Serine:glyoxylate aminotransferase (SGAT) is a key enzyme of photorespiration.Changes in SGAT transcript levels and in enzyme activity followingshifts from light to dark or dark to light and after cultivation in mediasupplemented with 6-benzyladenine (6-BA), a synthetic cytokinin, wereanalysed in the green alga Chlamydomonas reinhardtii. Cells grown indarkness had higher transcript levels than those grown in the light andtranscript levels decreased upon transfer of cultures from dark to light; incontrast, they increased upon shift from light to dark. However, higherenzyme activities were detected in light-grown vs. dark-grown cells, whichcould be correlated with an elevated photorespiratory activity of the lightgrowncultures. Application of the synthetic cytokinin into the mediumpromoted both the accumulation of SGAT mRNA and enhanced SGATenzyme activity. Under these conditions an accelerated multiplication ofcells as well as higher final cell densities were observed. However, thesynthetic cytokinin did not affect chlorophyll content, which is differentfrom cytokinin effects observed in higher plants. The presence of cytokininlikeactivities in C. reinhardtii was confirmed using high-performance liquidchromatography. One of these activities was identified by mass-spectroscopyto be zeatin riboside.

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) induces selectivelyapoptosis in various tumor cells and virus-infectedcells, but rarely in normal cells. A chloroplast expressionvector, p64TRAIL, containing the cDNA codingfor the soluble TRAIL (sTRAIL), was constructed withclpP-trnL-petB-chlL-rpl23-rpl2 as Chlamydomonasreinhardtii plastid homologous recombinant fragmentsand spectinomycin-resistant aadA gene as aselect marker. The plasmid p64TRAIL was transferredinto the chloroplast genome of C. reinhardtii bythe biolistic method. Three independently transformedlines were obtained by 100 mg/L spectinomycinselection. PCR amplification, Southern blotanalysis of the sTRAIL coding region DNA and cultivationcells in the dark all showed that the exogenousDNA had been integrated into chloroplast genome ofC. reinhardtii. Western blot analysis showed thathuman soluble TRAIL was expressed in C. reinhardtiichloroplast. The densitometric analysis of Westernblot indicated that the expressed human sTRAILprotein in the chloroplasts of C. reinhardtii accountedfor about 0.43%―0.67% of the total soluble proteins.These experimental results demonstrated the possibilityof using transgenic chloroplasts of green alga asbioreactors for production of biopharmaceuticals.

The chlorophyll a (Chl a) fluorescence techniquewas applied to investigate damage of PS II during senescenceof excised half-fronds in Spirodela polyrrhiza P143.The green explants showed a typical Chl a fluorescencetransient, OJIP. After cultivation of explants under long-dayconditions for 8 days, all the J, I, and P steps disappeared,but a clear K band, an indication of senescence, wasobserved. JIP-test showed that at this time point, the photosyntheticperformance index (PI) dropped to zero and theactive reaction center (RC) per leaf cross-section (RC/CS)declined to 18%. As the oxygen-evolving complex (OEC)and the chlorophyll content all remained above 42%, it isproposed that the decline in RC contributes more to theappearance of the K band. Supplementation of 6-benzyladenine(6-BA) into the medium at the beginning of cultivationcaused dramatic increase in PI, OEC, RC/CS, andchlorophyll content, and at any time before the 8th dayreversed the senescence process of the explants. When 6-BA was added after 8 days of cultivation, the PI did notincrease anymore, RC/CS and OEC were maintained at22% and above 40%, respectively, and chlorophyll contentdecreased continuously further. These data support a viewthat the decline in RC is crucial for initiation of the irreversiblesenescence phase of explants cultivated under longdayconditions.

The relationship between senescence process and photorespiration was investigated in Spirodela polyrrhiza P143. The senescence of halffrondscultivated under long-day conditions was either delayed by elevating CO2 or enhanced by elevating O2 composition of the surroundingenvironment, which implicated the involvement of photorespiration in this process. Application of sucrose into the cultivation medium wasnot found to be able to postpone the senescence process. In testing the effect of upstream substances of photorespiratory pathway onsenescence, it was observed that applied at 0.5 mM, L-glycine and L-serine, but not glycollate, glyoxylate or formate, did induce senescence ofboth half-fronds and intact plants. No similar effect was observed of other amino acids, such as Arg, Trp and Cys, etc. As the role of L-serine ininducing senescence has been reported earlier, the possible role of L-glycine in inducing senescence of half-fronds under long-day conditionswas further investigated. It was found that the peak value of endogenous glycine accumulated during senescence occurred later than that ofserine and that the application of MOA, a specific inhibitor of glycine decarboxylase, inhibited the accumulation of serine and resulted in theaccumulation of glycine, but did not cause the irreversible process of senescence. Therefore, a causative role of L-glycine in the senescence ofhalf-fronds under long-day conditions could be ruled out. Measurement of changes in the endogenous serine level demonstrated that theappearance of endogenous serine peak value was postponed when half-fronds were cultivated at elevated CO2, which provides furtherevidence that L-serine is a specific factor of inducing senescence in duckweed.

f novel α,α,α-trifluoro-m-tolyl pyridazinone derivatives was synthesised. Herbicidal activitiesof the two intermediate compounds and 15 pyridazinone derivatives were evaluated through barnyardgrass andrape cup tests and Spirodela polyrrhiza (L.) Schleiden tests. Selected compounds were also evaluated undergreenhouse conditions. Bleaching activities were observed at 10 μgml−1 and some compounds exhibited herbicidalactivities at a rate of 300 g ha−1. The relationship between crystal structures and herbicidal activities is discussedthrough a comparison of two compounds (5a and 5f). 2006 Society of Chemical Industry

【目的】揭示籼粳基因组的多态性。【方法】选用分布于水稻12 条染色体上的RFLP 对多个水稻品种进行分析,结合生物信息学方法分析这些籼粳特异性标记的籼粳特异性及其序列分子的生物学基础。【结果】筛选到28 个RFLP 籼粳特异性探针,籼粳特异性标记的生物信息学分析表明,这些标记在基因组水平上存在差异。【结论】这些标记能对水稻品种进行籼粳稻分子分类。水稻基因组序列图的完成和释放,使得这些籼粳特异性标记揭示的基因组多态性可以在基因组序列水平上进行分析和验证。

Half-fronds of Spirodela polyrrhiza accumulated high level of endogenous serine when they had been cultivated under long-day conditionsand on a cytokinin-free medium for 4 days, after which the irreversible senescence processwas triggered. When 1 M6-benzyl adenine (6-BA)was included in the medium, the accumulation of endogenous serine was significantly inhibited, and at the same time the biological activityof half-fronds was increased. Application of exogenous l-serine into medium at 1mM initiated senescence of half-fronds, as well as intactplants, under the conditions of long-day and darkness. Application of 6-BA into the medium with l-serine postponed the appearance of serinelevel peak and senescence syndrome, although the senescence process was not prevented. It was also observed that half-fronds cultivated in thedarkness on a medium with only inorganic nutrients had a very low level of endogenous serine and maintained their biological activity for upto 2 months. These data shows that high level of endogenous l-serine can initiate senescence in Spirodela polyrriza and cytokinin can inhibitthe accumulation of endogenous l-serine. The possible reason for the accumulation of endogenous serine under long-day conditions wasinvestigated. The activity of serine glyoxylate aminotransferase (SGAT), which catalyzes the conversion of l-serine to hydroxypyruvate, wasfound to decrease gradually in half-fronds during cultivation under long-day conditions, but to be maintained by 6-BA application. Northernblot analysis was used to detect the changes in the level of transcript encoding SAGT, and results showed that the expression of this gene wasup-regulated by cytokinin but down-regulated during senescence.
