邹国林,1965年9月-1970年7月在武汉大学生物系微生物学专业本科学习;1979年2月-1980年9月在武汉大学生物系生物化学专业进修;1980年9月-1983年7月在武汉大学生物系生物化学专业作研究生,获理学硕士学位;1991年8月-1992年9月任美国西东大学化学系访问学者;从1983年7月至今,在武汉大学生物系(现生命科学学院)任教,历任助教、讲师、副教授、教授、硕士生导师、博士生导师;历任生物化学教研室副主任、生物系副系主任、生物化学与生物物理系副系主任、系主任、生命科学学院副院长。现任应用生物化学与分子生物学实验室主任、湖北省中小企业共性技术生物应用研发推广中心主任。兼任武汉市人大常委、科教文卫委员会委员,民建湖北省委会副主委、民建武汉市委会主委、民建中央科教委员会委员;湖北省暨武汉生物物理学会理事长,湖北省实验动物学会理事,国家教育部中小学教材审定委员会委员,湖北省政府科技奖励评审委员会委员,工业微生物湖北省重点实验室学术委员会主任(湖北工业大学)、中药资源与植物化学湖北省重点实验室学术委员会委员(湖北中医学院)、天然产物研究与应用湖北省重点实验室学术委员会委员(三峡大学)等。一直从事本科生和研究生教学工作,主讲多门课程;已培养博士后、博士生和硕士生30多人,目前正在培养的博士后、博士生和硕士生有20多人。自1988年至2004年,一直在院系岗位上任领导职务,主要负责科研工作。已承担国家、省部级及市科研项目20多项,在国内外学术刊物上已发表研究论文和综述文章280多篇,出版两部教材,获得过国家教育部科技进步奖等多项奖励,是享受国务院政府特殊津贴的有特殊贡献的专家。他领导的实验室隶属于病毒学国家重点实验室,主要从事应用基础和应用研究。该室的主要研究方向有三个:一是酶与催化,既研究天然酶,又研究模拟酶,研究它们的结构与功能的关系、催化机理、分子改造;二是金属螯合物(重点是博莱霉素、阿霉素等抗癌抗生素以及金属卟啉、酞菁、菲咯啉等)与自由基,这些金属螯合物与氧自由基密切相关,研究它们的分子药理,模拟酶性质及自由基理论;三是药学,重点研究抗病毒、抗菌、抗癌药物,包括中草药有效成分的发现、分离与表征,抗病毒基因工程疫苗的研制,药用酶的基因工程改造等。这3个研究方向之间有许多的交叉与渗透。

Daunomycin (DM) is a clinically used antitumor anthracycline antibiotic, which is transported primarily by human serum albumin (HSA) in the blood. Binding characteristics are therefore of interest for both the pharmacokinetics and pharmacodynamics of DM. A new optical biosensor technique based on the resonant mirrorwas used to characterize interaction ofDMwith HSA at different temperatures and the affinity constants were obtained. The HSA-DM interaction is exothermic with having favorable enthalpy and entropy followed by the integrated van't Hoff equation analysis. Fluorescence studies showed that DM has an ability to quench the intrinsic fluorescence of HSA through a static quenching procedure according to the Stern–Volmer equation and DM displays a pH-dependent binding affinity to HSA. Molecular modeling calculations showed that the DM binds HSA to a non-classical drug binding site and further analysis of the binding site of DM within the HSA molecule suggested that hydrophobic contacts, hydrogen bond formation and electrostatic interactions account for the binding of DM.

A three-dimensional structural model of nattokinase (NK) from Bacillus natto was constructed by homology modeling. High-resolution Xray structures of Subtilisin BPN' (SB), Subtilisin Carlsberg (SC), Subtilisin E (SE) and Subtilisin Savinase (SS), four proteins with sequential, structural and functional homology were used as templates. Initial models of NK were built by MODELLER and analyzed by the PROCHECK programs. The best quality model was chosen for further refinement by constrained molecular dynamics simulations. The overall quality of the refined model was evaluated. The refined model NKC1 was analyzed by different protein analysis programs including PROCHECK for the evaluation of Ramachandran plot quality, PROSA for testing interaction energies and WHATIF for the calculation of packing quality. This structure was found to be satisfactory and also stable at room temperature as demonstrated by a 300 ps long unconstrained molecular dynamics (MD) simulation. Further docking analysis promoted the coming of a new nucleophilic catalytic mechanism for NK, which is induced by attacking of hydroxyl rich in catalytic environment and locating of S221.

The objective of the present study was to investigate the feasibility of a subunit vaccine and a live bacteria vaccine to protect chickens against infectious bursal disease virus (IBDV) infection. The gene for VP2 of a new wild-type very virulent IBDV (vvIBDV) strain was cloned into an Escherichia coli expression system. Following expression, the recombinant VP2 and the induced expression bacteria were used to vaccinate chickens against virulent IBDV (vIBDV). Three weeks after the vaccination, chickens were inoculated with IBDV strain BC 6/85 by intranasal route or eyedrop route, prior to challenge anti-IBDV serum antibody was detected by AGP. All chickens vaccinated with recombinant VP2 could be detected anti-IBDV antibody. The subunit vaccine of recombinant VP2 conferred protection for 90-100% chickens, live bacteria vaccine of recombinant VP2 conferred protection for 85.7% chickens. The results indicate that E. coli BL21/pET28a-VP2 could be used to develop recombinant VP2 vaccine against infectious bursal disease in chickens

Reactive oxygen species play an important role in the mediation of cell killing. But the mechanistic links between reactive oxygen species (ROS) and cell death remains unclear. There was a speculation that ROS, especially hydroxyl radicals can induce necrosis but not apoptosis in cells treated with copper-1,10-phenanthroline, IICu(OP)2. In this paper, liver carcinoma cell line (Bel-7402) was treated with IICu(OP)2 and its effect was examined by several means. Cells were found to undergo changes characteristic of apoptosis. Hoechst staining showed apoptotic body appeared in the cells induced by IICu(OP)2. When DNA extracted from the cells treated with IICu(OP)2 was analyzed by agarose gel electrophoresis it generated 'ladder' pattern of discontinuousDNAfragments. Sub-G1 peakwas detected in treated cells. Furthermore, two different flowcytometric methods were used, each allowing us to relate the apoptotic cells to the position the cell-cycle position. Apoptosis induced by IICu(OP)2 was limited to G1-phase cells. Using cyclin analysis, the expression of cyclin E in G1 was blocked. Thus, it was concluded that IICu(OP)2 can induceG1-phase specific apoptosis in Bel-7402.
