。用等离子体原子发射光谱( ICP)、元素分析( EA)等手段对该纳米粒子进行表征,确定了其最简分子式为(RhB)4(TeI8)。

A fast protein liquid chromatographic method for purification of selenium-containing phycocyanin (Se-PC) from selenium-enriched Spirulina platensis was described in this study. The purification procedures involved fractionation by ammonium sulfate precipitation, DEAE-Sepharose ion-exchange chromatography and Sephacry S-300 size exclusion chromatography. The purity ratio (A620/A280) and the separation factor (A620/A655) of the purified Se-PC were 5.12 and 7.92, respectively. The Se concentration of purified Se-PC was 496.5 ugg-1 protein, as determined by ICP-AES analysis. The purity of the Se-PC was further characterized by UV-VIS and fluorescence spectrometry, SDS-PAGE, RP-HPLC and gel filtration HPLC. The apparent molecular mass of the native Se-PC determined by gel filtration HPLC was 109 kDa, indicating that the protein existed as a trimer. SDS-PAGE of the purified Se-PC yielded two major bands corresponding to the a and b subunits. A better separation of these two subunits was obtained by RP-HPLC. Identification of the a and b subunits separated by SDS-PAGE and RP-HPLC was achieved by peptide mass fingerprinting (PMF) using MALDI-TOF-TOF mass spectrometry.

研究了Te (IV) 胁迫下螺旋藻鲜活细胞与冻融破壁后细胞的光谱性质,吸收光谱表明Te (IV)胁迫下藻细胞的特征吸收峰明显降低,对于冻融后的藻细胞,藻蓝蛋白的吸收峰成为主峰,且随着Te (IV)浓度的增大其强度显著降低,说明高浓度Te (IV) 胁迫对藻蓝蛋白的损伤最为明显;荧光光谱显示,荧光峰的位置较对照组没有发生移动,但强度明显降低,表明在高浓度的Te (IV) 胁迫下螺旋藻中主要的光合色素遭到明显的损伤;冻融细胞残渣的荧光未观察到发射峰。

A simple and selective voltammetric method based on selenium-gold film modified glassy carbon electrode has been developed for investigating electrochemical reaction mechanism of selenocystine.With N2 saturated, redox reactions between selenocystine (SeC) and selenocysteine (SeCys) were judged to be two simple electron-transfer processes. With air saturated, the reduction reaction was diagnosed to be EC catalytic reaction (the chemical oxidation reaction of the SeCys by O2 (C) following the electron-transfer reaction (E)) and oxidation reaction is a simple electron-transfer process. With pure O2 saturated, only reduction peak was observed and the reaction was judged to be EC catalytic reaction. The electron-transfer numbers of redox reaction were calculated to be 2 by chronocoulometry and rotating disk electrode.

本文对合成的7 种含硒芳香杂环化合物进行了GC/ MS分析研究。结果表明:BS、MB、BBS和DBBS等4个化合物在色谱柱内的保留时间与它们的相对分子质量呈线性关系。所有化合物均可获得特征质谱,表现出含单个硒原子的分子离子或碎片离子特征峰簇,硒的两种主要同位素在峰簇中表现为主要峰M与(M-2)的相对丰度比约为2:1,可为鉴定含硒分子离子或碎片离子提供重要信息。新化合物1,2,52硒二唑并[3,4-d ]嘧啶-5,7-( 4H,6H )二酮(SPDO)在色谱柱内出现11.83 min 和7.96 min两个具有相同的质谱的色谱峰,被认为是互变异构体的峰。

研究了不同分子结构的硒杂环化合物-金属离子在乙醇溶液中的发光, 硒杂环均为苤硒脑类化合物(Piazselenoles,简称Pis):Piazselenole (PS); 4,5-benzopiazselenole (BP); 5-methyl-piazselenole (MP); 4,4’-dipiazselenole(DP)。结果表明:Pis的Em随分子共轭体系增大或共轭效应增强而出现有规律地红移;Sn (IV)对Pis的发光有明显的增敏作用:使DP的Em蓝移;对MP荧光减弱,对BP和DP均有明显的荧光增强作用,对常温下不发光的PS,在Sn (IV)-PS乙醇溶液有较强的特征荧光(Ex=355 nm, Em=426 nm);此外,Cr(III), Cd(II), Cu(II), Sb(III) 和Sn (IV)均使BP荧光增强,Fe (III)和Fe (II)浓度较大时使BP荧光减弱,Zn (II) 却几乎没有影响。
