摘要:目的:本课题拟解决临床病人PTX耐药问题,观察关键靶标分子GPRC5A对肺癌敏感和PTX耐药细胞的迁移的影响,并探讨其机制。方法:GEO数据库筛选GPRC5A为潜在耐药靶点。荧光定量PCR(qPCR)和蛋白免疫印迹(Western blot,WB)检测肺癌敏感和耐药细胞中GPRC5A的mRNA和蛋白的表达量,在人肺癌紫杉醇耐药细胞H1703中用CRISPR-Cas9技术敲除GPRC5A,WB验证敲除效率。将细胞分为NCI-H1703对照组、NCI-H1703-PTX实验组、H1703-PTX-KO-GPRC5A实验组,比较Transwell小室细胞迁移的情况。结果:3个GSE数据集以logFC大于1.4倍和p<0.0001的筛选标准交互到2个基因,选取GPRC5A是潜在紫杉醇耐药靶标分子,发现紫杉醇耐药细胞中GPRC5A的mRNA和蛋白表达量显著高于其对应的敏感细胞,H1703肺癌细胞迁移能力远低于H1703-PTX耐药细胞,耐药细胞敲除GPRC5A后,细胞迁移能力显著下降。结论:GPRC5A是潜在的肺癌紫杉醇耐药的靶标分子,并促进肺癌细胞的迁移。?????

CRISPR knockout of GPRC5A inhibits the migration of paclitaxel-resistant cells in lung cancer
Abstract:Objective: To investigate the key role of GPRC5A on lung cancer sensitivity and the migration of paclitaxel (PTX) resistant cells. Methods: The GEO database screened GPRC5A as a potential drug-resistant target. We use Quantitative real-time polymerase chain reaction (qPCR) and Western blot(WB)to detect the mRNA and protein expressions of GPRC5A in sensitive and resistant lung cancer cells. CRISPR-Cas9 technology was employed to knockout GPRC5A in human lung cancer PTX-resistant cell H1703, and WB was used to verify the knockout efficiency. Cells were divided into the NCI-H1703 control group, NCI-H1703-PTX experimental group, and H1703-PTX-KO-GPRC5A experimental group. Cell migration was compared using Transwell chamber assays. Results: Three GSE datasets were intersected using a screening criterion of logFC greater than 1.4 and p<0.0001, resulting in two genes, and GPRC5A was selected as a potential target of the paclitaxel resistance. The mRNA and protein levels of GPRC5A in paclitaxel-resistant cells were significantly higher than in their corresponding sensitive cells. The migration ability of H1703 lung cancer cells was much lower than that of H1703-PTX resistant cells, and the knockout of GPRC5A significantly decreased the migration ability of the resistant cells. Conclusion: GPRC5A, as a target of paclitaxel resistance, promotes migration of lung cancer cells.
