摘要

用110g/100mL纤维素酶(Onozuka RO10),011g/100mL果胶酶(Mecerozyme RO10)及10mmol/L CaCl2•2H2O,017mmol/L KH2PO4和015mol/L甘露醇的混合酶液,从14~18d苗龄的不结球白菜子叶上分离出高产率的原生质体。原生质体培养在KM8P1附加2,4OD 015mg/L、6OBA0125mg/L、NAA 015mg/L、葡萄糖910g/100mL、蔗糖110g/100mL、半乳糖0103g/100mL液体培养基上,分裂旺盛。形成愈伤组织后经芽诱导和生根培养,获得了再生植株。

摘要

取不结球白菜OguCMS 5d苗龄的下胚轴,在20mg/mL纤维素酶(Onozuka R210),10mg/mL果胶酶(Mecerozyme R210)及CaCl2•2H2O 10mmol/L、KH2PO40.7mmol/L、甘露醇0.5mol/L的酶液中游离36h,500r/min下离心收集下胚轴原生质体。原生质体在KM8P1附加2,42D0.5mg/L、62BA 0.25mg/L、NAA1.0mg/L的培养基上培养21d形成小愈伤组织,经MS附加2,42D1.0mg/L增殖愈伤组织,在MS附加62BA 10.0mg/L和NAA 0.3mg/L培养基上进行芽分化培养,并在无任何激素的MS培养基上生根,14d后即可获得完整的再生植株。

Bulk segregant analysis (BSA) and random amplified polymorphic DNA (RAPD) were employed to identify molecular markers linked to a self-incompatibility gene in self-incompatible and self-compatible near-isogenic lines of non-heading Chinese cabbage (B. campestris L. ssp. chinensis Makino var. communis Tsen et Lee). 【期刊论文】Creation of a New Germplasm of CMS Non-Heading Chinese Cabbage

摘要

A new germplasm ZS6(A)10 was created by asymmetric cell fusion with progeny plants identified in both field and laboratory. Results showed that the material displayed 100% sterility rate, 100% sterility degree, it had 4 nectaries and no chlorosis at low temperature. The seed-set was identical to that of the maintainer, which was significantly higher than that of the parental sterile line. The chromosome number of the progeny plant was the same as the maintainer (2n=20). The POD isoenzyme of the radicle and cotyledons, and the EST isoenzyme of the hypocotyl were notably different with those of the maintainer and the parental sterile line. The cpDNA and mtDNA contents of the progeny plants were intermediate between the maintainer and the parental sterile line. PCR analysis of cpDNA and mtDNA confirmed that ZS6(A)10 was a somatic hybrid and its progeny 98HQiu-45 was a new germplasm of Cytoplasmic Male Sterile (CMS) non-heading Chinese Cabbage.
