Four new steroidal alkaloids, puqienine A (1), puqienine B (2), N-demethylpuqietinone (3), and puqietinonoside (4), along with a known steroidal alkaloid, puqietinone, were isolated from the bulbs of Fritillaria puqiensis. The structures of these compounds were elucidated on the basis of spectroscopic analysis. The compounds exhibited significant antitussive activity on ammonia liquor-induced cough in mice. Furthermore, the compounds were evaluated for activities against A549 human lung carcinoma cell line, BGC-823 human stomach adenocarcinoma cell line, SMMC-7721 human hepatocarcinoma cell line, and HL-60 human promyelocytic leukemia cell line.

The present paper describes the development of a most simple, sensitive, and specific gas chromatographic method to date, for the direct determination of seven major bioactive isosteroidal alkaloids, namely ebeiedine, ebeiedinone, ebeienine, hupehenine, isoverticine, verticine, verticinone and imperialine, in Fritillaria species, a commonly used antitussive traditional Chinese medicinal (TCM) herb. In the present study, a commercially available Supelco SAC-5 capillary column (30m, 30.25mm, 0.25mm) specifically designed for the analysis of steroids was utilized for the direct determination of Fritillaria alkaloids. Calibration curves were obtained by spiking authentic compounds and the internal standard (solanidine) into herbal samples prior to extraction. Extraction was conducted simply by shaking the pre-alkalized diethyl ether solution 2 (5.0ml) containing dried herb (0.1g) for 2h. All calibration curves showed good linear regressions (r.0.995) within test ranges. The assay was reproducible and accurate with the overall intra- and inter-day variation and accuracy of less than 10% and more than 90%, respectively. The developed GC method was successfully utilized to analyze seven major bioactive alkaloids in seven Fritillaria species, and the results demonstrate that this direct GC analytical method is suitable for the quality control of this commonly used antitussive TCM herb.

Cordyceps sinensis is a well-known traditional Chinese medicine, and some of the active components are nucleosides. The analysis of nucleosides in Cordyceps material has been performed by reversed-phase high-performance liquid chromatography (HPLC) with gradient elution or by spectrometry. Here, we have explored the possibility of using capillary electrophoresis to determine the content of three major nucleosides (adenosine, guanosine and uridine) in Cordyceps. Capillary electrophoresis needs no gradients, and it provides a better separation due to its higher efficiency. In order to optimize the resolution, the separation of adenosine, guanosine and uridine was determined in Cordyceps with respect to the variation of buffer concentration, pH, temperature, and voltage. By using the calibrated electrophoresis system, the separation was achieved for the three nucleosides in less than 10 min with a background electrolyte consisting of 0.2M boric acid-sodium hydroxide buffer, pH 8.5. The nucleoside contents of various types of natural Cordyceps and cultured Cordyceps mycelia were determined and compared. There was a great variation of nucleoside content in different sources of Cordyceps; the cultured Cordyceps mycelia, however, contains a much higher concentration than the natural Cordyceps.

A new HPLC coupled with evaporative light scattering detection (ELSD) method has been developed for the simultaneous quantitative determination of seven major saponins, namely macranthoidin B (1), macranthoidin A (2), dipsacoside B (3), hederagenin 28-O--d-glucopyranosyl(6→1)-O--d-glucopyranosyl ester (4), macranthoside B (5), macranthoside A (6), and hederagenin-3-O-[1]-larabinopyranosyl(2→1)-O-[1]-l-rhamnopyranoside (7) in Flos Lonicerae, a commonly used traditional Chinese medicine (TCM) herb. Simultaneous separation of these seven saponins was achieved on a C18 analytical column. The mobile phase consisted of (A) acetonitrile-acetic acid (95:0.5) and (B) 0.5% aqueous acetic acid using a gradient elution of 29% A at 0-10min, 29-46%A at 10-25min and 46%A at 25-30min. The drift tube temperature of ELSD was set at 106℃, and with the nitrogen flow-rate of 2.6l/min. All calibration curves showed good linear regression (r2>0.9922) within test ranges. This method showed good reproducibility for the quantification of these seven saponins in Flos Lonicerae with intra- and inter-day variations of less than 3.0% and 6.0%, respectively. The validated method was successfully applied to quantify seven saponins in five sources of Flos Lonicerae, which provides a new basis of overall assessment on quality of Flos Lonicerae.

A method of precolumn derivatization GC with FID detection was developed for a simultaneous analysis of five major steroidal alkaloids of Fritillaria species, namely ebeiedine, ebeiedinone, verticine, verticinone and imperialine. Derivatization was carried out by trimethylsilylation of the hydroxyl-containing Fritillaria alkaloids to the corresponding trimethylsilylates with trime thy lsily limidazole. Reaction conditions were optimised and the alkaloids derivatives were characterised by on-line GC-MS. The validated GC method demonstrated a good linearity at the sampling ranges used. This analytical method is simple, convenient and reproducible. The developed assay was successfully applied to the determination of the major pharmacologically active alkaloids in three commonly used antitussive Fritillaria species: F. cirrhosa, F. thunbergii and F. pallidiflora.
