马爱群,男,汉族,1957年生于湖北,心血管博士,教授,主任医师,博士生导师。现任西安交通大学第一医院副院长、大内科主任、心内科主任。国务院特殊津贴专家,教育部骨干教师,陕西省“3•5”人才,陕西省卫生厅“2•15”人才,原西安医科大学首批跨世纪学术带头人。
主要从事心力衰竭、血管相关疾病及心脏分子遗传学等方面的研究。先后主持国家自然科学基金2项、卫生部科研基金4项、教育部项目3项、省自然基金1项及国际合作项目4项,指导博士研究生申请到国家自然科学基金7项,是国家重大项目“973”医学类核心专家组成员,负责其中一项子课题研究,并以主要成员参加“九五”攻关及国家、省部级研究课题10项。共发表文章180余篇,其中SCI收录10篇。主编专著8部,参编专著6部。获省部级科技成果奖4项,厅级科技成果奖4项。

【期刊论文】微管蛋白在体外牵张刺激诱导心肌细胞肥大反应中的作用

目的 探讨细胞骨架微管蛋白在牵张刺激心肌细胞肥大反应中的作用。方法 通过体外牵张培养的心肌细胞,采用3H-亮氨酸掺入法和四甲基偶氮唑盐微量酶反应比以法反映心肌细胞肥大指示和海参心肌细胞数目,激光共聚焦定性、定量微管蛋白的合成和分布情况。结果 20%的持续性牵张引起心肌细胞的3H-亮氨酸掺入量[12h为(1870.1±112.5)cpm,24h为(2015.5±19.37)epm],与对照组[12h为(1122.7±12.5)epm,24h为(1210.7±222.7)cmp]比较显著增加,微管解聚剂秋水仙素(4μmol/L)可以明显抑制3H-亮氨酸掺入量[12h为(1292.7±142.6)epm,24h为(1327.8±97.4)epm]。牵张12h即可见微管蛋白荧光强度增强,24h部分细胞呈现微管蛋白结构坍塌,纹理模糊,荧光强度分布不均,微管蛋白最高光密度值1600(任意单位),可被4μmol/L秋水仙素抑制。结论 细胞骨架微管蛋白系统是牵张刺激诱导心肌细胞肥大反应的细胞内信号传递途径之一。

A number of mechanical cell stimulators have been used to study the effect of mechanical stmulation on cells in vitro. But the efficiencey of these devices is not fully desirable. We recently developed a new device for mechaical cell stimulation, the centrifugal force stretcher, and compared its efficacy with that of the traditional Flexercell Strain Unit. When the mechanical stretcher circumrotates with certain speed, cardiac myocytes attached on the plate are stretched and elongated by centrifugal force. Neonatal rat cardiac myocytes were isolated by enzymatic dissociation from the hearts of 3~5d old Sprague Dawley rats, and were mechanically stimulated by traditional 20% strech and 180r/min centrifugal force for 12 and 24h. The effects of mechanical stimulation on the hypertrophic response of neonatal rat cardiac myocytes and production of angiotensin Ⅱ(Ang Ⅱ) were examined. Compared with the non-stretch group. the radioactivity of 3H-leucine incororated into the stretch-stimulated cardiac myocytes in the centrifugal force stretch group was significantly higher[(1295. 17±51.19)vs(1122.67±51.63) in 12h; (1447.5±35.96)vs(1210.67±90.92)in 24h, P<0.05].Ang Ⅱ was also dramatically increased by 128% in 12h (P<0.05) and 139% in 24h (P<0.01). After the myocytes was stretched for 24h, the LDH level in the mediun in the Flexercell Strain Unit group was significantly higher than that in the centrifugal force group [(14.5±8.7) U/L vs (7.8±4.3)U/L, and Ang Ⅱ secretion of the cardiac myocytes, and the damage to the cells bronght by this stimulator is relatively slighter in comparison with the Flexercell Strain Unit.

目的:探讨OxLDL在动脉粥样硬化血管局部免疫病理损伤中的作用机制。方法:采用地高辛标记的寡核苷酸控针原位杂交技术,观察OxLDL对人外周血单核细胞MCP-1、PDGF、bFGF、IL-104种致炎细胞因子mRNA表达的影响。结果:OxLDL(10~30mg/L)同人外周血单核细胞孵育24小时后能够以剂量依赖性方式显著提高MCP-1、bFGF、FDGF、IL-104种致炎细胞因子mRNA表达(均p<0.001)。结论:OxLDL能以剂量依赖性方式直接刺激人外周血单核细胞分泌致炎细胞因子mRNA。

Mutations of the cardiac β-mysin heavy-chain (β-MHC) gene cause hypertrophic cardiomyopathy (HCM). Recent genetype-phenotype correlation studies have shown that mutations carry prognostic significance, We studied five unrelated polymerase chain reaction-single-strand conformation polymorphism (PCR-SSCP) method and the cycle sequencing of the PCR produts. A previously reported heterozygous mutation Arg719Gln (arginine→glutamine in codon 719) in exon 19 was found in one family. The proband is a 30-year-old female diagnosed at age of 25 years when she presented with showed moerate asymmetrical septal hypertrophy with left atrial enlargement. There was no obstruetion of the left ventricular outflow tract (LVOT) The patient also developed atrial fibrillation. The proband's mother and on of her ststers had similar clinical manifestations and both died sudenly at the age of 38 years. In addition. two silent nucleotide substitutions (ACT63ACC, TTT244TTC) in the cardiac β-MHC gene were klentified in the other four families.There synonyumous mutations did not cosegregate with the disease in the fanilies and they were also present in the 60 healthy and age-matched control subjects. Of the five families studied. we did not find any they were also present in the 60 healthy four families. The missense mutation Arg719Gln found in the Chines family is associated with a malignant phenotype f severe clinical symptoms and poor survival prognosis. This mutation also causes atrial enlargement and atrial fibrillation. Our study provides further evidence that the mutation, which alters the charge of the myosin heavy chain, is associated with a serious clinical outcome.

目的 研究高血压病(EH)胰岛素抵抗的相关因素。方法 观察EH串者和正常对照者的血清胰岛素样生长因子IGF-1)、血清生长激素、红细胞膜胰岛素受体活性、红细胞内ATP含量和24h尿钙、镁和红细胞钙、镁量,采用多因素相关和多元逐步回归的方法分析。结果 (1)EH组胰岛素敏感指数、血清生长激素、红细胞胰岛素低亲合受体结合位数、24h尿镁和红细胞镁含量降低,血IGF-红细胞胰岛素受体解离常数KD2,红细胞钙升高,差异有显著性。(2)EH组中,胰岛素敏感指数与平均动脉压血压、高血压家族史、体重指数、血清IGF-1、腰臂围比星负相关,与红细胞胰岛素低亲合受体结合位数、红细胞鲜含量呈正相关,有统计学意义。(3)EH组中,红细胞膜胰岛素低亲合受体水平与红细胞内镁含量、24h尿钙、尿镁排泄量正相关。(4)红细胞内镁含量与24h尿镁、尿钙排泄量正相关。结论 EH胰岛素抵抗的独立相关因素。细胞内镁水平降低是EH与胰岛素抵抗菌素联系的中心环节。

【期刊论文】大鼠容量超负荷性心肌肥厚时环磷酸腺苷、原癌基因蛋白、微管蛋白的变化及意义

目的 动态观察大鼠容量超负荷胜心肌肥厚模型中微管蛋白(tubulin)的变化以及环磷酸腺苷(cAMP)、D原癌基因蛋白(c-myc蛋白)对机械信号刺激的应答反应。方法 应用免疫方法、本光共聚焦扫描观察容量负荷后不同时点大鼠心股中tubulin、cAMP、c-myc蛋白的变化。结果 (1)容量负荷后tubulin光密度及象分布即刻均低于正常,在6h最为明显,在4d时星现恢状况,7d、11d、20d时微管结构排列星进行性紊乱趋势;(2)容量负荷后即刻及4~6h,eAMP明显下隆,其后逐渐上升,1周时超过正常水平,2周时下降,5月时显著下降;4~6h,cAMP蛋白表达率最高,其后逐渐下降,5个月时c-myc蛋白表达很低;(3)在慢性空量超负荷基础上给予急性容量荷时cAMP星现反常反应、c-myc蛋白无应答。结论 肥厚心股存在机械信号跨膜传递障碍。微管结构紊乱可能参与心股肥厚的形成过程及机械信号的跨膜传递过程。

Essential Hypertension (EH) is correlated with a metabolic disturbance characterized by insulin resistance (IR). In this study, there were observed in 47 subjects with EH and 30 subjects with normal blood pres-sure. Serum levels of Insulin-like growth factor-1 (IGF-1), serum levels of growth hormone (GH), the act v-ity of erythrocyte Insulin receptors (EIR), and ATP levels In erythrocytes, the insulin sensitivity index (ISI) was used to study the correlative factors of essential hypertension. 1. Among patients with EH, [SI. GH, and low-affinity Insulin binding sites of EIRs (RT2) were found to be in significantly lower amounts, IGF-1 levels and the KD2 of the erythrocyte insulin receptors were noted to be significantly h]gher. Compared with the control group, there was a mart ed digerenca between EH group and the control group. However, no statisgcal difference was observed between the hypertensive group and the group with normal blood pressure as regards erythrocyte ATP levels, high-affinity fnsulin binding sites of EIRs (RT1). and the KD of EIRs. 2. In the hypertensive group, the ISI was negatively correlated with mean arterial blood pressure (MBP), a family history of hypertension. the body mass index (BMI), the waist-hip ratio (WHR) and IGF-1 levels (r=-0.614△, -0.354**, -0.386**, -0.472**, -0.298*, △ρ<0.001, **ρ<0.01, *ρ<0.05), were positively correlated with RT2 and GH levels (r=0.301**, 0.275*, **p<0.01, *p<O.05), There were no statistically significant differences betwe ISI and age, sex, smoking history, drinking, RT1, KD1, and ATP levels In erythrocytes. 3, The ISI was user as the dependent variable in multiple linear stepwise regression analysis. MBP (X1), a famity history of ER (X2), WHR (X2), GH (X4), IGF-1 (X5), RT2 (X6), and the body mass index (X7) was used as independent variables. X1, X2, X3, X5, X6, and X7 were used in the eclualions, The results indicate that petients with EH also tend to have IR. We suggest that MBP, a family history of ypertension. BMh WHR, IGF-1, and RT2 might be independent factors affecting IR in cases of essantial hypertension.
