刘义,博士,男,1970年11月出生,武汉大学化学与分子科学学院教授、博士生导师、博士后合作导师、享受国务院政府特殊津贴的专家、教育部高校优秀青年教师教学科研奖励计划入选者。
1988年入武汉大学化学系,本科、硕士、博士分别于1992、1994、1997年毕业于武汉大学化学系,1997年获理学博士学位,并留校任教。1997-1999年在武汉大学生命科学学院从事化学与生命科学交叉领域博士后研究;2000年在中科院水生所做高级访问学者;2002年在中科院大连化物所做高级访问学者;2003年8月至2004年2月在英国Queen’s University Belfast和University of East Anglia做高级访问学者。1999年晋升为副教授;2001年破格晋升为教授;2002年遴选为博士生导师;2004年3月至7月受香港裘搓基金会资助在香港浸会大学做访问教授。
于1999、2000、2002、2003年分别入选武汉大学青年学术骨干重点培养对象、教育部高校青年骨干教师培养计划、教育部高校优秀青年教师教学科研奖励计划、湖北省新世纪高层次人才培养工程。

The process that T4 phages multiply in host cells of Escherichia coli B was determined using LKB-2277 Bioactivity Monitor by means of stopped-flow method, and the growth was measured turbidometrically at the same time at 37 8C. By analyzing thermocurves, quantitative parameters could be obtained to characterize the interactions of host cells and phages. The parameters such as ka, Pmax, G etc. change regularly with the decrease of multiplicity of infection (MOI) value. Infection/lysis equations were fitted and the lytic rate constant kL was obtained. The results show that the metabolic activity of infected cells is more intensive than that of normal cells. The phenomenon of lysis inhibition (LIN) was first detected with the microcalorimetric method, and the mechanism is discussed.

The heat output of the non-growth metabolism of Chlorella vulgaris has been determined using an LKB-2277 BioActivity Monitor. The experimental results indicate that there is a turn-point on the metabolic thermogenic curves, which can be divided into two parts, an early phase and a later phase. The thermogenic curves of the metabolism of mitochondria isolated from the heart of chicken Star-cross 288 and the eect of La3+ on it were studied by using an LKB-2277 BioActivity Monitor, ampoule method, at 37℃. After isolation from the chicken heart tissue, mitochondria still have metabolic activity and can live for a long time by using the stored nutrients. From the thermogenic curves, we obtained the thermokinetic equations under dierent conditions. The kinetics show that La3 has changed the metabolism completely.

The microcalorimetric bioassay for acute cellular toxicity is based on metabolic heat production from cultured cells. Microcalorimetry is a quantitative, inexpensive, and versatile method for toxicology research. The biological response to toxicants is the inhibition of the heat production rate in cells and toxicity is expressed as the concentration of toxicant that is 50% effective in this inhibition (IC50). In this paper, the effect of Cd2+on Rhizopus nigricans growth was investigated at 25℃ The relationship between growth rate constants (k) and concentration of Cd (C) shows a logarithmic normal distribution, and described as k=1.2742×1061 exp[-1.810×10-3 (C+2 283.0)2], and IC50 is 0.72μg/ml. These signals are readily obtained by an LKB 2277-204 heat 50 conduction microcalorimeter.

The effect of Na2SeO3 on the growth of Halobacterium halobium R1 was investigated by means of microcalorimetry at 37°C. The biological response to toxicants is observed as the inhibition of the rate constant of growth of living cells. A low concentration of Na2SeO3 stimulated the growth of H. halobium R1, and a high concentration of Na2SeO3 inhibited the growth of H. halobium R1. Toxicity may be expressed as the half-inhibition concentration (IC50). The rate constants of growth (k) and the concentrations of Na2SeO3 (c) shows a linear relationship: k=1.790×10-6-2.27×10-3c. The value of IC50 obtained from the accompanying figure of I-c is 679 μg/mL.

The water-soluble quaternary ammonium salt of chitosan was synthesized by reaction of chitosan with glycidyl trimethylammonium chloride. It was characterized by Fourier transform infrared and carbon-13 magnetic resonance spectra. Its microbiostatic effects against Staphylococcus aureus, Escherichia coli and Candid albicans were explored by microcalorimetry. The antimicrobial activity of the quaternized chitosan was stronger in alkaline condition than that in weak acidic condition.

The interaction between monoammonium glycyrrhizinate (MAG) and bovine serum albumin (BSA) were studied by fluorescence and absorption spectroscopy. The quenching mechanism of fluorescence of bovine serum albumin by monoammonium glycyrrhizinate was discussed. The binding sites number n and apparent binding constant K were measured by fluorescence quenching method. The thermodynamic parameters [1]H◦, [1]G◦, [1]S◦ at different temperatures were calculated. The distance r etween donor (bovine serum albumin) and acceptor (monoammonium glycyrrhizinate) was obtained according to F˝orster theory of non-radiation energy transfer. The results of synchronous fluorescence spectra and UV-vis absorption spectra show that the conformation of bovine serum albumin has been changed.

The binding of Schiff base selenide, (2-hydroxy-benzimido)ethyl-n-hexylselenide, to bovine serum albumin (BSA) was studied using fluorescence spectroscopy. The measurement was performed in Tris-HCl buffer aqueous medium at pH 7.4. Stern-Volmer graphs were plotted and quenching constants were estimated. The quenching constant at 303 K was (1.639-0.046)1013 L mol-1 s-1. Decreased quenching was observed as temperature increased, but at the temperature range of 303-313 K, the association of Schiff base selenide to BSA was not significantly different. The static quenching presented in the system of Schiff base selenide and BSA. A complex was possibly formed between Schiff base selenide and BSA, which was responsible for the quenching of the fluorescence of BSA. This fact was also confirmed by differences in the absorption spectra of BSA before and after Schiff base selenide addition. The hydrophobic interaction was found to play a main role in the binding according to the thermodynamic parameters, enthalpy change (H) and entropy change (S) of reaction. Schiff base selenide most likely binds to the hydrophobic pockets within sub domain IIA of BSA, which can be proved by competition experiments for sodium dodecyl sulfate. By constant-wavelength synchronous fluorescence spectra, the influence of (2-hydroxy-benzimido)-ethyl-n-hexylselenide on the surrounding environment of tyrosine and tryptophan residues in BSA was also investigated. The red shift of the fluorescence peak of tryptophan residues indicated that the hydrophobic amino acid structure surrounding tryptophan residues in BSA collapsed slightly after the addition of (2-hydroxy-benzimido)ethyl-n-hexylselenide.

An on-line solution-reaction isoperibol calorimeter has been constructed. The performance of the apparatus was evaluated by measuring the molar enthalpy of solution of KCl inwater at 298.15 K. The uncertainty and the inaccurary of the experimental results were within±0.3% compared with the recommended reference data. Using the calorimeter, the molar enthalpies of reaction for the following two reactions: LaCl3-7H2O(s)+2Hhq(s)+NaAc(s)=La(hq)2Ac(s)+NaCl(s)+2HCl(g)+7H2O(l) and PrCl3-6H2O(s)+2Hhq(s)+NaAc(s)= Pr(hq)2Ac(s)+NaCl(s)+2HCl(g)+6H2O(l), were determined at T=298.15 K, as−(78.3±0.6) and−(97.3±0.5) kJ mol−l, respectively. From the above molar enthalpies of reaction and other auxiliary thermodynamic quantities, the standard molar enthalpies of formation of La(hq)2Ac and Pr(hq)2Ac, at T=298.15 K, have been derived to be−(1535.5±0.7) and−(1536.7±0.6) kJ mol−l, respectively
