【期刊论文】IL-8 在兔动脉粥样硬化模型中斑块处的蛋白和基因表达

白细胞介素-8(Interleuk in-8,IL-8)是趋化因子CXC亚家族的一员,对中性粒细胞有趋化作用,可以诱导平滑肌细胞的增殖和迁移,是一个促血管生成因子,在动脉粥样硬化慢性炎症过程中起着重要作用。本文通过建立兔高脂血症动脉粥样硬化斑块模型,动态监测了在动脉粥样硬化发展过程中IL-8的蛋白和基因表达,以了解IL-8在动脉粥样硬化中的作用。采用免疫组化的方法定位IL-8在高脂血症兔动脉粥样硬化斑块模型主动脉升弓部血管壁的蛋白表达,8、12周模型组内膜显著增厚并有明显阳性染色,半定量分析A S模型组阳性染色的面积分别是对照组的4.48、8.76倍,平均积分光密度(IOD)值分别是对照组的4.16、4.36倍。采用双抗夹心EL ISA法测定兔血管组织匀浆液中的IL-8蛋白表达量,8、12周A S模型组IL-8蛋白含量与总蛋白比值和对照组比较均有显著性差异,分别是对照组的1.84、2.06倍。用原位杂交的方法定位IL-8mRNA的表达,8周模型组血管内膜有强的阳性染色,IL-8mRNA的表达增加。本研究通过建立动脉粥样硬化模型,动态监测了整个动脉粥样硬化过程中IL-8的蛋白和基因表达。IL-8的蛋白和基因上调主要是在高脂血症兔动脉粥样硬化模型的脂纹期。

In order to study the role of hemorheology in pathogenesis of acute hemorrhagic necrotizing pancreatitis (AHNP), forty four adult hybrid dogs were used and divided into 5 groups. Groups I and II were operation control (8 dogs) and acute interstitial pancreatitis group (8 dogs) respectively. Groups III-V suffered from AHNP by injection of sodium taurocholate in major pancreatic duct. Furthernore, groups IV and V were treated with Dextran 40 solution and Salvia Miltiorrhizae intravenous injection respectively. Arterial blood pressure and central venous pressure in all 5 groups did not exhibit significant changes before and after operation. Serum amylase and ]ipase level of groups III-V increased obviously after AHHP, but they decreased more rapidly in groups IV and V after treatment with drug than those in group III. Henorheologic changes were measured with Low Shear 30 sinus. Plasma viscosity of all 5 groups had no evident changes before and after operation. There were no significant changes in henatocrit (HCT), apparent viscosities of whole blood at 0.512 sec-1 (η0.512) and 51.2 see-1 (751.2), aggregation index of RBC (AI), viscous component (η') and elastic component (η") of complex viscosity; and elastic molulus (G') of whole blood in groups I and II had no significant alteration before and after operation. However, the parameters mentioned above in groups III-V increased significantly after AHNP had been induced by experiment. They decreased continuously after treatment with drug in groups IV and V. However, Salvia Miltiorrhizae injection in group V had better improvement in henorheological alteration than DX40 in group IV did. The results suggested that abnornality of hemorheology was an important factor to introduce the disturbance of nicrocirculation of pancreas during AHNP and correction of henorheological abnoruality could improve symptons of pancreatitis.

We investigated neutrophil activation, specifically F-actin content and distribution, in situations mimicking the in vivo environment using steady and oscillatory shear. Under low steady shear (<150 s−1) F-actin levels were decreased for both treated (n-formyl-L-methioryl-L-leucyl-L-phenylalanine (fMLP)) and untreated neutrophils. The F-actin content increased with a change to higher steady shear levels. Neutrophils show the same behavior of decreased F-actin content for oscillatory shear (26.7 s−1) as they did for steady shear. In both situations, the low shear levels caused a decrease in F-actin content. However, as the magnitude of the shear rate increased, cells showed a reversal to increasing F-actin content. Shear caused a decrease in F-actin in the cell cortex for both control and fMLP treated cells. Ctyochalasin B (CB), a common F-actin assembly blocker, significantly decreased F-actin content. The results indicate that neutrophils regulate their actin network based on the level and type of shear stress they encounter in the bloodstream.

The adhesion of leukocytes to vascular surface is an important biomedical problem and has drawn extensive attention. In this study, we propose a compound drop model to simulate a leukocyte with a nucleus adhering to the surface of blood vessel under steady shear flow. A two-dimensional computational fluid dynamics (CFD) is conducted to determine the local distri-bution of pressure on the surface of the adherent model cell. By introducing the parameter of deformation index (DI), we investigate the deformation of the leukocyte and its nucleus under controlled conditions. Our numerical results show that: (i) the leukocyte is capable of deformation under external exposed flow field. The deformation index increases with initial contact angle and Reynolds number of external exposed flow. (ii) The nucleus deforms with the cell, and the de-formation index of the leukocyte is greater than that of the nucleus. The leukocyte is more de-formable while the nucleus is more capable of resisting external shear flow. (iii) The leukocyte and the nucleus are not able to deform infinitely with the increase of Reynolds number because the deformation index reaches a maximum. (iv) Pressure distribution confirms that there exists a region downstream of the cell, which produces high pressure to retard continuous deformation and provide a positive lift force on the cell. Meanwhile, we have measured the deformation of human leukocytes exposed to shear flow by using a flow chamber system. We found that the numerical results are well consistent with those of experiment. We conclude that the nucleus with high viscosity plays a particular role in leukocyte deformation.

We studied hemor-heologic alterations in acute pancreatitis in 2g dogs to find out that during the early stage of induction of acute necrotizing pancrea-tiffs, all the hemorheologic parameters were signifi-cantly elevated, and continued to increase at the restintervals but no difference between the acute in-terstitial pancreatitis and control groups. We suggest that increased blood viscosity and tendency of ery-throeytes to aggregate with rouleaux formation are the causes of microeirculatory thrombosis in pan-creas. They may be th key factors in the patbo. genesis of pancreatic necrosis in acute necrotizing pancreatitis.
