1983年毕业于上海第一医学院, 分别于1989年和1994年在上海医科大学获医学硕士和博士学位。1996~1997年赴日本大阪大学医学部老年病医学教研室进修一年,为外国人客座研究员身份。现任复旦大学上海医学院生理学教授,博士生导师。自1986年以来, 主要从事中枢神经系统对心血管活动的调节,应激性高血压的发病机制,以及针刺降压机制的研究。至今已发表科研论文50余篇于国际和国内一级杂志上。研究成果曾获国家教委、卫生部、上海市和中国中西医结合学会科技进步奖多项。

Experiments were per foxed on Wistar or Sprague-Dawley rats of both sexes. Microinjection of cortieosterone (10 or 40ng/0.1μl/site) into the rostral ventrolateral medulla (rVLM) caused an increase in systolic blood pressure (SBP). heart rate (HR) and pressor response induced by stimulation of the dorsal periaqueductal grey (dPAG) in the midbrain. Microiujection of aldosterone (10 or 40 ng/0.1μl/site) into the rVLM had similar effects showing a higher leveI and longer period than that of eorticosterone. All thee effects wen dose dependent. Mieroinjection of glucocorticoid antagonist RU 38486 (40ng/0,1μl/site) or mineralocorticoid an tagonist spironolactone (40ng/0.1μl/site) caused a decrease in SBP, H R and the pressor response induced by stimulation of the dPAG The inhibitory effects of splronolactone were more apparent. These results suggest that both corticoids could exert ccntral modulatory effects on the resting cardiovascular activities and facilitate the pressor response during a defense reaction, and the rVLM is an essential area for the location of the central modulation. These effects may play an important role in the incidence and development of hypertension induced by strcss.

Hypertension was induced in Sprague-Dawley and Wistar rats by irregular foot shrieks combined with a buzzing noise for 2h twice a day for 1-2 weeks. The plasma cat echo[amine, cortico-sterone, angiotensin Ⅱ, glucose and lipids were found to increase in parallel. The acetylcho[ine (ACh) and choline ace-tyltransferase in rostrai ventrolateral medulla (rVLM) in-creased markedly). Microinjection of ACh or cholinergic ago-nists into rVLM induced a pressor effect, and microinjection of M receptor blockers had a depressor effect. Elecrtophysio-logical studies showed that the stress-induced hypertension was closely related to the activation ofa cholinergie system in rVLM. Microinjection of cortieoids into rVLM had led to a pressor response which could be blocked by Ru38486, spiro-nolactone, cholinergic blockers or verapamih Microinjection of morphine and μ-or δ-receptor agonists into rVLM caused bradycardia and a reduction of arlerial pressure that could be blocked by naloxone.

The experiments were performed on Wistar or Sprague-Dawley rats of both seaes divided at random into stress and control groups The rats in the stress groups were put into cages and subjected to electric foot shocks and noises for 9-15 days. which caused an increase in blood pressure (BP) and heart rate (HR) In hyper tenslve rats DKJ-21 (4mg/1ml) was injured intravenously (Lv) and 0.5-10h after administration the BP and HR dropped from the high level to normotensive level. In normotensive rats. however, administration DKJ-I had no effect on BP 01 HR In separate groups of normtensive rats. pretreatment of DKJ-21 (4mg/l ml.iv) blocked the pressor and tachycardiac effect induced by microinjection of physostigmine (0.4μg/0.l/site) corticosterone (40ng/0.1μl/site) or aldostcronc (40ng/0.1μl/site) into the rostra1 ventrolateral medulla (rVLM) Furthemore. DKJ-21 also attenuated the enhancement ol the pressor response to stimulation of the defense area in the midbrain, which was induced by microinjection oF drugs (mentioned above) into the rVLM These results indicate that i.v DKJ-21 can selectively block the muscarinic receptors in the rVLM in stress-induced hypertensive rats. which suggests that abnormal enhamcement of cholinergic mechanism in the rVLM tony be related to hypertensive effects of corticoids in this area

Experiments were performed on anaesthetized Wistar or Sprague-Dawley rats of both sexes, Microinjection of an acetycholinesterase inhibitor physostigmine (0.4μg/0.1μl/site) or acetylcholine pressure (BP) heart rate (HR) and the pressor response produced by stimulation of the dorsal periaqueductal grey (Dpag) in the midbrain Prior microinjection of the calcium channel blocker verapamil (0.24μg/0.1μl/site) into the same sites blocked the cardiovascular effect in response to the respective microinjection of the drugs mentioned above Moreover verapanmil pretreatment blocked the pressor and tachyardiac effect induced by respective microinjection of corticosterone (40μg/0.1μl/site) or aldosterone (40μg/0.1μl/site) into the Rvlm, as well as the enhancement of the pressor response to stimulation of the dPAG induced by microinjection of corticoids into the rVLM These results suggest that the enhancement of cardiovascular activites mediated by cholinergic mechanisms may be due to the activation of postsynaptic calcium channels of neurons in the rVLM The corticosteroid effect seems to be mediated by similar mechanisms

本工作观察到应激性高血压大鼠发病过程中伴随着出现血浆儿茶酚胺、皮质酮以及血糖。血脂的平行升高。肾上腺皮质激素微量注射至延髓头端腹外侧区(rVLM)可引起升压、加快心率及增强防御性升压反应,该作用可能是应激性高血压发病机制的重要环节之一,其机制可能通过对该区心血管神经元的快速膜效应而实现,并与该区胆碱能系统活动的异常增强和钙通道的激活有关。而rVLM的NO系坑对该区心血管神经元具有抑制性作用。

To elucidate the role of the rostral veetrolateral medulla (RVLM) in cardiovascular control through the release of central amino acid neurotransmitters, experiments were per-formed in Sprague-Dawtey (normotensive) rats and spontane-ously hypertensive rats (SHR) anesthetized with urethane by using microdialysis sampling from the RVLM for determination of amino acid neurotransmitters. The baseline release of the excitatory amino acid neurotransmitter, glutamate (GLU) from the RVLM in SHR was higher and these of the inhybit cry amino acid neurotransmitters, glycine (GLY), taurine (TAU), and y-aminobutyric acid (GABA), were lower than in normotensive rats. Microinjection of angiotensin ll (ANG Ⅱ) into the RVLM caused a dose-dependent increase in mean arterial pressure (MAP) and heart rate (HR), accompanied by increased release of GLU in the RVLM. In contrast, microinjection of the ANG ll type 1 receptor (AT1) antagonist CV 11974 into the RVLM reduced MAP and HR, accompanied by increased release of GLY, TAU and GABA. These changes in MAP and HR after administration of ANG ll or AT, antagonist were partially blacked by the use of the corresponding antagonist of each amino acid neurotrans-mitter. Furthermore, these effects were more prominently seen in SHR than in normotensive rats. These results suggest that the release of amino acid neurotransmitters mediate the cardiovaseutar effects of the aag[otensin system in the RVLM, which may be involved in the generation of hypertension in SHR.

采用微量注射、微透析、高效液相色谱-荧光测定等技术和方法,观察和分析了血管紧张衷-(1-7)[Ang(1-7)]在延髓头端腹外侧(RVLM)与氨基酸类递质释放之问的关系。在麻醉大鼠RVLM注射Ang(1-7)可引起血压升高。同时伴RVLM兴奋性氨基酸(EAA)释放增多;在RVLM注射Ang-(1-7)选择性受体拮抗剂Ang779可引起血压降低,同时伴RVLM EAA释放减少和抑制性氪基酸(LAA)释放增多。Ang(1-7)的升压作用和Ang779的降压作用均可被相应的氨基酸受体拮抗剂部分阻断。结果提示,Ang-(1-7)在RVLM的升压效应可能部分是通过EAA释放增多所致;而Ang799在RVLM的降压效应可能部分是通过EAA释放减少、IAA释救增多所致。

The hypertensive rat model was made by chro'nic stress of electric foot-shocks and noises. On such hypertensive rats. when anesthetized with urethane and chloralose, the electroacupuncture (EA) to bilateral "Zusanli (st.36)" acupolnts for 20min, could resuh in a depressor (including both systolic and diastolic pressure) and bradycardiac response as well as an attenuation in the maximum of left ventficular pressure, end diastolic pressure and ±dp/dt. fit power spectrum analysis of heart rate variability aspect, EA could increase all total variance, very low frequency component, low frequency component and the ratio of low frequency component and high frequency component, When EA with microinjection of Nω-Nitro-L-Arginine, a blocker of the formation of nitric oxide, into the ventral periaqueductal gray matter (vPAG), the above effects of EA were abolished or reduced significantly. The results suggest that the depressor effect of EA on stress-induced hypertensive rats might be mediated by nitric oxide in the vPAG due to activation of sympathegc inhibitory system and by attenuated cardiac activities.

研究下丘脑前核微量注射褪黑素及其受体拮抗剂对正常血压和应激性高血压大鼠心血管活动的影响。方法:微量注射褪黑素及其受体拮抗剂至下丘及前区前核,记录血压、平均动脉和心率。结果:微量注射褪黑素可降低平均动脉压,其竞争性ML1受体拮抗剂luzindole可完全阻断褪黑素的降压反应,而其ML2受体拮抗剂prazosin不能阻断褪黑素的降压反应。结论:褪黑素为一种降压因子,其降压反应主要通过激活ML1受体,而不是ML2受体来介导的,下丘脑前核是褪黑素影响心血管活动的重要中枢部位。

First published January 11. 2002; 10.1152/japplphysiol. 00261. 2001. -In the present study, the changes of amino acids release in the spinal cord aider the application of angiotensin H (ANG ID in tee rostral ventrolaterul medulla (RVLM) and the distribution of ANG receptors on neurons of the RVLM were investigated. A micredialysis experiment showed that micminjection of an-giotonste Ⅱ into the RVLM sigulficantly (P<0.01) increased the release of aspartate and glutamate in the intetmledioiat-eral column of the spinal cord. Immunofluorecence tete-nique combined with confocal microscopy demonstrated that most of the glutematergic and GABAergic neurons in the RVLM of both Wistar and spantaneously hypertensive rats (SHR) were double labeled with ANG type 1 (AT2) recptor. Immuaocytochemical studies demonstrated that tee mean optic density of AT, receptor oftbe cell surFace as we)] as the whole cell was higher (P<0.05) in SHR than that in Wistar rats, indicating teat tee higher expression of AT1 receptors in the RVLM may contribute to the higher responsiveness of SHR to ANG II stimulation. Immunogold staining and elec-tronmicroscopic study demonstrated that AT1 receptor in the RVLM was distributed on the rough endoplasmic retjculum, cell membrane, and nerve processes The moults suggest teat effects evoked by ANG II in the RVLM are closely related to gintamatergic and GABAergie pateways. These results indi-rectly support the hypothesis that ANG Ⅱ in the RVLM may activate vasomotor sympathetic gintamatergic neurons, lead-ing to an increase in sympathetic nerve activity and arterial bled pressure.
